Dinophysis and Phalacroma species containing diarrheic shellfish toxins and pectenotoxins occur in coastal temperate waters all year round and prevent the harvesting of mussels during several months each year in regions in Europe, Chile, Japan, and New Zealand. Toxicity varies among morphologically similar species, and a precise identification is needed for early warning systems. Molecular techniques using ribosomal DNA sequences offer a means to identify and detect precisely the potentially toxic species. We designed molecular probes targeting the 18S rDNA at the family and genus levels for Dinophysis and Phalacroma and at the species level for Dinophysis acuminata, Dinophysis acuta, and Dinophysis norvegica, the most commonly occurring, potentially toxic species of these genera in Western European waters. Dot blot hybridizations with polymerase chain reaction (PCR)-amplified rDNA from 17 microalgae were used to demonstrate probe specificity. The probes were modified along with other published fluorescence in situ hybridization and PCR probes and tested for a microarray platform within the MIDTAL project (http://www.midtal.com). The microarray was applied to field samples from Norway and Spain and compared to microscopic cell counts. These probes may be useful for early warning systems and monitoring and can also be used in population dynamic studies to distinguish species and life cycle stages, such as cysts, and their distribution in time and space.  相似文献

This study is the first report of post-nesting migrations of loggerhead sea turtles (Caretta caretta) nesting in Sarasota County (Florida, USA), their most important rookery in the Gulf of Mexico (GOM). In total, 28 females (curved carapace length CCL between 82.2 and 112.0 cm) were satellite-tracked between May 2005 and December 2007. Post-nesting migrations were completed in 3–68 days (mean ± SD = 23 ± 16 days). Five different migration patterns were observed: six turtles remained in the vicinity of their nesting site while the other individuals moved either to the south-western part of the Florida Shelf (n = 9 turtles), the Northeast GOM (n = 2 turtles), the South GOM (Yucatán Shelf and Campeche Bay, Mexico, and Cuba; n = 5 turtles) or the Bahamas (n = 6 turtles). In average, turtles moved along rather straight routes over the continental shelf but showed more indirect paths in oceanic waters. Path analyses coupled with remote sensing oceanographic data suggest that most of long-distance migrants reached their intended foraging destinations but did not compensate for the deflecting action of ocean currents. While six out of seven small individuals (CCL < 90 cm) remained on the Florida Shelf, larger individuals showed various migration strategies, staying on the Florida Shelf or moving to long-distance foraging grounds. This study highlights the primary importance the Western Florida Shelf in the management of the Florida Nesting Subpopulation, as well as the need of multi-national effort to promote the conservation of the loggerhead turtle in the Western Atlantic. The presence of large amounts of Se-laden agricultural drainage sediment in the San Luis Drain, Central California, poses a serious toxic threat to wildlife in the surrounding environment. Effective management of the drainage sediment becomes a practical challenge because the sediment is polluted with high levels of Se, B, and salts. This two-year field study was conducted to identify the best plant species that are salt and B tolerant and that have a superior ability of volatilizing Se from drainage sediment. The drainage sediment was mixed with clean soil, and vegetated with salado alfalfa (Medicago sativa 'salado'), salado grass (Sporobulus airoides 'salado'), saltgrass-turf (Distichlis spp. 'NYPA Turf'), saltgrass-forage (Distichlis spicata (L.) Greene), cordgrass (Spartina patens 'Flageo'), Leucaenia (Leucaena leucocephola), elephant grass (Pennistum purpureum), or wild type-Brassica (Brassica spp.). Results show that elephant grass produced the greatest amount of biomass and accumulated highest concentrations of B. Highest concentrations of Se, S, and Cl were observed in wild-type Brassica. Biogenic volatilization of Se by plants and soil microbes was greater in summer. Among the treatments, the mean daily rates of Se volatilization (microg Se m(-2)d(-1)) were wild-type Brassica (39) > saltgrass-turf (31) > cordgrass (27) > saltgrass forage (24) > elephant grass (22) > salado grass (21) > leucaenia (19) > salado alfalfa (14) > irrigated bare soil (11) > non-irrigated bare soil (6). Overall, rates of Se volatilization in drainage sediment were relatively low due to high levels of sulfate. To manage Se in drainage sediment by phytoremediation, the biological volatilization process needs to be enhanced substantially under field conditions.  相似文献

We develop a novel statistical approach for classifying generalists and specialists in two distinct habitats. Using a multinomial model based on estimated species relative abundance in two habitats, our method minimizes bias due to differences in sampling intensities between two habitat types as well as bias due to insufficient sampling within each habitat. The method permits a robust statistical classification of habitat specialists and generalists, without excluding rare species a priori. Based on a user-defined specialization threshold, the model classifies species into one of four groups: (1) generalist; (2) habitat A specialist; (3) habitat B specialist; and (4) too rare to classify with confidence. We illustrate our multinomial classification method using two contrasting data sets: (1) bird abundance in woodland and heath habitats in southeastern Australia and (2) tree abundance in second-growth (SG) and old-growth (OG) rain forests in the Caribbean lowlands of northeastern Costa Rica. We evaluate the multinomial model in detail for the tree data set. Our results for birds were highly concordant with a previous nonstatistical classification, but our method classified a higher fraction (57.7%) of bird species with statistical confidence. Based on a conservative specialization threshold and adjustment for multiple comparisons, 64.4% of tree species in the full sample were too rare to classify with confidence. Among the species classified, OG specialists constituted the largest class (40.6%), followed by generalist tree species (36.7%) and SG specialists (22.7%). The multinomial model was more sensitive than indicator value analysis or abundance-based phi coefficient indices in detecting habitat specialists and also detects generalists statistically. Classification of specialists and generalists based on rarefied subsamples was highly consistent with classification based on the full sample, even for sampling percentages as low as 20%. Major advantages of the new method are (1) its ability to distinguish habitat generalists (species with no significant habitat affinity) from species that are simply too rare to classify and (2) applicability to a single representative sample or a single pooled set of representative samples from each of two habitat types. The method as currently developed can be applied to no more than two habitats at a time.  相似文献

The monitoring of dissolved zinc in the De?le river was undertaken at three different periods of the years 2008 and 2009. Electrolabile concentrations of Zn were estimated every 2 hours for several weeks by using an ATMS (Automatic Trace metal Monitoring System) based on voltammetric measurements using a solid Ag-Hg rotating disc working electrode. Complementary measurements were carried out with DGT (Diffusive Gradient in Thin films) pistons deployed directly in the river for 24 hours. Water samples filtrated at 0.45 μm were also analysed by HR-ICP-MS to estimate the total dissolved concentrations of zinc and other trace metals. High frequency monitoring of zinc over several weeks in the De?le river indicated that the concentration could change significantly over short time periods. Resuspension of polluted sediment and biological activities are two main factors that control the behaviour of zinc in the De?le river. Furthermore, in May 2009, daily cycles of the electrolabile zinc fraction have been observed at relatively constant total dissolved concentration. It is assumed that this particular behaviour of zinc is based on an exchange between colloids and/or nonelectrolabile forms and free cation and inorganic complexes at a daily time scale.  相似文献
