An indirect enzyme-linked immunosorbent assay (EIA) for metolachlor (2-chloro-N-(2-ethyl-6-methylphenyl)-N-(2-methoxy-1-methylethyl)acetamid e) detection in river water and soil was developed using serum obtained from rabbits immunized against the acid of metalaxyl ((N-(2,6-dimethylphenyl)-N-(methoxy-acetyl)-DL-alanine methyl ester) conjugated to bovine serum albumin. The assay had a linear working range from 1 to 50 ng/ml with a mean I50 value of 13.6 ng/ml and a lower detection limit of 2.0 ng/ml. Both the mean interwell and interassay coefficients of variation were less than 4% over the range of the standard curves for samples which had been prepared in phosphate buffered saline (PBS), river water, or soil extract. Assay cross-reactivity to the following four structurally related chloro-acetanilide pesticides were: propachlor (0%), metazachlor (0%), alachlor (23%), and metalaxyl (5,000%). Mean recoveries of metolachlor in spiked (2.0 to 32.0 ng/ml range) PBS, river water, and soil extract were 102%, 103%, and 110%, respectively. Soil samples were taken over a 56-d period from field plots treated with metolachlor and analyzed by GC and EIA. The correlation coefficient for comparison of the two methods was 0.96 with the slope of the linear regression line being 0.78. Furthermore, no statistical difference (P less than 0.05) was found between the dissipation curves of metolachlor derived from GC data versus EIA data.  相似文献

In May 1983, granular formulations of carbofuran, chlorpyrifos, disulfoton, fonofos, isofenphos, phorate, and terbufos were applied in incorporated bands to duplicate 2 m2 field plots of clay loam. Insecticide concentrations were determined in the bands at 0,1,2,3,4,6,8,10,12,16, and 20 wk. Following spring cultivation, the insecticides were applied to the same plots in 1984 and 1985. In addition, carbofuran was applied to previously untreated plots in 1984 and all 7 materials were applied to previously untreated plots in 1985. Sampling and analysis were carried out as in 1983. Persistence was assessed on the basis of the disappearance rates measured for the 1st 8 wk and of a calculated Effectiveness Potential (the ratio of the average residue in the upper 5 cm of the band at 8, 10 and 12 wk and the published LC95 for western corn rootworm in clay loam soil). Soils treated with carbofuran and isofenphos in 1984 and all soils treated in 1985 were tested for anti-insecticide activity. Soil cores from some carbofuran, chlorpyrifos and terbufos treated plots were sectioned vertically to establish the distribution of the insecticides during 1985. In addition, granular and pure chemical forms of isofenphos and carbofuran were applied at 10 ppm to anti-isofenphos and anti-carbofuran active and control soils (from field plots) maintained at 10 and 20% moisture in the laboratory to assess the effect of formulation and moisture on persistence in active soils. Insecticide concentrations were determined at 0,1,3,7, 10,14,21,28, and 35 days. The persistence of chlorpyrifos, terbufos and phorate was relatively constant over the 3 years and between plots receiving single and multiple treatments. Disulfoton and fonofos behavior was more variable and that of carbofuran and isofenphos was extremely variable. Anti-insecticide activity against carbofuran and isofenphos was detectable 2 wk after an initial application and was still present the following spring. Anti-insecticide activity against fonofos, terbufos sulfoxide, phorate sulfone and disulfoton sulfone was also generated in this soil. Anti-insecticide activity against chlorpyrifos, disulfoton, terbufos and phorate was not present. Carbofuran, chlorpyrifos and terbufos (+ metabolites) present in the upper 5 cm of soil averaged 93, 94 and 94%, respectively, of the total core contents over 12 wk. Oxygen evolution and uptake by whole thalli of the large marine alga Laminaria longicruris de la Pylaie were measured for 24 h, once every 2 weeks for a year, using large chambers to incubate the plants on the sea bed. Diel rates of photosynthesis and respiration were calculated from these measurements and continuous light measurements were used to extrapolate the data between observation dates. The resulting estimates were combined with measurements of growth and carbon content to give an annual carbon budget for a typical mature plant. Annual net assimilation was 6.8 mgC per cm2 of frond surface (71 cal cm-2). Approximately 45% of this appeared in the production of new frond tissue, and a further 12% was accounted for by storage of carbon in mature frond tissue. About 8% was needed for stipe growth, and the remaining 35% was assumed to be lost as dissolved organic carbon. Diel net photosynthetic rates reached a maximum in June and July and were negative only in November, indicating an ability to produce a photosynthetic surplus throughout winter. In early winter the plants drew on stored reserves to supplement photosynthesis in providing carbon for growth, but from January onwards photosynthesis provided more than enough carbon for growth.  相似文献

A method is described for studying the release of soluble organic material during photosynthesis by benthic algae. The method simulates presumed natural conditions in that the excreted matter is rapidly and continuously removed. This also facilitates analysis of the soluble compounds and the study of factors affecting excretion. Axenic cultures ofPhaeodactylum tricornutum (Bohlin) of good photosynthetic activity, as determined by measurement of oxygen output, were exposed as a surface layer to labelled carbon dioxide in a perspex fixation cell. Deep culture experiments also involved rapid separation of excreted solutes. Levels of excretion of 1 to 3% of total fixation were found. Some of the excreted material had been inside the cell for some hours. The proportion of fixed carbon excreted was similar in old and fresh media and was not affected by the presence of buffer. Co-precipitation with iron and copper was successful in extracting the labelled solutes from the filtrates, but the specific activity was too low to permit complete identification. There appeared to be differences between the materials excreted in surface and in deep culture. A discussion of related studies leads to a suggestion that it may be useful to consider the released solutes as falling into two categories; the first a low but constant level of excretion of more complex compounds, and the second a potentially much higher but variable level of excretion of simpler compounds.  相似文献

The seasonal photosynthetic responses and daily carbon gain of upper intertidal, low intertidal and subtidal (3 to 4 m depth) populations ofColpomenia peregrina were examined over a 2 yr period (1986–1988) in Santa Catalina Island, California, USA. The populations showed significant differences in their photosynthetic responses, daily carbon balance and carbon-specific growth rates when normalized to tissue area or to chlorophyll content. The substantial plasticity with respect to photosynthetic responses shown byC. peregrina is considered to be an important factor in facilitating the colonization of both intertidal and subtidal habitats. This species appears to have a cellular carbon metabolism influenced by responses to season and tidal elevation. Highest net daily carbon balance, predicted carbonspecific growth rates and net growth efficiency were achieved in upper intertidal habitats during summer. These parameters decreased in winter and progressively declined with increasing depth as plants become increasingly exposed to low-light regimes. The diminishing net daily carbon balance and predicted carbon-specific field growth rates found during winter suggest that standing stock and lower subtidal limits of distribution ofC. peregrina are at least partly controlled by these two factors.  相似文献

A widely accepted view of intertidal community organizatiton in the NW Atlantic proposes that fucoid vegetation is maintained by the actions of predators which remove species competitively superior toFucus species. Herbivory is an important component of these predatory interactions, but has been studied largely with reference to the interaction betweenF. vesiculosus andLittorina littorea. There are many species of fucoids and herbivorous invertebrates on the shores of the NW Atlantic and this paper reports field and laboratory experiments performed in 1987 (in or near Halifax, Canada) on the effects of grazing by three species ofLittorina on adults and juveniles of four species ofFucus. In laboratory experiments, portions ofFucus species were presented singly (no choice) or together (multiple choice) to single species ofLittorina. AdultF. distichus was grazed to only a small extent.F. evanescens andF. vesiculosus adult tissues were heavily grazed in most laboratory experiments.F. spiralis adults were heavily grazed in no choice experiments, but were grazed only slightly in multiple choice tests. When adult fucoid tissues were exposed to a mixed suite of grazers in the field, onlyF. vesiculosus was grazed heavily. Tests done on adult tissues showed clearly thatFucus species are highly variable in their vunerability to grazers. Moreover, we found variability among periwinkle species in grazing rates on adultFucus. We also found a great disparity in the feeding rates of different periwinkle species on juveniles ofFucus. Therefore, it is not possible to extrapolate from a single periwinkle species/single fucoid species interaction when attempting to identify vegetation structuring processes.  相似文献

The importance of species interactions and recruitment variability was examined during the first year and a half of primary succession (1988–1989) on an exposed rocky seashore near Halifax, Nova Scotia. Previous work suggested that emergent rock on these shores is normally dominated by fucoid rockweeds because predatory whelks control the sessile animal competitors, mussels and barnacles, and because herbivorous littorinids control ephemeral algal competitors. Abundances of all species except seasonal ephemeral algae were very small throughout this experiment and we found no significant effects of carnivory, herbivory, plant-animal competition or plant-plant competition. A slight facilitation of Fucus recruitment is attributed to a thin mat of ephemeral, blue-green algae. Very few other studies have directly manipulated intertidal ephemeral algae. As primary succession may be very rare in this assemblage, these results may be specific to these circumstances, but they highlight the varying importance of species interactions with variable recruitment. In particular, it appears that variations in recruitment success may be important to community structure, even when recruitment is not limited by propagule supply. The scale of the study also provides insight into successional processes occurring after the recent, extensive ice-scour of exposed seashores in this region.  相似文献

Kelp regeneration was observed for the first time in St. Margaret's Bay, Nova Scotia, Canada in an area known to have been devoid of macroalgae for several years. The regeneration was destroyed by sea urchins within 10 months. Experimentally induced kelp regeneration met a similar fate under normal grazing pressure. At the lowest sea urchin biomass and density encountered re-establishment of mature kelp stands seems highly unlikely. The sea urchin population required to suppress kelp regeneration is fed by benthic microalgae. Diatoms and other pioneer algal community species were found in the guts of sea urchins. The mean standing crop of benthic microalgae was found to be, 2.2 g C m-2 and production estimated as ca 15 g C m-2yr-1 at 8m depth. Most of the primary production of St. Margaret's Bay has been lost with the disappearing kelp populations.  相似文献

The emergence ofNephrops norvegicus (L.) from their burrows was studied on several fishing grounds around Scotland at 28 to 62 m depth. Various methods were used: sampling by trawl and baited creels, observations by television and time-lapse cameras. Peak emergence generally occurred around dusk and dawn. In shallow water (30 m) there was also a significant level of activity during the intervening night period. Day/night variations in the size composition of catches suggest that relatively more time is spent out of the burrow with increasing size. There was good agreement between the various methods employed, suggesting that trawl and camera avoidance has little influence on the pattern of emergence observed. The use of bait in some experiments confirmed that emergence is essentially a feeding activity.  相似文献

Clinal differentiation of stipe traits in Laminaria (simplices) populations occurs on a gradient of exposure to wave action. Crossability tests and the production of fertile hybrids established interfertility between populations from the extremes of the morphological range. Reciprocal transplantation experiments between exposed and sheltered sites did not produce significant changes in stipe morphology. Quantitative genetic analysis of the degree of resemblance between relatives indicated a relatively high genetic component in the interpopulation differentiation. This is consistent with the results of the transplantation experiments. Within-population heritability estimates at the exposed site were low, perhaps because of intensive local stabilising selection which acts to reduce genetic variability. The question of conspecificity of L. saccharina and L. longicruris in view of these results is discussed.  相似文献
