This study presents the microbiological characterization of the anaerobic sludge used in a two-stage anaerobic reactor for the treatment of organic fraction of urban solid waste (OFUSW). This treatment is one alternative for reducing solid waste in landfills at the same time producing a biogas (CH(4) and CO(2)) and an effluent that can be used as biofertilizer. The system was inoculated with sludge from a wastewater treatment plant (WWTP) (Río Frío Plant in Bucaramanga-Colombia) and a methanogenic anaerobic digester for the treatment of pig manure (Mesa de los Santos in Santander). Bacterial populations were evaluated by counting groups related to oxygen sensitivity, while metabolic groups were determined by most probable number (MPN) technique. Specific methanogenic activity (SMA) for acetate, formate, methanol and ethanol substrates was also determined. In the acidogenic reactor (R1), volatile fatty acids (VFA) reached values of 25,000 mg L(-1) and a concentration of CO(2) of 90%. In this reactor, the fermentative population was predominant (10(5)-10(6)MPN mL(-1)). The acetogenic population was (10(5)MPN mL(-1)) and the sulphate-reducing population was (10(4)-10(5)MPN mL(-1)). In the methanogenic reactor (R2), levels of CH(4) (70%) were higher than CO(2) (25%), whereas the VFA values were lower than 4000 mg L(-1). Substrate competition between sulphate-reducing (10(4)-10(5)MPN mL(-1)) and methanogenic bacteria (10(5)MPN mL(-1)) was not detected. From the SMA results obtained, acetoclastic (2.39 g COD-CH(4)g(-1)VSS(-1)day(-1)) and hydrogenophilic (0.94 g COD-CH(4)g(-1)VSS(-1)day(-1)) transformations as possible metabolic pathways used by methanogenic bacteria is suggested from the SMA results obtained. Methanotrix sp., Methanosarcina sp., Methanoccocus sp. and Methanobacterium sp. were identified.  相似文献

Marine mammals from Queensland, Australia, are bioaccumulating elevated concentrations of a range of polybrominated natural products. In this study, we detected three new polybrominated dimethoxybiphenyls (PBDMBs) in the blubber of selected marine mammal samples which were identified as 2,6'-dimethoxy-3,3',5-tribromobiphenyl (2,6'-diMeO-BB 36), 2,2'-dimethoxy-3,3'-dibromobiphenyl (2,2'-diMeO-BB 36), and 6,6'-dimethoxy-3,3'-dibromobiphenyl (6,6'-diMeO-BB 11). These three PBDMBs are structurally related to the known natural product 2,2'-dimethoxy-3,3',5,5'-tetrabromobiphenyl (2,2'-diMeO-BB 80). In the first part of this study, 2,2'-diMeO-BB 80 was photochemically debrominated under UV irradiation. This resulted in seven of eight possible mono- to triBDMBs as debromination products. In the second part of this study, the structure of all PBDMBs debromination products was investigated. This was supported by synthesis of two diBDMB and one triBDMB via bromination and subsequent methylation of 2,2'-biphenyldiol. Structures of the remaining PBDMBs were tentatively assigned by considering the retention times, mass spectra and amounts formed during UV irradiation of 2,2'-diMeO-BB 80 . In the third part of this study, blubber of marine mammals from Australia was analysed for PBDMBs using gas chromatography in combination with electron ionization mass spectrometry (GC/EI-MS) in the selected ion monitoring mode. In these samples, 2,2'-diMeO-BB 80 was found at concentrations of 200-1800ngg(-1) lipid weight (lw). The latter represents the highest concentration reported for this compound in environmental samples. 6,6'-diMeO BB 11, 2,2'-diMeO BB 36, and 2,6'-diMeO BB 36 were present at approximately 7ngg(-1) lipids, or 0.43-1.5% of diMeO-BB 80. No further PBDMBs were detected in the samples. The di- and triBDMBs identified in marine mammal blubber have not been reported as natural products. They may represent either new natural products or transformation products of 2,2'-diMeO-BB 80.  相似文献

Pollen storage in a colony of Apis mellifera is actively regulated by increasing and decreasing pollen foraging according to the “colony's needs.” It has been shown that nectar foragers indirectly gather information about the nectar supply of the colony from nestmates without estimating the amount of honey actually stored in the combs. Very little is known about how the actual colony need is perceived with respect to pollen foraging. Two factors influence the need for pollen: the quantity of pollen stored in cells and the amount of brood. To elucidate the mechanisms of perception, we changed the environment within normal-sized colonies by adding pollen or young brood and measured the pollen-foraging activity, while foragers had either direct access to them or not. Our results show that the amount of stored pollen, young brood, and empty space directly provide important stimuli that affect foraging behavior. Diclofenac is a non-steroidal anti-inflammatory drug, which tends to be relatively persistent in the environment. Now, a fixed-bed column bioreactor filled with sediment from the creek Münzbach (Freiberg/Saxony) under aerobic conditions showed rapid removal of diclofenac in a concentration range of 3-35 microM without previous adaptation. The conversion of higher concentrations up to 260 microM was accompanied by conspicuously decreased turnover rates indicating a toxic effect of this drug or its resulting metabolic burden on the indigenous microflora. A major metabolite occurred transiently and was identified by NMR and MS to be the p-benzoquinone imine of 5-hydroxydiclofenac. Abiotic adsorption to the biofilm was shown to determine the further fate of this reactive product of 5-hydroxydiclofenac (aut-)oxidation. The apparent lack of a degradative potential for this compound as well as the failure to detect an enrichment of diclofenac-depleting microbial activity both indicate a cometabolic nature of diclofenac transformation. 4'-Hydroxy-diclofenac, the favoured transformation product of eucaryotic diclofenac metabolism, could not be identified. The ability to convert diclofenac was shown to be widespread among biofilms from different river sediments, but measured rates obviously do not correlate with the total microbial activity. In addition, application of sediments from locations exposed to communal waste water effluents did not indicate any form of adaptation measured as an increased specific diclofenac depletion rate.  相似文献

Bisphenol A is an endocrine disruptor. Complete mineralization of bisphenol A is therefore a primary environmental issue. Here, the combination of ozonation and photocatalysis by TiO2 is proposed for the degradation and final mineralization of bisphenol A. TiO2 films deposited onto two sides of an Al lamina show good stability and high surface roughness. We used a specific experimental setup employing two facing ultraviolet lamps and TiO2 layers, together with an ozone flux. High-performance liquid chromatography–mass spectrometry determinations on bisphenol A solutions sampled at different reaction times and Fourier Transform Infrared analyses of the oxide at the end of the reaction were performed to study the reaction intermediates and the overall degradation mechanism. Our results show that pollutant mineralization achieved with the combined method is far higher, of 55% in the case of 0.3 mM bisphenol A, than those obtained by individual treatments such as photolysis (<3%), ozonation (6%), photocatalysis (6%), and by other combined processes: photolytic ozonation (13%) and catalytic ozonation (15%). This finding is explained by the occurrence of highly synergistic effects.  相似文献

Background

The coexistence of agricultural production with and without the use of genetically modified (GV) crops is supposed to be made possible in Germany by regulations, which include minimal distances of GV-fields to potentially susceptible crop fields and habitats. To explore the impact of these regulations on region specific coexistence potentials, we broadened the applicability of an existing method for the simulation of the spatial distribution of arable fields cropped with conventional, organic and GV-maize. We used simulations which combine a variety of minimum distances of GV-maize fields to assess regional specific options and limitations for coexistence.

Results

An existing method was extended to be applicable for different spatial scales, from the large (e.g. Federal State) to small (e.g. municipality). Input data consisted of cropping statistics, geometry of arable fields and protected areas. Scenarios of cropping situations included various minimal distances between GV-maize fields and protected areas and various proportions of maize within the areas. The results of the simulations represent possible distribution patterns of non-GV and GV-maize fields as well as the size of the remaining area in which additional GV-maize can be grown without violating the minimal distance rules. As suspected, increasing proportions of GV-maize and increasing minimal distances lower the areas suitable for additional GV-maize. However, the relation between the area of GV-maize grown and those suitable for GV-maize cultivation varied between the scenarios. Moreover, the variability between the municipalities was even more evident, due to varying landscape structure (proportion of maize, the ratio total arable land to maize, proportion of protected areas). Areas with high proportions of GV-maize, of protected areas and of maize could be problematical for coexistence. We discuss these parameters with regard to other coexistence studies.

Conclusions

Our method is suitable to simulate the spatial distribution of fields cultivated with GV-crops and non-GV-crops on various scales. Simulations on the scale of a Federal State reveals those areas, in which coexistence could be problematical. Simulations on a county scale, however, allow more insight into options and restrictions for coexistence in relation to landscape structural characteristics, which also can be transferred to larger scales. On the scale of municipalities simulations can help to analyse the limits of coexistence in areas of high conflict potential, moreover this level is more realistic with regard to practical agricultural decisions on the farm level.

Hintergrund

Die Koexistenz verschiedener landwirtschaftlicher Produktionsformen ?C mit und ohne Anbau von gentechnisch ver?nderten Pflanzen (GVO) ?C soll durch gesetzlich fixierte Regeln erm?glicht werden, die unter anderem die Mindestabst?nde von GVO-Anbaufl?chen zu potenziell empfindlichen anderen Ackerfl?chen und Biotopen festlegen. Hier wurde eine Methode weiterentwickelt zur Simulation der r?umlichen Verteilung der Anbaufl?chen von konventionellem, ?kologischem und GV-Mais um regionsspezifische Koexistenzpotenziale von gentechnisch ver?ndertem Mais, sowie potenzielle Konfliktgebiete zu identifizieren.

Ergebnisse

Eine für Brandenburg entwickelte GIS-Simulationsmethode wurde durch die Verwendung fl?chendeckend vorhandener Daten so erweitert, dass sie bundesweit übertragbar ist und auf unterschiedlichen r?umlichen Skalenebenen angewandt werden kann, von gro?r?umig (Bundesland) bis lokal (Gemeinde). Als Eingangsdaten wurden Anbaustatistiken sowie Geometrien der Ackerfl?chen und von FFH- und Naturschutzgebieten verwendet. In den Szenarien wurden Abstandsregelungen zwischen Maisanbaufl?chen und zu Schutzgebieten und der GV-Maisanteil variiert. Die Ergebnisse der Simulation sind m?gliche r?umliche Verteilungen von Nicht-GV-Mais und GV-Mais sowie die potenziell für den Anbau von GV-Mais zur Verfügung stehende Fl?che. Mit zunehmendem GV-Maisanteil und Mindestabst?nden zu Schutzgebieten wird die für den GV-Mais zur Verfügung stehende Fl?che st?rker ausgesch?pft. Der Anteil des GV-Mais an der potenziell für den Anbau von GV-Mais zur Verfügung stehenden Fl?che variierte zwischen den Szenarien, und noch st?rker jedoch regional zwischen den Landkreisen, verursacht durch deren verschiedene agrar- und landschaftsstrukturelle Ausstattung (Maisanbauanteil, Verh?ltnis Ackerlandsanteil/Maisanbauanteil, Schutzgebietsanteil). Ein r?umliches Konfliktpotenzial bei der Umsetzung der Koexistenz ist in Gebieten hohen Nutzungsdrucks zu erwarten, d.h. in denen sowohl der Maisanbauanteil an der Ackerfl?che und der Anbauanteil von GV-Mais, als auch der Schutzgebietsanteil hoch sind. Diese Faktoren werden diskutiert in Bezug zu Ergebnissen weiterer Koexistenzstudien.

Schlussfolgerungen

Die vorgestellte Methode ist geeignet, die r?umliche Verteilung des Anbaus von Nicht-GV-Mais und GV-Mais auf unterschiedlichen Skalenebenen zu simulieren: Die Ebene eines Bundeslandes liefert Hinweise auf Gebiete, in denen die Koexistenz problematisch sein k?nnte und kann als Grundlage weiterer Berechnungen, wie zum Beispiel der Modellierung von Genflüssen auf Landesebene dienen. Die Simulation auf der Ebene eines Landkreises oder einer Gemeinde erm?glicht genauere Aussagen über die M?glichkeiten und Grenzen der Koexistenz. Auf der Ebene der Landkreise k?nnen z.B. unterschiedliche agrar- und landschaftsstrukturelle Situationen untersucht und für eine nachfolgende Regionalisierung angewandt werden. Die Ebene der Gemeinden erlaubt die Analyse der Grenzen der Koexistenz für Gebiete mit h?herem r?umlichen Konfliktpotenzial. Simulationen auf lokaler Ebene erscheinen darüber hinaus n?her an den Entscheidungsm?glichkeiten der landwirtschaftlichen Praxis.  相似文献

The activity concentration of (90)Sr was determined in several deer bones from Austria. Strontium specific ion exchange columns with 4',4'(5')-di-t-butylcyclohexane-18-crown-6 from Eichrom Industries, Inc. were used for separation. The yield of the chemical procedure was quantified with AAS. Directly after column separation, the solution containing (90)Sr was mixed with the scintillation cocktail HiSafe III and measured by liquid scintillation counting. Prevention of (210)Pb contamination and reusability of the separation columns was investigated as well as the activity distribution within the bones. Results were compared with pre-Chernobyl measurements in Austria; a correlation between activity concentration of (90)Sr and site altitude was found.  相似文献

The present work experimentally investigates the dynamics of unsteady gravity currents produced by lock-release of a saline mixture into a fresh water tank. Seven different experimental runs were performed by varying the density of the saline mixture in the lock and the bed roughness. Experiments were conducted in a Perspex flume, of horizontal bed and rectangular cross section, and recorded with a CCD camera. An image analysis technique was applied to visualize and characterize the current allowing thus the understanding of its general dynamics and, more specifically, of the current head dynamics. The temporal evolution of both head length and mass shows repeated stretching and breaking cycles: during the stretching phase, the head length and mass grow until reaching a limit, then the head becomes unstable and breaks. In the instants of break, the head aspect ratio shows a limit of 0.2 and the mass of the head is of the order of the initial mass in the lock. The average period of the herein called breaking events is seen to increase with bed roughness and the spatial periodicity of these events is seen to be approximately constant between runs. The rate of growth of the mass at the head is taken as a measure to assess entrainment and it is observed to occur at all stages of the current development. Entrainment rate at the head decreases in time suggesting this as a phenomenon ruled by local buoyancy and the similarity between runs shows independence from the initial reduced gravity and bed roughness.  相似文献

Meroplankton are seasonally important contributors to the zooplankton, particularly at inshore sites, yet their feeding ecology is poorly known relative to holoplankton. While several studies have measured feeding in decapod larvae, few studies have examined the feeding rates of decapod larvae on natural prey assemblages throughout the reproductive season. We conducted 8 feeding experiments with Necora puber, Liocarcinus spp. and Upogebia spp. zoea larvae collected from the L4 monitoring site off Plymouth (50°15.00′N, 4°13.02′W) during spring–summer 2009 and 2010. This period spanned moderate-to-high food availability (0.5–1.6 µg chl-a L?1), but a great range in food composition with small cells <20 µm dominating in 2010. Daily rations averaged 17, 60 and 22 % of body C for the 3 respective decapod species. Clearance rates differed according to prey type, and all 3 decapod genera showed evidence of selection of dinoflagellates. Importantly, small cells including nano- and pico-plankton were ingested, this being demonstrated independently by flow cytometric analysis of the feeding experiments and molecular analysis. PCR-based analysis of the haptophyte portion of the diet revealed ingestion of Isochrysis galbana by decapod larvae in the bottle incubations and Isochrysis galbana and Phaeocystis globosa by decapod larvae collected directly from the field. This study has shown that pico- and nano-sized plankton form an important supplement to the diverse and variable diet of decapod larvae.  相似文献
