Repetitive element sequence-based polymerase chain reaction (rep-PCR) is one of the commonest methods used to identify sources of fecal contamination of water systems. In this work, BOX-A1R-based repetitive extragenic palindromic-PCR (BOX-PCR) was used to discriminate Escherichia coli strains originating from different animals and water sources, and the suitability of the technique for bacterial source tracking (BST) was evaluated. A total of 214 strains from humans, 150 strains from animals, 55 strains from sewage and 77 strains from water bodies were analyzed by the BOX-PCR technique. When maximum similarity between the fingerprints was used, a correct classification rate of 84% was achieved for strains from human and animal sources. Furthermore, 95% of the strains found in sewage were classified as being from human sources by at least one of the four classification tools used. Classification of the strains found in water bodies in the State of S?o Paulo was based on the fingerprints obtained for human and animal sources. Most of the sampling sites appeared to be affected by mixed sources of fecal contamination. The use of BOX-PCR for BST could be especially valuable in developing countries, where simplicity and cost are important considerations.  相似文献

The aim of this study was to investigate the relationship between antibiotic residues found in the muscle of cage-farm-raised Nile tilapia (Oreochromisniloticus), the occurrence of resistant bacteria, and the sanitary practices adopted by farmers in Ilha Solteira reservoir, Brazil. Nine fish (three small fish, 40–200 g; three medium-sized fish, 200–500 g; and three large fish, 500–800 g) were collected from four cage farms every three months from April 2013 to January 2014. Ten antibiotic residues were determined using liquid chromatography-mass spectrometry, and bacteria were isolated and tested for antibiotic resistance to determine the multiple antibiotic resistance (MAR) index. Only three antibiotics (oxytetracycline, tetracycline, and florfenicol) were detected in the muscle of Nile tilapia, and their residues were the highest in small fish; however, the MAR index was higher in large fish. In addition, a direct positive relationship between the MAR index and the concentration of antibiotic residues in Nile tilapia was found. Overall, the adoption of prophylactic management practices improved the sanitary status of cage farms, reducing bacterial infections and hampering the development of antibiotic-resistant bacteria.  相似文献

In this work the archaea and eubacteria community of a hypersaline produced water from the Campos Basin that had been transported and discharged to an onshore storage facility was evaluated by 16S recombinant RNA (rRNA) gene sequence analysis. The produced water had a hypersaline salt content of 10 (w/v), had a carbon oxygen demand (COD) of 4,300 mg/l and contains phenol and other aromatic compounds. The high salt and COD content and the presence of toxic phenolic compounds present a problem for conventional discharge to open seawater. In previous studies, we demonstrated that the COD and phenolic content could be largely removed under aerobic conditions, without dilution, by either addition of phenol degrading Haloarchaea or the addition of nutrients alone. In this study our goal was to characterize the microbial community to gain further insight into the persistence of reservoir community members in the produced water and the potential for bioremediation of COD and toxic contaminants. Members of the archaea community were consistent with previously identified communities from mesothermic reservoirs. All identified archaea were located within the phylum Euryarchaeota, with 98 % being identified as methanogens while 2 % could not be affiliated with any known genus. Of the identified archaea, 37 % were identified as members of the strictly carbon-dioxide-reducing genus Methanoplanus and 59 % as members of the acetoclastic genus Methanosaeta. No Haloarchaea were detected, consistent with the need to add these organisms for COD and aromatic removal. Marinobacter and Halomonas dominated the eubacterial community. The presence of these genera is consistent with the ability to stimulate COD and aromatic removal with nutrient addition. In addition, anaerobic members of the phyla Thermotogae, Firmicutes, and unclassified eubacteria were identified and may represent reservoir organisms associated with the conversion hydrocarbons to methane.  相似文献

The occurrence of diseases in cultivars has caused significant losses in global food production. The advancement of nanobiotechnology makes it possible to obtain new products to be used in the control of pathogens in cultivars. Silver nanoparticles can be synthesized by microalgae and are widely known for their antimicrobial activity. In addition, the biomass produced in microalgal culture for the biosynthesis of the nanoparticles also demonstrates antimicrobial properties, as it can increase the antibacterial and antifungal potential of the silver nanoparticles. In this context, this article addresses the use of microalgae to biosynthesize silver nanoparticles simultaneously with biomass production. In addition, we demonstrate the antimicrobial potential of these nanomaterials, as well as of the microalgal biomass produced in biosynthesis, to use in the control of pathogens in agriculture.  相似文献

Fishery wastewater treatment can be compromised due to seasonal production. The use of sequencing batch reactors is not completely successful, despite flexibility being one of the principal advantages. Most research on activated sludge is performed using synthetic wastewater to ensure a stable and constant feed. The current work compared biomass morphology and settling ability using image analysis of synthetic and real fishery wastewaters, with and without NaCl addition.  相似文献

To evaluate the spatial variability of ozone concentrations, two studies were undertaken in the montane environment of Trentino region, northern Italy, in 2007. In the first study, a 225 km2 area was considered. Here, a randomized design was used to evaluate the variability of ozone concentration at 1 and 225 km2 scale. Measurements were carried out by passive samplers between May and June 2007. In a second study, the whole 6207 km2 area of Trentino was considered. The area is covered by five grid cells of the European Monitoring and Evaluation Programme (EMEP). A systematic 15 × 15 km grid was used to allocate 15 passive samplers over the entire province, resulting into 1–4 samplers for each of the 5 EMEP grid cells (2500 km2 each) overlapping the study area. Measurements were carried out between June and September 2007. Accuracy of passive samplers was checked by direct comparison with conventional ozone analysers. Significant differences (P = 0.034) were found in ozone concentration among 1 × 1 km quadrates within the 225 km2 study area, while variability within the 1 × 1 km grid cells (coefficient of variation, CV′ = 0.12) slightly exceed the measurement error (CV′ = 0.08). At larger scales (225, 2500 and 6207 km2), ozone concentration shows much higher variability (CV′ from 0.18 to 0.28, with peak values at 0.40). Reported differences lead to very different AOT40 estimates even within the same EMEP grid cell. These findings suggest that 1 × 1 km resolution seems appropriate for ozone concentration modelling. On the other hand, significant sub-grid variation may exist at the resolution adopted by the EMEP model. Coupled with the likely variability of other important meteorological, soil and vegetation variables, our findings suggest that ozone risk assessment for vegetation based on large-scale modelled AOT40 and flux needs to be considered with great caution. The evidence reported in this paper asks for more detailed national-scale modelling, and the development of methods to incorporate local scale variations into large-scale models.  相似文献

A rugged and specific method based on tandem solid-phase extraction and liquid chromatography-tandem mass spectrometry for the determination of anti-infectives in raw sewage and wastewater plant effluents was developed. Analyte recoveries from spiked effluents ranged from 68 to 104%. Two specific selected reaction monitoring transitions and their peak area ratios were used to avoid false positives and confirm the presence of the targeted substances. Detection limits allowed low nanogram per litre detection (0.3-22 ng L(-1)). The method was successfully applied to real samples from the Montréal wastewater treatment plant. All the studied anti-infectives were found in the wastewater samples in concentrations ranging from 39 to 276 ng L(-1). Mean flows of anti-infectives were estimated from effluent concentrations and it was found that large amounts (>118 g day(-1) up to 830 g day(-1)) are discharged in the receiving waters of the St Lawrence River.  相似文献

It is known that Brazil still has a privileged position of water quantity and quality, but water use has not proceeded in a responsible manner and often results in impairment of quality. This study aims to evaluate limnological parameters, parasites and bacteria, and concentrations of heavy metals (Cd, Pb, Cu, Cr, Mn, Hg, and Zn) in surface water of Ribeirão Preto Stream. The Ribeirão Preto Stream is located in urban areas under anthropogenic influence. The results showed that the levels of dissolved oxygen values were lower than those established by the National Environmental Council (CONAMA Resolution No 357/2005). The reading of electrical conductivity showed values typical of impacted environments. The parasitological analysis revealed the presence of nematode larvae. The bacteriological analysis showed higher values for total coliform and Escherichia coli than those set by the Brazilian National Environment Council (CONAMA). The heavy metals Cd, Pb, Cu, Cr, Mn, Hg, and Zn showed concentrations in accordance with the guidelines established by CONAMA. The results provide data on the quality of these waters and showed the necessity to protect the watercourse from point sources of contamination, recommending their continued monitoring.  相似文献

Objective

To evaluate fetal brain development using MRI (magnetic resonance imaging) in CDH (congenital diaphragmatic hernia).

Methods

52 isolated left CDH and 104 control fetuses were imaged using MRI. Brain morphometry (Biparietal diameter—BPD, brain fronto-occipital diameter—BFOD, third ventricle, posterior ventricles, transcerebellar diameter—TCD, anteroposterior and craniocaudal cerebellar vermis diameter—AP and CC) and cortical structures (bilateral cingulate fissure—CF, insular fissure—IF, insular depth - ID) were compared with controls using Mann–Whitney test.

Results

Median gestational age at MRI (p = 0.95)and the median biparietal diameter (p = 0.737) were comparable. Among morphometric parameters, only the brain fronto-occipital diameter was significantly smaller in CDH (p = 0.001) and the third ventricle was significantly greater in CDH (<0.0001). Among cortical structures, the cingulate and insular fissures were significantly deeper in CDH fetuses (p < 0.0001) as the insular depth ID was smaller in CDH (p < 0.03).

Conclusions

CDH fetuses have a smaller fronto-occipital diameter, reduced insular depth, deeper cingulate and insular fissure, and greater third ventricle width as compared to controls. These findings suggest that left CDH may have an impact on fetal brain development with an overall reduction in brain volume.  相似文献
