Wolbachia are symbiotic bacteria found in many arthropods and filarian nematodes. They often manipulate the reproduction of host arthropods. In the present study, female-biased sex-ratio distortion in the butterfly Eurema hecabe was investigated. Breeding experiments showed that this distorted sex ratio is maternally inherited. When treated with tetracycline, adult females of the thelygenic line produced male progeny only. After PCR using Wolbachia-specific primers for the ftsZ gene a positive result was seen in the thelygenic females, but not in male progeny from tetracycline-treated females, or individuals from a Tokyo population with normal sex ratio and reproduction. Cytological observations showed that thelygenic females lack the sex chromatin body (W chromosome). The results strongly suggest that the sex-ratio distortion in E. hecabe is due to feminization of genetic males by Wolbachia.  相似文献

Characterization of the effective cellulose degrading strain CTL-6   总被引:1,自引:0,他引:1  
An efficient cellulose degrading bacteria exists in the thermophilic wheat straw-degrading community, WDC2. However, this strain cannot be isolated and cultured using conventional separation techniques under strict anaerobic conditions. We successfully isolated a strain of effective cellulose degrading bacteria CTL-6 using a wash, heat shock, and solid-liquid alternating process. Analysis of its properties revealed that, although the community containing the strain CTL-6 grew under aerobic conditions, the purified strain CTL-6 only grew under anaerobic culture conditions. The strain CTL-6 had a striking capability of degrading cellulose (80.9% weight loss after 9 days of culture). The highest efficiency value of the endocellulase (CMCase activity) was 0.404 μtmol/(min-mL), cellulose degradation efficiency by CTL-6 was remarkably high at 50-65°C with the highest degradation efficiency observed at 60°C. The 16S rRNA gene sequence analysis indicated that the closest relative to strain CTL-6 belonged to the genus Clostridium thermocellum. Strain CTL-6 was capable of utilizing cellulose, celiobiose, and glucose. Strain CTL-6 also grew with Sorbitol as the sole carbon source, whereas C.therrnocellum is unable to do so.  相似文献

In order to develop effective conservation strategies for endangered migratory species, the link between feeding and breeding grounds needs to be clarified. In this study, the genetic compositions of consecutive Japanese feeding aggregations of green turtles (Chelonia mydas) along the Kuroshio Current were examined by mixed-stock analyses of mitochondrial DNA control-region sequences. The results indicated that the southern feeding aggregation around Yaeyama (24.3°N, 124.0°E) was sourced from various Pacific rookeries in the Yaeyama, Ogasawara, Western Pacific, and Indian Oceans and Southeast Asia. Among northern feeding aggregations, the Ginoza (26.5°N, 128.0°E) aggregation was also sourced from the Western Pacific Ocean, but the Nomaike (31.4°N, 130.1°E), Muroto (33.2°N, 134.2°E), and Kanto (35.6°N, 140.5°E) aggregations were contributed mostly by the closer Ogasawara rookeries. The reduced contribution from tropical Pacific rookeries to northern feeding aggregations and the significant correlation between genetic differentiation and geographical distance matrices of feeding aggregations indicated that most hatchlings from these regions transported by the Kuroshio Current settle in upstream feeding grounds along the Japanese archipelago, implying that current flow influences the composition of feeding aggregations. Differences in the composition of relatively close neritic feeding aggregations have important conservation implications, for which both regional and multinational conservation strategies are needed.  相似文献

The aim of this paper is to present an evaluation method to aid decision makers in the prioritization and selection of appropriate countermeasures at the planning stage of site remediation. We introduced a hierarchical network (hiernet) decision structure and applied the Analytic Network Process (ANP) supermatrix approach to measure the relative desirability of the remedial alternatives using the decision maker's value judgment as input. A simplified illustrative example is presented to elucidate the process, as it is being applied to evaluate the feasible remedial countermeasures of a contaminated site caused by uncontrolled landfill. Four decision models derived from the generalized hiernet were examined to describe the effect of hierarchic functional dependence, inner dependence and feedback cycle on the derivation of the priority weights. The ANP could provide a more flexible analytical framework to break down one's judgment through a more elaborate structure in a systematic way to understand the complexity of the decision problem. The proposed method therefore may not only aid in selecting the best alternative but also may help to facilitate communication to understand why an alternative is preferred over the other alternatives through the analysis of the derived weights and its underlying decision structure.  相似文献

Forest conversion is one of the greatest global threats to biodiversity, and land-use change and subsequent biodiversity declines sometimes occur over a variety of underlying geologies. However, how forest conversion and underlying geology interact to alter biodiversity is underappreciated, although spatial variability in geology is considered an integral part of sustaining ecosystems. We aimed to examine the effects of forest conversion to farmland, the underlying geology, and their interaction on the stream fishes’ diversity, evenness, and abundance in northeastern Japan. We disentangled complex pathways between abiotic and biotic factors with structural equation modeling. Species diversity of stream fishes was indirectly shaped by the interaction of land use and underlying geology. Diversity declined due to nutrient enrichment associated with farmlands, which was mainly the result of changes in evenness rather than by changes in species richness. This impact was strongest in streams with volcanic geology with coarse substrates probably because of the differential responses of abundant stream fishes to nutrient enrichment (i.e., dominance) and the high dependency of these fishes on large streambed materials during their life cycles. Our findings suggest that remediation of deforested or degraded forest landscapes would be more efficient if the interaction between land use and underlying geology was considered. For example, the negative impacts of farmland on evenness were larger in streams with volcanic geology than in other stream types, suggesting that riparian forest restoration along such streams would efficiently provide restoration benefits to stream fishes. Our results also suggest that land clearing around such streams should be avoided to conserve species evenness of stream fishes.  相似文献

A large amount of rice straw is generated and left as much in paddy fields, which causes greenhouse gas emissions as methane. Rice straw can be used as bioenergy. Rice straw pellets are a promising technology because pelletization of rice straw is a form of mass and energy densification, which leads to a product that is easy to handle, transport, store and utilize because of the increase in the bulk density. The operational conditions required to produce high quality rice straw pellets have not been determined. This study determined the optimal moisture content range required to produce rice straw pellets with high yield ratio and high heating value, and also determined the influence of particle size and the forming temperature on the yield ratio and durability of rice straw pellets. The optimal moisture content range was between 13% and 20% under a forming temperature of 60 or 80 °C. The optimal particle size was between 10 and 20 mm, considering the time and energy required for shredding, although the particle size did not significantly affect the yield ratio and durability of the pellets. The optimized conditions provided high quality rice straw pellets with nearly 90% yield ratio, ?12 MJ/kg for the lower heating value, and >95% durability.  相似文献

Resurrection of the iron and phosphorus resource in steel-making slag   总被引:4,自引:0,他引:4  
 This research focused on the treatment of steel-making slags to recycle and recover iron and phosphorus. The carbothermal reduction behavior of both synthesized and factory steel-making slag in microwave irradiation was investigated. The slags were mixed with graphite powder and heated to a temperature higher than 1873 K to precipitate a lump of Fe–C alloy with a diameter of 2–8 mm. The larger the carbon equivalent (Ceq, defined in the text), the higher the fractional reduction of iron and phosphorus. An increase in the SiO2 content of slag led to a considerable improvement in the reduction for both iron and phosphorus because of the improvement in the fluidity of the slags and an increase in the activity coefficient of P2O5 in the slags. The extraction behavior of phosphorus from Fe–P–Csatd alloy was also investigated at 1473 K by carbonate flux treatment. For all the experiments with a processing time longer than 10 min, the phosphorus in the fluxes could be concentrated to more than 9% (w/w) showing that it could be used as a phosphorus resource. Compared with K2CO3 flux treatment, that using Na2CO3 was more effective for the extraction of phosphorus, and this was attributed to the lower evaporation of Na2CO3. We have measured the elemental concentrations in aerosols with a 2-h time resolution in two different types of working environment: a chemistry laboratory dealing with the processing of advanced nanoparticulate materials and a medium-sized machine workshop. Non-stop 10-day and 12-day samplings were performed at each location in order to determine the concentration trends during the non-working/working and weekday/weekend periods. Supplementary measurements of PM10 aerosols with a 2-day sample collection time were performed with a standard Gent PM10 sampler to compare the elemental concentrations with the time-averaged concentrations detected by the 2D step-sampler. The concentrations were determined a posteriori by analyzing the x-ray spectra of aerosol samples emitted after 3-MeV proton bombardment. The PM10 samples collected in the chemistry laboratory were additionally inspected by scanning electron microscopy (SEM) and energy-dispersive X-ray spectroscopy (EDX) to determine the chemical compositions of the individual particles. In the workshop, a total PM10 mass sampling was performed simultaneously with a minute resolution to compare the signal with typical outdoor PM10 concentration levels. A factor analysis of the time-resolved dataset points to six and eight factors in the chemistry laboratory and the machine workshop, respectively. These factors describe most of the data variance, and their composition in terms of different elements can be related to specific indoor activities and conditions. We were able to demonstrate that the elemental concentration sampling with hourly resolution is an excellent tool for studying the indoor air pollution. While sampling the total PM10 mass concentration with a minute resolution may lack the potential to identify the emission sources in a “noisy” environment, the time averaging on a day time scale is too coarse to cope with the working dynamics, even if elemental sensitivity is an option.  相似文献
