A general framework for designing sampling programs is described. As part of the sampling program the problem of concern, or reason for sampling, needs to be clearly stated and objectives specified. The development of a conceptual model will assist the clarification of objectives and the choice of indicators to be sampled.Objectives can then be stated as testable hypotheses and decisions made about the samallest difference/changes that are to be detected/observed by the sampling.To allow the collection of representative samples, and the statistical analysis of data to be collected, the potential sources of variability in the data must be considered. Site, selection, frequency and replication must account for the expected variability.Before field collection of samples occurs, the sample collection device needs to be tested as to its efficiency to collect a representative sample. It also will usually be necessary to consider how samples are to be preserved to inhibit biological and chemical change. All sample programs require a quality assurance program to identify, measure and control errors.As well as the above the cost-effectiveness of the program should be evaluated in terms of maximizing the information obtained/cost.  相似文献

The filamentous fungus Scopulariopsis brevicaulis biomethylates inorganic antimony(III) compounds to trimethylstibine, that can be detected in culture headspace gases. Dimethylantimony and trimethylantimony species have been detected in the medium of these cultures, but the origin of these species was controversial. We now show that the dimethylantimony species is a true intermediate on the pathway to trimethylstibine (rather than arising from trimethylstibine oxidation or as an analytical artifact) because no dimethylantimony species are formed on trimethylstibine oxidation, as determined by using HG-GC-AAS. Furthermore, the dimethylantimony and trimethylantimony species can be separated, by using anion exchange chromatography, and so the dimethylantimony species is not an analytical artifact, formed during the hydride generation process. The antimony biomethylation mechanism was further probed by measuring incorporation of the methyl group, from 13CD3-L-methionine and CD3-D-methionine, into methylantimony species and, for comparison, into methylarsenic species. The percentage incorporation of the labeled methyl group into methylarsenic and methylantimony species was not significantly different. The incorporation from 13CD3-L-methionine was 54% and 47% for antimony and arsenic, respectively. The incorporation from CD3-D-methionine was 20% and 16% for antimony and arsenic, respectively. It appears that the biomethylation of arsenic and antimony occur by very similar, perhaps identical, mechanisms.  相似文献

Ceratium tripos dominated a multi-species dinoflagellate patch in the sub-surface chlorophyll maximum in August 1978 on the Southern California shelf. The specific growth rate () ofC. tripos averaged 0.25 d-1. Patch length was about 45 km along the shelf. Several members of the subsurface dinoflagellate assemblage were also present in surface samples, but only during the daytime. These apparent vertical migrators includedProrocentrum micans, C. furca, Gonyaulax polyedra and other less common forms. The growth ofC. tripos in the California patch is compared with that in aC. tripos patch off New York in 1976.  相似文献

A non-thecate dinoflagellate, Gymnodinium splendens, was studied in a 12 d laboratory experiment in 2.0x0.25 m containers in which light, temperature, and nutrients could be manipulated. Under a 12 h light: 12 h dark cycle, the dinoflagellates exhibited diurnal vertical migrations, swimming downward before the dark period began and upward before the end of the dark period. This vertical migration probably involved geotaxis and a diel rhythm, as well as light-mediated behavior. The vertical distribution of nitrate affected the behavior and physiology of the dinoflagellate. When nitrate was present throughout the container, the organisms resembled those in exponential batch culture both in C:N ratios and photosynthetic capacity (Pmax); moreover, they migrated to the surface during the day. In contrast, when nitrate was depleted, C:N ratios increased, Pmax decreased, and the organisms formed a subsurface layer at a depth corresponding to the light level at which photosynthesis saturated. When nitrate was present only at the bottom of the tank, C:N ratios of the population decreased until similar to those of nutrient-saturated cells and Pmax increased; however, the dinoflagellates behaved the same as nutrient-depleted cells, forming a subsurface layer during the light period. Field measurements revealed a migratory subsurface chlorophyll maximum layer dominated by G. splendens. It was just above the nitracline during the day, and in the nitracline during the night, which concurs with our laboratory observations.  相似文献

In the Gulf of St. Lawrence, Canada, productivity-determining biophysical interactions occur in the upper 0 to 30 m of the water column. The eggs and larvae of several commercially important marine invertebrates and fishes (e.g. Gadus morhua L.) are found in this layer. Measurements of the diffuse attenuation coefficients for ultraviolet-B radiation (280 to 320 nm, UV-B) at various locations in this geographic region indicated maximum 10% depths (the depth to which 10% of the surface energy penetrates at a given wavelength) of 3 to 4 m at a wavelength of 310 nm. This represents a significant percentage of the summer mixed-layer water column: organisms residing in this layer are exposed to UV-B radiation. Laboratory experiments using a Xenon-arc-lamp based solar simulator revealed that cod embryos exposed to UV-B exhibited high wavelength-dependent mortality. The strongest effects occurred under exposures to wavelengths below 312 nm. This susceptibility was also dependent upon developmental stage; mortality was particularly high during gastrulation. At the shorter wavelengths (<305 nm) UV-B-induced mortality was strongly dose-dependent, and not significantly influenced by dose-rate. The biological weighting function (BWF) derived for UV-B-induced mortality in cod eggs is similar to that reported for naked DNA – suggesting that the mortality is a direct result of DNA damage. There was no evidence of a detrimental effect of ultraviolet-A radiation (320 to 400 nm). Calculations based upon the BWF indicate that, under current noon surface irradiance, 50% of cod eggs located at or very near (within 10 cm) the ocean surface will be dead after 42 h of exposure. Under solar spectral irradiance simulating a 20% decrease in ozone layer thickness, this time drops to 32 h. These are first-order estimates based upon surface irradiance taken at a time of day during which the values would be maximal. Nonetheless, they illustrate the relative changes in UV-B impacts that will result from ozone layer depletions expected over the coming decades. It is also important to point out that variability in cloud cover, water quality, and vertical distribution and displacement of cod eggs and larvae within the mixed layer, can all have a greater effect on the flux of UV-B radiation to which fish eggs are exposed than will ozone layer depletion at these latitudes. Hundreds of millions of people around the world are exposed to elevated concentrations of inorganic and organic arsenic compounds, increasing the risk of a wide range of health effects. Studies of the environmental fate and human health effects of arsenic require authentic arsenic compounds. We summarize here the synthesis and characterization of more than a dozen methylated and thiolated arsenic compounds that are not commercially available. We discuss the methods of synthesis for the following14 trivalent(Ⅲ) and pentavalent() arsenic compounds: monomethylarsonous acid(MMA~Ⅲ), dicysteinylmethyldithioarsenite(MMA~Ⅲ(Cys)_2), monomethylarsonic acid(MMA~Ⅴ),monomethylmonothioarsonic acid(MMMTAⅤ) or monothio-MMA~Ⅴ, monomethyldithioarsonic acid(MMDTA~Ⅴ) or dithio-MMA~Ⅴ, monomethyltrithioarsonate(MMTTA~Ⅴ) or trithio-MMA~Ⅴ,dimethylarsinous acid(DMA~Ⅲ), dimethylarsino-glutathione(DMA~Ⅲ(SG)), dimethylarsinic acid(DMA~Ⅴ), dimethylmonothioarsinic acid(DMMTA~Ⅴ) or monothio-DMAⅤ, dimethyldithioarsinic acid(DMDTA~Ⅴ) or dithio-DMA~Ⅴ, trimethylarsine oxide(TMAO~Ⅴ), arsenobetaine(AsB), and an arsenicin-A model compound. We have reviewed and compared the available methods,synthesized the arsenic compounds in our laboratories, and provided characterization information. On the basis of reaction yield, ease of synthesis and purification of product, safety considerations, and our experience, we recommend a method for the synthesis of each of these arsenic compounds.  相似文献

The effects of nano-scale and micro-scale zerovalent iron (nZVI and mZVI) particles on general (dehydrogenase and hydrolase) and specific (ammonia oxidation potential, AOP) activities mediated by the microbial community in an uncontaminated soil were examined. nZVI (diameter 12.5 nm; 10 mg g−1 soil) apparently inhibited AOP and nZVI and mZVI apparently stimulated dehydrogenase activity but had minimal influence on hydrolase activity. Sterile experiments revealed that the apparent inhibition of AOP could not be interpreted as such due to the confounding action of the particles, whereas, the nZVI-enhanced dehydrogenase activity could represent the genuine response of a stimulated microbial population or an artifact of ZVI reactivity. Overall, there was no evidence for negative effects of nZVI or mZVI on the processes studied. When examining the impact of redox active particles such as ZVI on microbial oxidation-reduction reactions, potential confounding effects of the test particles on assay conditions should be considered.  相似文献

Most groundwater modelers avoid using static heads measured from active production wells because they can introduce a bias into model calibration. However, in the deep confined Cambrian-Ordovician Sandstone Aquifer System in the Central Midcontinent of North America, dedicated observation wells are sparse and remote from areas of most concentrated pumping. As a result, in areas where drawdown is the greatest and modeling is most needed, only static heads from production wells are available for calibration. This paper evaluates two leading sources of discrepancies in using production well data, spatial and temporal structural error (S.E.). A simple Theis solution is used to evaluate the potential magnitude of spatial S.E. when calibrating a regional MODFLOW model with coarse cell resolution. Despite theoretical analyses indicating that spatial S.E. could be significant, statistical analysis of the model results suggests that temporal S.E. is dominant. Long (ranging over decades) or frequent (monthly) head datasets are key in understanding temporal S.E., to better capture water-level variability. In this study, the range in static head observations impacted estimates of the remaining time a well could extract water from the aquifer by 0.1 to 16.0 years. This uncertainty in future water supply is highly relevant to stakeholders and must be assessed in hydrographs depicting risk.  相似文献

Thirty cases of cervical cystic hygroma were diagnosed in the first trimester of pregnancy. Karyotype analysis was available in 29 (97 per cent). Fifteen (52 per cent) had a chromosomal abnormality. The ultrasound appearance was described as posterior cervical, lateral cervical, or cervical hygroma with hydrops. Of the 14 euploid embryos, six were electively aborted, two are undelivered, and six have been delivered as phenotypically normal infants. In cases in which the chromosomes were normal and the pregnancy continued, all lesions resolved by 18 weeks.  相似文献
