GOAL, SCOPE AND BACKGROUND: Pentachlorophenol (PCP) is the second highest volume pesticide used in the United States. It is a mutagenic compound whose exposure poses significant health effects, One of the most desirable, environmentally friendly treatment methods is bioremediation. For soil-based contamination, the effectiveness of bioremediation will also be affected by the presence of an active indigenous population, sorption of the contaminant onto the soil, and environmental parameters. METHODS: Two pure strains and their mixed culture were used to evaluate PCP biodegradation in two different field soils, Columbia (CO) and New Mexico (NM). Biostimulation of the indigenous microbes was evaluated by adding nutrients. The efficiency of adding bacteria strains (bioaugmentation) for degrading PCP was determined with Arthrobacter sp., Flavobacterium sp. and a 50:50 mixture of the two bacteria strains. RESULTS: In CO soil, only 24%, 12% and 25% of the initial PCP concentration were degraded by Flavobacterium sp., Arthrobacter sp. and mixed culture, respectively. Arthrobacter sp. was used in NM soil with two initial concentrations and achieved degradation efficiencies of 57% and 61% for 361 and 95 mg kg- concentrations, respectively. Discussion. Analysis via statistical methods showed that the bacteria had different efficiencies on PCP degradation in each soil. 2 CONCLUSIONS: All bacteria catalyzed a higher PCP degradation when present in NM soil. Second, Flavobacterium sp. degraded more PCP than Arthrobacter sp. in CO soil. The mixed culture achieved the highest degradation efficiency regardless of the initial concentration or soil origin. RECOMMENDATIONS AND PERSPECTIVES: The effect of the soil properties, such as the soil organic matter (SOM) on PCP biodegradation should be investigated. Future work can also investigate the effect of aging time on biodegradation.  相似文献

We conducted field surveys and laboratory analyses to test the effects of soil characteristics in habitat mitigation sites and natural sites on the growth and condition of blue elderberry (Sambucus mexicana), which is the sole host plant for the federally threatened Valley elderberry longhorn beetle (Desmocerus californicus dimorphus). Thirty mitigation and 16 natural sites were selected throughout the range of the beetle. We found that although plant relative growth rates were comparable between mitigation sites and a natural site, mitigation sites contained substantially less soil nutrients than mitigation sites. Within mitigation sites, elderberry health and growth were positively correlated with the amount of total nitrogen in soils and less strongly with other soil nutrients and soil moisture. Analyses demonstrated reductions in the relative growth rate of elderberry as mitigation sites aged, and that soil nutrients and soil moisture became depleted over time. For mitigation sites, it took approximately seven years to develop basal stem diameters that have been linked to successful beetle colonization. Mitigation sites have smaller shrubs than natural sites and growth slows as mitigation sites age, thus delaying convergence of conditions between natural and mitigation sites. Analyses of soil particle size and whether sites were within the 100-year floodplain (as an indicator of riparian conditions) were inconclusive. We recommend investigating fertilizing and optimum planting densities for elderberry at restoration and mitigation sites, as well as increasing the duration of irrigation and monitoring.  相似文献

Goals, Scope and Background It has been observed that hydrocarbon treated wastewaters still contain high COD and a number of intermediates. This suggests that the required catabolic gene pool for further degradation might be absent in the system or, that its titer value is not significant enough. By providing the desired catabolic potential, the overall efficiency of the treatment system can be improved. This study aims to demonstrate this concept by bioaugmentation of a lab-scale reactor treating refinery wastewater with a consortium having the capacity to complement the alkB genotype to the available microbial population. Methods Two reactors were set up using activated biomass collected from a refinery treatment plant and operated at a continuous mode for a period of 8 weeks. The feed to both reactors was kept constant. Crude oil was spiked regularly. One reactor was bioaugmented with a consortium previously described for crude oil spill remediation. The efficiency of the bioaugmented reactor was demonstrated by reduced COD. The changes in the microbial population over a period of time were analyzed by RAPD. Catabolic activity of the biomass in both reactors was monitored by PCR. The presence of the catabolic loci was confirmed by Southern Hybridization. Results and Discussion 52.2% removal of COD was observed in the bioaugmented reactor while only 15.1% reduction of COD was observed in the reactor without bioaugmentation. The change in microbial population can be seen from the 4th week, which also corresponds to improved catabolic activity. The presence of the bedA locus was seen in all samples, which indicates the presence of aromatic degraders, but the appearance of the alkB locus, from the 6th week onwards, which was observed only in the samples from the bioaugmented reactor. The results suggest that the gene pool of the bioaugmented reactor has catabolic loci that can degrade accumulated intermediates, thus improving the efficiency of the system. Conclusions In this study, improvement of efficiency of bioremediation was demonstrated by addition of catabolic loci that are responsible for degradation. Bioaugmentation was carried out in biomass that was collected from an ETP (effluent treatment plant) treating hydrocarbon containing wastewater to study the strategies for improvement of the treatment system. Biostimulation, only marginally improved the efficiency, when compared to bioaugmentation. The improved efficiency was demonstrated by COD removal. The presence of the alkB locus suggests the importance of a catabolic gene pool that acts on accumulated intermediates. It is well documented that straight chain aliphatics and intermediates of aromatic compounds after ring cleavage, accumulate in refinery wastewater systems, thereby hindering further degradation of the wastewater. Supplementation of a catabolic gene pool that treats the lower pathway compounds and alkanes will improve the overall efficiency. In this study, results suggest that the alkB locus can also be used to monitor the degradative mode of the activated biomass. Recommendations and Perspective . Pollution from petroleum and petroleum products around the globe are known to have grave consequences on the environment. Bioremediation, using activated sludge, is one option for the treatment of such wastes. Effluent treatment plants are usually unable to completely degrade the wastewater being treated in the biological unit (the aerator chambers). The efficiency of degradation can be improved by biostimulation and bioaugmentation. This study demonstrates the improved efficiency of a treatment system for wastewater containing hydrocarbons by bioaugmentation of a consortium that supports degradation. Further experiments on a pilot scale are recommended to assess the use of bioaugmentation on a large scale. The use of molecular tools, like DNA probes for alkB, to monitor the system also needs to be explored.  相似文献

Using previously published histological data on multiple, monthly samples of Sardina pilchardus collected in the central Aegean and Ionian Seas (September 1999–August 2000, and November 2000–February 2001), the Mediterranean sardine was treated as a case study to investigate the biological characteristics of ephemeral spawning aggregations in multiple-spawning clupeoids. Actively spawning (Day0) females in the Mediterranean sardine, i.e., the daily class of spawners caught a few hours prior, during, or after the spawning act, were shown to separate spatially from late (Day1+) spawners and non-spawning females, taking with them a large proportion of conspecific males which were also in advanced spawning condition and in better somatic condition compared to the remaining population. In addition, information from 28 stocks of multiple-spawning clupeoids from a wide range of geographic locations, belonging to 14 species and 2 families (Engraulidae and Clupeidae), was reviewed and analyzed pertinent to the formation of ephemeral spawning aggregations. Results from the latter analysis indicated similar patterns of spatial segregation of Day0 spawners in the reviewed clupeoid stocks as in the Mediterranean Sardine, which strongly suggested that the formation of ephemeral spawning aggregations is a common behavioral trait among multiple-spawning clupeoids. Anarchy, where honeybee workers produce males in the presence of a queen, is extremely rare in natural honeybee populations, suggesting that there are colony-level costs associated with being anarchistic. Yet, no significant costs have yet been identified. A possible reason for this may be that researchers have only focused on the behaviour of anarchistic workers, which have been shown to perform worker-tasks as do wild-type workers. Possibly, therefore, costs associated with anarchy should be sought in anarchistic queens and not workers. A potential cost could be a lower survival rate of eggs laid by anarchistic queens perhaps because their egg-marking signal is not as clear as that of a wild-type queen. In this paper, we determined the removal rate of eggs laid by anarchistic queens in standard worker-policing bioassays. Our results show that eggs laid by anarchistic queens are removed at a higher rate than eggs laid by wild-type queens. This does not seem to be due to differences in hydrocarbons found on the surface of eggs, as both egg types showed the same alkanes and alkenes in similar proportions. We postulate that higher removal rates of queen-laid eggs due to recognition errors may be one reason that anarchy is rare in natural honeybee populations.  相似文献

Increased salinity in spawning and nursery grounds in the Savannah River estuary was cited as the primary cause of a 97% decrease in adult striped bass (Morone saxatilis) and a concomitant 96% decrease in striped bass egg production. Restoration efforts focused on environmental remediation and stock enhancement have resulted in restored salinity patterns and increased egg and adult abundances. However, future water needs or harbor development may preclude further recovery by reducing freshwater inflow or increasing salinity intrusion. To assess the effect of potential changes in the salinity regime, we developed models relating discharge, tidal phase, and salinity to striped bass egg and early larval survival and re-cast these in a quantitative Bayesian belief network. The model indicated that a small upstream shift (≤1.67 km) in the salinity regime would have the least impact on striped bass early life history survival, whereas shifts >1.67 km would have progressively larger impacts, with a 8.33-km shift potentially reducing our estimated survival probability by >28%. Such an impact could have cumulative and long-term detrimental effects on the recovery of the Savannah River striped bass population. The available salinity data were collected during average and low flows, so our model represents some typical and some extreme conditions during a striped bass spawning season. Our model is a relatively simplistic, “first-order” attempt at evaluating potential effects of changes in the Savannah River estuarine salinity regime and points to areas of concern and potential future research.  相似文献
