The expansion of invasive Japanese knotweed s.l. is of particular concern because of its aptitudes to rapidly colonize diverse environments, especially anthropized habitats generally characterized by their pollution with heavy metals. Whether the presence of heavy metals impacts the performance traits of this plant is a central question to better understand its invasive properties, though no controlled approach to assess these effects was yet reported. In this aim, we undertook greenhouse experiments where rhizome fragments of Japanese knotweed s.l. (Fallopia japonica and Fallopia × bohemica) were grown during 1 and 3 months, in a soil pot artificially polluted or not with heavy metals added in mixture (Cd, Cr, Pb, Zn). Our results showed that (i) the presence of heavy metals delayed rhizome regeneration and induced lowered plant part weights but did not affect plant height after 3 months; (ii) the effect of metals on the metabolic profiles of belowground part extracts was only detectable after 1 month and not after 3 months of growth, though it was possible to highlight the effect of metals independently of time and genotype for root extracts, and torosachrysone seemed to be the most induced compound; and (iii) the hybrid genotype tested was able to accumulate relatively high concentrations of metals, over or close to the highest reported ones for this plant for Cr, Cd and Zn, whereas Pb was not accumulated. These findings evidence that the presence of heavy metals in soil has a low impact on Fallopia sp. overall performance traits during rhizome regeneration, and has a rather stimulating effect on plant growth depending on pollution level.  相似文献

Applying Csikszentmihalyi's ( 1990 ) flow theory of optimal experience to the workplace, two studies examined the relationships of employees' perceived skill and challenge at work and need for achievement with their positive mood, intrinsic task interest, and extra‐role performance. Among achievement‐oriented employees only, high skill and challenge was associated with greater positive mood, task interest, and performance than other skill/challenge combinations. The mechanism of toxicity of ionic copper and copper complexes to growth, photosynthesis, respiration, ATP levels and mitochondrial electron-transport chain-activity in two marine diatoms, Nitzschia closterium (Ehrenberg) W. Smith (Hasle, 1964) and Asterionella glacialis Castracane, and one freshwater green alga, Chlorella pyrenoidosa Chick was investigated. Copper ions depressed both cell division and photosynthesis in A. glacialis and C. pyrenoidosa, whereas ionic copper concentrations which were inhibitory to cell division in N. closterium had no effect on photosynthesis, respiration, ATP production, electron transport or membrane ultrastructure. This suggests that in N. closterium, copper does not act on the chloroplast, the mitochondrion, or the cell membrane, since if it did, the above parameters should be affected. Copper-ethylxanthogenate was exceptional amongst the copper complexes in that it stimulated respiration, mitochondrial electrontransport and ATP formation in N. closterium under conditions of strongly inhibited cell division and slightly stimulated photosynthesis. Ionic copper toxicity may result from an intracellular reaction between copper and reduced glutathione (GSH), leading to a lowering of the GSH:GSSG ratio and suppression of mitosis. In addition, copper inhibits the enzyme catalase and reduces cell defence mechanisms against H2O2 and oxygen-free radicals. Lipid-soluble copper complexes are more toxic than ionic copper because both the metal and the ligand are introduced into the cell. Toxicity of ionic copper is ameliorated by trivalent metal ions in the growth medium, including those of Mn, Co, Al, Fe and Cr which form a layer of metal (III) hydroxide around the algal cell, adsorb copper and reduce its penetration into the cell. The degree of insolubility of the metal (III) hydroxide is related to its ability to protect against copper toxicity. In addition, manganese and cobalt catalytically scavenge damaging H2O2 and superoxide radicals, respectively, produced by the cell.  相似文献

In response to the recent focus by the U.S. EnvironmentalProtection Agency on bioassessment of lakes, a multimetric index was developed for New Jersey lakes and reservoirs using benthicmacroinvertebrates. Benthic samples were collected fromreference and impaired lakes with muck and intermediate sedimentsin central and northern New Jersey during summer 1997. We used astepwise process to evaluate properties of candidate metrics andselected five for the Lake Macroinvertebrate Integrity Index(LMII): Hilsenhoff Biotic Index (HBI), percent chironomidindividuals, percent collector-gatherer taxa, percentoligochaetes/leeches, and number of Diptera taxa. We scoredmetrics as the fraction of the best expected value (based on allsites) achieved at a site and summed them into the LMII. Evaluation of the LMII showed that it discriminated well betweenreference and impaired lakes and was strongly related to severalpotential stressors. Chemical and physical gradients distinguished between reference and impaired lakes, and the LMIIsummarized these gradients well. The LMII corresponded stronglywith land use, but some lakes with more urban land use stillachieved high scores. Based on a power analysis, the ability ofthe LMII to detect differences in condition was sensitive to thenumber of samples from each lake.  相似文献

According to the hologenome theory, the microbiota contributes to the fitness of the holobiont having an important role in its adaptation, survival, development, health, and evolution. Environmental stress also affects the microbiota and its capability to assist the holobiont in coping with stress factors. Here, we analyzed the diversity of cultivable bacteria associated with Manila clam tissues (mantle, gills, hemolymph) in two non-contaminated sites (Portugal and France) and one metal-contaminated site (Portugal). A total of 240 isolates were obtained. Representative isolates (n = 198) of the overall diversity were identified by 16S rDNA sequencing and subjected to functional characterization. Isolates affiliated with Proteobacteria, Actinobacteria, Firmicutes, and Bacteroidetes. Proteobacteria (mostly Pseudoalteromonadaceae and Vibrionaceae) were dominant in non-contaminated sites while Actinobacteria (mostly Microbacteriaceae) dominated in the metal-contaminated site. The main factor affecting the microbiota composition was contamination. No significant differences were observed between clam tissues and geographic regions. Several isolates tested positive for antibacterial activity, biofilm formation, protease, and siderophore production. The results show that the Manila clam harbors a diverse microbiota that may contribute to clam protection and overall fitness, as well as to its adaptation to stressful environments. In addition, the Manila clam microbiota is revealed as a promising source of novel probiotics with potential application in aquaculture.  相似文献

When field pollution is heterogeneous due to localized pesticide application, as is the case of chlordecone (CLD), the mean level of pollution is difficult to assess. Our objective was to design a decision support tool to optimize soil sampling. We analyzed the CLD heterogeneity of soil content at 0–30- and 30–60-cm depth. This was done within and between nine plots (0.4 to 1.8 ha) on andosol and ferralsol. We determined that 20 pooled subsamples per plot were a satisfactory compromise with respect to both cost and accuracy. Globally, CLD content was greater for andosols and the upper soil horizon (0–30 cm). Soil organic carbon cannot account for CLD intra-field variability. Cropping systems and tillage practices influence the CLD content and distribution; that is CLD pollution was higher under intensive banana cropping systems and, while upper soil horizon was more polluted than the lower one with shallow tillage (<40 cm), deeper tillage led to a homogenization and a dilution of the pollution in the soil profile. The decision tool we proposed compiles and organizes these results to better assess CLD soil pollution in terms of sampling depth, distance, and unit at field scale. It accounts for sampling objectives, farming practices (cropping system, tillage), type of soil, and topographical characteristics (slope) to design a relevant sampling plan. This decision support tool is also adaptable to other types of heterogeneous agricultural pollution at field level.  相似文献

The effect of phenanthrene on the bacterial community was studied on permanent grassland soil historically presenting low contamination (i.e. less than 1 mg kg?1) by polycyclic aromatic hydrocarbons (PAHs). Microcosms of soil were spiked with phenanthrene at 300 mg kg?1. After 30 days of incubation, the phenanthrene concentration decreased rapidly until its total dissipation within 90 days. During this incubation period, significant changes of the total bacterial community diversity were observed, as assessed by automated-ribosomal intergenic spacer analysis fingerprinting. In order to get a deeper view of the effect of phenanthrene on the bacterial community, the abundances of ten phyla and classes (Actinobacteria, Acidobacteria, Bacteroidetes, Alphaproteobacteria, Betaproteobacteria, Gammaproteobacteria, Firmicutes, Verrucomicrobiales, Gemmatimonadetes, and Planctomycetes) were monitored by quantitative polymerase chain reaction performed on soil DNA extracts. Interestingly, abundances of some bacterial taxa significantly changed as compared with controls. Moreover, among these bacterial groups impacted by phenanthrene spiking, some of them presented the potential of phenanthrene degradation, as assessed by PAH-ring hydroxylating dioxygenase (PAH-RHDα) gene detection. However, neither the abundance nor the diversity of the PAH-RHDα genes was significantly impacted by phenanthrene spiking, highlighting the low impact of this organic contaminant on the functional bacterial diversities in grassland soil.  相似文献
