ABSTRACT

Phenanthrene is a toxic and mutagenic pollutant that can cause severe environmental and human health issues. The bioremediation of these polyaromatic hydrocarbons (PAHs) is possible with a biosurfactant by enhancing hydrophobicity. In this study, the production of a biosurfactant by Bacillus pumilus 1529 and its effects on the phenanthrene biodegradation pathway were examined. Biosurfactant production was determined using hemolytic activity, emulsification index, and surface tension. For phenanthrene metabolite detection, samples at 0, 7, 14, and 21 incubation days were analysed by gas chromatography-mass (GC-mass) spectrometry. The results showed that Bacillus pumilus 1529 can reduce surface tension to 22.83?±?1.1?mN?m?1. Furthermore, the GC-mass spectrometry analysis showed that 1-hydroxy-2-naphthoic acid, benzaldehyde, o-phthalic acid, and phenylacetic acid were notable phenanthrene metabolites produced during phenanthrene biodegradation. Biodegraded phenanthrene and its metabolites have a less toxic effect on the germination of safflower seeds than non-biodegraded phenanthrene. The IC50 of phenanthrene on seed germination after biodegradation was increased to approximately 113?mg?L?1. In general, biodegradation aided by biosurfactant producing bacteria contributed to turning the toxic phenanthrene into less harmful metabolites with lower phytotoxicity effects, indicating that its application in the bioremediation of PAHs is promising.  相似文献

Endeis laevis (Grube) is the more littoral of the two British members of the Endeidae (Pycnogonida). The process of vitellogenesis is examined. It closely resembles that of annelids and Limulus polyphemus, in which the majority of the yolk is synthesised within the oocyte with only a small contribution from outside the oocyte. This contrasts with the method in insects in which most of the yolk comes from outside the oocyte. The vitellogenic process is slow, the eggs accumulating yolk over the winter. Although E. laevis has two reproductive cycles each year, only one brood is produced, juveniles occurring over a restricted period (July and early August).  相似文献

A permeable reactive barrier (PRB) for remediation of coal spoil heap drainage in Northumberland, UK, is described. The drainage has typical chemical characteristics of pH<4, [acidity]>1400 mg/L as CaCO3, [Fe]>300 mg/L, [Mn]>165 mg/L, [Al]>100mg/L and [SO4]>6500 mg/L. During 2 years of operation the PRB has typically removed 50% of the iron and 40% of the sulphate from this subsurface spoil drainage. Bacterial sulphate reduction appears to be a key process of this remediation. Treatment of the effluent from the PRB results in further attenuation; overall reductions in iron and sulphate concentrations are 95% and 67% respectively, and acidity concentration is reduced by an order of magnitude. The mechanisms of attenuation of these, and other, contaminants in the drainage are discussed. Future research and operational objectives for this novel, low-cost, treatment system are also outlined.  相似文献

Macropore flow is a key factor determining pesticide fate, but models accounting for this process need parameters that cannot be easily measured. This study was conducted to investigate the use of inverse techniques to estimate parameters controlling macropore flow and pesticide fate in the dual-permeability model MACRO. Undisturbed columns were sampled at three landscape positions (hilltop, slope, hollow) with contrasting texture and organic carbon content. Transient leaching experiments were performed for an anionic tracer and the herbicide MCPA (4-chloro-2methylphenoxy acetic acid) during a 4-mo period, first under natural rainfall, and then under controlled irrigation in the laboratory. The tracer breakthrough for the liner-textured soil from the hilltop showed strong evidence of macropore flow, resulting in a rapid leaching of MCPA, while leaching was minimal from the organic-rich hollow soil, since macropore flow was weaker and adsorption stronger. The MACRO model was linked to the inverse modeling program SUFI (Sequential Uncertainty Fitting) to enable calibration of nine key model parameters. Based on calculated model efficiencies, MACRO-SUFI gave generally good predictions of water movement and tracer and pesticide transport, although some errors were attributed to difficulties in simulating the effects of soil moisture on degradation and the timing of water outflows. Even after calibration, significant uncertainties remained for some key parameters controlling macropore flow. Nevertheless, the parameter estimates were significantly different between landscape positions and could also be related to basic soil properties. The posterior uncertainty ranges could probably be reduced with a more exhaustive sampling of the parameter space and improved experimental designs.  相似文献

The objective of this study was to identify the main sources of variation in pesticide losses at field and catchment scales using the dual permeability model MACRO. Stochastic simulations of the leaching of the herbicide MCPA (4-chloro-2-methylphenoxyacetic acid) were compared with seven years of measured concentrations in a stream draining a small agricultural catchment and one year of measured concentrations at the outlet of a field located within the catchment. MACRO was parameterized from measured probability distributions accounting for spatial variability of soil properties and local pedotransfer functions derived from information gathered in field- and catchment-scale soil surveys. At the field scale, a single deterministic simulation using the means of the input distributions was also performed. The deterministic run failed to reproduce the summer outflows when most leaching occurred, and greatly underestimated pesticide leaching. In contrast, the stochastic simulations successfully predicted the hydrologic response of the field and catchment and there was a good resemblance between the simulations and measured MCPA concentrations at the field outlet. At the catchment scale, the stochastic approach underestimated the concentrations of MCPA in the stream, probably mostly due to point sources, but perhaps also because the distributions used for the input variables did not accurately reflect conditions in the catchment. Sensitivity analyses showed that the most important factors affecting MACRO modeled diffuse MCPA losses from this catchment were soil properties controlling macropore flow, precipitation following application, and organic carbon content.  相似文献

Much work has been done on gaseous emissions and leaching of nitrogenous compounds from whole soil profiles and also from soil surface measurements which are assumed to be mainly due to topsoil activity. In soils with an impervious clay subsoil, the boundary between the topsoil and subsoil may provide an interface for microbial activity, including N transformations. In this study, we investigated movement and transformations of two reactive N species (nitrate and urea) at the subsoil interface using a series of replicate, intact soil blocks, under two contrasting watering regimes. We measured fluxes in both liquid and gaseous phases and demonstrated that nitrate reaching the subsoil interface does not necessarily leach into water systems, but may denitrify immediately and could, therefore, add to atmospheric pollution through N2O production. On the other hand, ammonium reaching the subsoil interface either directly, or after mineralization, appears to be more mobile than expected and has the potential to pollute watercourses.  相似文献

A model is described that may help to resolve uncertainty and controversy over the long-term consequences of sludge applications to arable land, especially with regard to the effects of sludge adsorption characteristics on trace metal solubility and bioavailability (e.g., the sludge "time bomb" or sludge "protection" hypotheses). Mass balances of organic and inorganic material derived from sludge and crop residues are simulated. Each pool has a potentially different adsorption affinity for trace metals, and this leads to changes in the adsorption capacity of sludge-amended soil that influence leaching and crop uptake. Model simulations were compared with measured changes in organic carbon and ethylenediaminetetraacetic acid (EDTA)-extractable cadmium contents in a clay loam soil following 41 years of sludge applications. The model adequately reproduced the data, although discrepancies in the vertical distribution of Cd were attributed to the effects of macropore transport and root-uptake driven recirculation. A Monte Carlo sensitivity analysis demonstrated that the most important parameters affecting leaching and crop uptake were the Cd loading and parameters controlling adsorption, especially the partition coefficient for sludge-derived inorganic material and the exponent regulating the effect of pH on sorption. Scenario simulations show that no general conclusions can be drawn with respect to the validity of the sludge "time bomb" and sludge "protection" hypotheses. Either may occur, or neither, depending on three key system parameters: the ratio of sludge adsorption capacity to the initial adsorption capacity of the soil, the proportion of the sludge adsorption capacity contributed by the inorganic fraction, and the sludge Cd loading.  相似文献

Past research on nitrogen (N) inputs, losses and surpluses focused on separate components of grassland management, i.e., grazed or cut swards and the impact of fertiliser or slurry applications. In practice, however, grassland is both grazed and cut for conservation, and N fertiliser is supplied from both organic and inorganic sources. A whole systems approach was used to evaluate the effects of combinations of management strategies designed to reduce N losses on N budgets, and herbage and animal production in South West England. Three systems with contrasting N inputs were compared: CN, conventional mineral N application and broadcast slurry; TN, tactical mineral N application with slurry injection and the early housing of cattle; GC, a mixed grass/white clover sward with no mineral N addition and slurry injection. Comparisons were made on two contrasting soil types: a freely-draining sandy loam (Gleysol, Site 1), and a poorly drained clay (Luvisol, Site 2). 1 ha farmlets were grazed to a target sward height by beef cattle for a 5-year (Site 1) or a 4-year (Site 2) period. Herbage surplus to grazing requirements was cut for silage. On average, 185 kg N ha−1 was applied annually to treatment TN compared with 280 kg N ha−1 for CN. An additional 76, 102 and 67 kg N ha−1 was applied in slurry to treatments CN, TN and GC, respectively. Substantial reductions in N surpluses were achieved for both treatments TN and GC compared with treatment CN (N surpluses ha−1: 254, 168 and 119 kg at Site 1, and 247 kg, 190 and 73 kg at Site 2, for CN, TN and GC, respectively). The highest N input for treatment CN was associated with the greatest animal and herbage production. More land was required for grazing on treatment GC and less herbage was cut for silage so that self-sufficiency was not attained for winter fodder on this treatment. The early removal of cattle on treatment TN did not result in a significant increase in the amount of herbage cut for silage. It was concluded that the combinations of mitigation options used were successful in reducing N surpluses compared with the conventional N management system, but animal and herbage production was reduced.  相似文献
