Cultures of a chorion biopsy taken from a pregnancy at risk of Bloom's syndrome revealed the high sister chromatid exchange frequency diagnostic of this rare disorder. To obtain the result, cultures were grown under standard conditions, with the addition of 10μM 5′;-bromodeoxyuridine for the final 48 h of incubation. This result demonstrates the feasibility of early prenatal diagnosis of Bloom's syndrome.  相似文献

Mowing is commonly implemented to Artemisia tridentata ssp. wyomingensis (Beetle & A. Young) S.L. Welsh (Wyoming big sagebrush) plant communities to improve wildlife habitat, increase forage production for livestock, and create fuel breaks for fire suppression. However, information detailing the influence of mowing on winter habitat for wildlife is lacking. This information is crucial because many wildlife species depended on A. tridentata spp. wyomingensis plant communities for winter habitat and consume significant quantities of Artemisia during this time. Furthermore, information is generally limited describing the recovery of A. tridentata spp. wyomingensis to mowing and the impacts of mowing on stand structure. Stand characteristics and Artemisia leaf tissue crude protein (CP), acid detergent fiber (ADF), and neutral detergent fiber (NDF) concentrations were measured in midwinter on 0-, 2-, 4-, and 6-year-old fall-applied mechanical (mowed at 20 cm height) treatments and compared to adjacent untreated (control) areas. Mowing compared to the control decreased Artemisia cover, density, canopy volume, canopy elliptical area, and height (P < 0.05), but all characteristics were recovering (P < 0.05). Mowing A. tridentata spp. wyomingensis plant communities slightly increases the nutritional quality of Artemisia leaves (P < 0.05), but it simultaneously results in up to 20 years of decrease in Artemisia structural characteristics. Because of the large reduction in A. tridentata spp. wyomingensis for potentially 20 years following mowing, mowing should not be applied in Artemisia facultative and obligate wildlife winter habitat. Considering the decline in A. tridentata spp. wyomingensis-dominated landscapes, we caution against mowing these communities.  相似文献

Hypoxic events frequently occur in the aquatic environment in association with micro pollutants, including heavy metals. Only a few studies are however available on the uptake and biological responses of heavy metals under hypoxic conditions. To elucidate the phenomenon, mirror carp Cyprinus carpio L. (16.13-16.22 g) were exposed chronically to dietary copper (Cu; 250 and 500 mg kg dry wt.−1) for 30 d under normoxic (8.25 mg O2 L−1) and hypoxic (∼3 mg O2 L−1) conditions and adopting an integrated approach, sub-lethal biomarker responses were determined at different levels of biological organisation. Level of oxidative DNA damage (as determined by modified Comet assay) showed strong significant difference following exposure to dietary Cu level under normoxic (1.6-fold) as well as under hypoxic condition at both Cu levels (2.1 and 2.5-folds respectively). Significant difference was also observed for haematological parameters (i.e. increased red and white blood cells, haematocrit value and haemoglobin concentration). Quantitative histology revealed alterations in tissues (i.e. liver and gills) for hypoxic and all dietary Cu treatment groups under both normoxic and hypoxic conditions suggesting a compensatory response to these organs (< 0.05). The order of Cu accumulation in tissues (as determined by ICP-OES) was liver > intestine > kidney > gill. Interestingly, SGR under both normoxic and hypoxic conditions reduced with elevating Cu levels (p = 0.019). Overall, the results provide evidence for enhanced toxicological responses in fish following exposure to Cu either alone or in combination with hypoxic condition and lends support to the evolving viewpoint that many water quality guidelines should be revisited in terms of new ecotoxicological criteria.  相似文献

Generic water quality guidelines (WQGs) are developed by countries/regions as broad scale tools to assist with the protection of aquatic ecosystems from the impacts of toxicants. However, since generic WQGs cannot adequately account for the many environmental factors that may affect toxicity at a particular site, site-specific WQGs are often needed, especially for high environmental value ecosystems. The Australian and New Zealand Guidelines for Fresh and Marine Water Quality provide comprehensive guidance on methods for refining or deriving WQGs for site-specific purposes. This paper describes three such methods for deriving site-specific WQGs, namely: (1) using local reference water quality data, (2) using biological effects data from laboratory-based toxicity testing, and (3) using biological effects data from field surveys. Two case studies related to the assessment of impacts arising from mining operations in northern Australia are used to illustrate the application of these methods. Finally, the potential of several emerging methods designed to assess thresholds of ecological change from field data for deriving site-specific WQGs is discussed. Ideally, multiple lines of evidence approaches, integrating both laboratory and field data, are recommended for deriving site-specific WQGs.  相似文献

The mixing efficiency of a plume in a filling box in an asymptotic state is 1/2, independent of the details of this state or how the plume is modelled. The mixing efficiency of a plume in an emptying-filling box in steady state is \(1 - \xi \), where \(\xi = h/H\), the depth of the ambient layer h normalised by the height of the box H. A deeper mixed layer therefore corresponds to a higher mixing efficiency. These results shed light on the interpretation of mixing efficiencies of open and closed systems.

A newly constructed coastal revetment at Colhuw Beach in the Glamorgan Heritage Coast, Wales, UK, was analysed using a recently developed environmental risk assessment package (ERA). Conflict with Heritage Coast conservation objectives is apparent and the act of building such a structure is questioned for a location where maintenance ofnatural beauty is an axiom of the coastal management philosophy. The likelyhood of revetment related environmental consequences of significant magnitude was analysed using estimated probability values derived from Bayesian theory. The damaging impact of the structure on the natural environment is out of all proportion to the level of storm protection afforded to the site. Analyses, such as carried out via ERA, would have shown planners, engineers and environmentalists, the inadequacies of such an investment and management strategy. The ERA approach can help to introduce greater clarity and consistency into decision making processes.  相似文献
