Fumonisin B1 (FB1) is a secondary metabolite produced by Fusarium verticillioides or Fusarium proliferatum, which present in food and feed. It causes hazardous effects on human and animal health. A monoclonal antibody (mAb) against FB1 was produced and a simple, reliable and sensitive, competitive, indirect enzyme-linked immunosorbent assay (ci-ELISA) for detection of FB1 was developed and the experiment conditions were optimized. The coating concentration of FB1-ovalbumin (FB1-OVA) was 500 ng mL?1, the action concentrations of anti-FB1 mAb and goat anti-mouse IgG were 1.28 × 104 and 1:5000, respectively. The 50% inhibitory concentration (IC50) was 11 ng mL?1, with a detectable range of 1.25–250 ng mL?1, and a limit of determination (LOD) of 1.15 ng mL?1. The cross-reactivity (CR) of the antibody against fumonisin B2 (FB2) was 60.4, and <1% against deoxynivalenol (DON), aflatoxin B1 (AFB1), ochratoxin A (OTA) or zearalenone (ZEN). In spiked samples (250 ng g?1, 500 ng g?1, 1000 ng g?1), the mean recoveries ranged from 86.7 ± 5% to 102 ± 4%, and the coefficient of variation (CV) ranged from 3% to 10%. A survey of 96 corn samples from Bozhou, Fuyang, Bengbu, and Hefei, in Anhui province, China, was performed. Frequencies of FB1 contamination were 83.3%, 95.8%, 20.8% and 91.7%, and the mean concentrations of positive samples were 0.702 μg kg?1, 0.883 μg kg?1, 0.074 μg kg?1, and 0.276 μg kg?1, respectively. The results of this study suggest that the ci-ELISA developed in this study can be used to identify FB1 in corn, furthermore, further study is needed to investigate FB1 contamination in food and feed to prevent its harmful health effects. 相似文献

The designation of biodiesel as an environmental-friendly alternative to diesel oil has improved its commercialization and use. However, most biodiesel environmental safety studies refer to air pollution and so far there have been very few literature data about its impacts upon other biotic systems, e.g. water, and exposed organisms. Spill simulations in water were carried out with neat diesel and biodiesel and their blends aiming at assessing their genotoxic potentials should there be contaminations of water systems. The water soluble fractions (WSF) from the spill simulations were submitted to solid phase extraction with C-18 cartridge and the extracts obtained were evaluated carrying out genotoxic and mutagenic bioassays [the Salmonella assay and the in vitro MicroFlow® kit (Litron) assay]. Mutagenic and genotoxic effects were observed, respectively, in the Salmonella/microsome preincubation assay and the in vitro MN test carried out with the biodiesel WSF. This interesting result may be related to the presence of pollutants in biodiesel derived from the raw material source used in its production chain. Background, aim, and scope It has been known that the pollutants of electronic wastes (E-wastes) can lead to severe pollution to the environment. It
has been reported that about 50% to 80% of E-wastes from developed countries are exported to Asia and Africa. It has become
a major global environmental problem to deal with ‘E-wastes’. E-waste recycling has remained primitive in Jinghai, China.
This not only produces enormous environmental pollution but also can bring about toxic or genotoxic effects on the human body,
threatening the health of both current residents and future generations living in the local environment. The concentration
of lead in the blood of children in the E-waste polluted area in China is higher than that of the control area. But little
is known about the cytogenetic effect to human beings caused by the pollution of E-wastes. In the present study, experiments
have been performed to investigate the genetics of permanent residents of three villages with numerous E-waste disposal sites
and to analyze the harmful effects of exposure to E-wastes.
Materials and methods In total, 171 villagers (exposed group) were randomly selected from permanent residents of three villages located in Jinghai
County of Tianjin, China, where there has been massive disposal of E-wastes. Thirty villagers were selected from the neighboring
towns without E-waste disposal sites to serve as controls. Chromosomal aberrations and cytokinesis blocking micronucleus were
performed to detect the cytogenetic effect, dic + r (dicentric and ring chromosome), monomer, fragments (acentric fragments,
minute chromosomes, and acentric rings), translocation, satellite, quadriradial, total aberrations, and micronuclear rate
were scored for each subject. DNA damage was detected using comet assay; the DNA percentage in the comet tail (TDNA%), tail
moment (TM), and Olive tail moment (OTM) were recorded to describe DNA damage to lymphocytes.
Results The total chromosome aberration rates (5.50%) and micronuclear rates (16.99%) of the exposure group were significantly higher
than in the control group (P = 0.000). The percentage of DNA in the comet tail, tail moment, and Olive tail moment detected by comet assay showed that
there was a significant difference in DNA damage in the exposure group (P = 0.000). The chromosome aberration, micronucleus rate, and DNA damage observed in women were significantly higher than those
in men. Chromosome aberration and micronuclear rates of both smokers and non-smokers in the exposure group are obviously higher
than that in the control group (P = 0.000).
Discussion The use of outdated (and unsafe) ways to deal with E-wastes can lead to exposure to a variety of substances harmful to human
health. The components of pollution may enter the human body through the air, drinking water, and food chain to damage human
genetic material, resulting in genomic instability. The rates of chromosomal aberration, micronucleus formation, and the degree
of DNA damage in women in the group exposed to electronic waste were significantly higher than in men. The reason for this
may be concerned with the traditional lifestyle of the local residents or the difference of sensitivity to the exposure to
E-wastes or any others. Further investigations are needed to provide evidence to demonstrate this.
Conclusions Here, we report the obviously cytogenetic toxicity to the exposure population by the E-waste pollution for the first time.
E-waste pollution may be a potential agent of genetic mutation, and may induce cytogenetic damage within the general population
exposed to the pollution. These findings need to be considered, and steps should be taken to protect the current population
and future generations from the effects of pollution with E-wastes.
Recommendations and perspectives The above results remind us that the impact of E-waste recycling on environmental quality of Jinghai should be evaluated soon.
Moreover, it is urgent for the government to prohibit E-waste import and its processing by outdated ways. The future studies
such as pollutant details of drinking water, air, and soil in the area as well as epidemiological investigations on the harmful
effect to children must be performed eagerly. All the data available do provide a compelling case for immediate action in
both countries to address workplace health and safety and waste management.

An in vitro approach was performed to assess the quality of drinking water collected at two treatment/distribution networks located near the source (Plant #1) and the mouth of River Po (Plant #2). The water was sampled at different points of each distribution network, before (raw water) and after the chlorine dioxide disinfection, and in two points of the pipeline system to evaluate the influence of the distribution system on the amount and quality of the disinfection by-product. Cytotoxicity and genotoxicity of water extracts were evaluated in human peripheral lymphocytes and Hep-G2 cells by the use of the micronucleus (MN) test and Comet assay. Raw water samples of both plants induced cytotoxic effects, but not the increases of MN frequency in Hep-G2 cells and in human lymphocytes. Increases of DNA damage in human leukocytes was detected by Comet assay for raw water of Plant #2 at concentration ≥ 0.25 Leq/mL. The disinfection process generally has reduced the toxicity of water samples, even if potential direct DNA-damaging compounds have been detectable in drinking water samples. The proposal approach, if currently used together with chemical analysis, can contribute to improve the monitoring drinking water. 相似文献

Biomonitoring studies have increased as a consequence of risks and effects to human health on exposure to environmental contaminants, mainly air pollutants. Genetic biomarkers are useful tools for the early assessment of exposure to occupational and environmental pollution. The objective of the present study was to investigate genotoxic effects on people residing and/or working downwind from an oil refinery in southern Brazil and the mutagenic activity of airborne particulate matter (PM10). Samples of peripheral blood and buccal mucosa cells were evaluated using the single-cell gel electrophoresis assay (comet assay) and the micronucleus (MN) assay, respectively. PM10 samples were collected in the target site and the organic matter extraced with dichloromethane was assessed for mutagenic activity in the Salmonella/microsome assay. The exposed group (n = 37) was compared to a reference group (n = 37) of subjects living in an urban area with limited traffic and industrial influence, located far from the main industrial areas. All PM10 organic extracts showed mutagenic positive responses and the effect decreased in the presence of S9 mix indicating that the predominant compounds present were direct-acting mutagens. The responses of YGs strains are consistent with aromatic amines and nitroarenes being present in the PM10 extracts. The group in the area under the influence of the oil refinery (exposed group) showed significantly higher DNA damage in lymphocytes than the reference group. The MN frequencies in buccal mucosa were very low for both groups and no difference between groups was observed. No association was found between age and tobacco smoking habit and level of DNA damages measured by the comet assay. The results indicate that the comet assay was a sensitive tool to detect DNA damage in subjects under the influence of an oil refinery, with marked genotoxic activity in the atmospheric environment. 相似文献

A previous study showed that the cancer mortalities are higher for residents who lived nearby the Songhua River heavily polluted by organic contamination. It is important to determine its risk of carcinogenic potential. Short-term genotoxic bio-assays using Salmonella, Sister Chromatid Exchange (SCE), and Micronuclei (MN) assays were employed to examine the genotoxic activity of ether extracts of water samples taken from the Songhua River. The results of the Salmonella bioassay indicated that there were indirect frame-shift mutagens in the water samples. A dose-response relationship for the SCE and MN assays was obtained. These results showed that organic extracts of water samples have genotoxic activity and the risk of carcinogenic potential to human health. The mutagenesis of water samples had changed compared to the results in 1994-1995. An increasing trend of risk of carcinogenic potential in the Songhua River after ten years should be noted and needs to be studied further. 相似文献

There is a growing interest for the application of biomakers to field-collected earthworms. Therefore we have evaluated the usability of native populations of endogeic, widely distributed earthworm Aporrectodea caliginosa in the assessment of soil genotoxicity using the Comet assay. Validation of the Comet assay on earthworm coelomocytes has been established using commercially available Eisenia fetida exposed to copper, cadmium, and pentachlorophenol, along with A. caliginosa exposed to copper in a filter paper contact test. Neutral red retention time (NRRT) assay was conducted on copper exposed and field-collected earthworms. Significant DNA and lysosomal damage was measured using Comet and NRRT assays in native populations of A. caliginosa sampled from the polluted soils in the urban area in comparison to the earthworms from the reference site. The results of this study confirm the employment of A. caliginosa as a suitable species for the in situ soil toxicity and genotoxicity field surveys. 相似文献

The present study was conducted in order to investigate pro-oxidant activity of dimethoate in liver and brain tissues following sublethal pesticide exposure for 5, 15 and 30 d by using SOD, GPx, CAT enzyme activities and lipid peroxidation as biomarkers as well as DNA damaging potential via detecting% Tail DNA, Tail moment and Olive tail moment as endpoints in erythrocytes of Oncorhynchus mykiss in an in vitro experiment. Antioxidant enzyme activities were found to elicit two staged response which was an initial induction followed by a sharp inhibition in liver tissue while a sustained increase in GPx activity and slight stimulation in SOD activity were detected in brain tissue. Lipid peroxidation showed an ascending pattern throughout the exposure period in both tissues and a decreasing trend was determined in tissue protein levels which was proved to be positively correlated with duration. Similar findings were obtained from outcomes preferred to quantify DNA damage and TM was decided to reflect the extent of damage more sensitively because of determined positive correlation with concentrations applied. Considering these results, it can be concluded that oxidative stress condition evoked by dimethoate could not be responded effectively and genotoxic nature of pesticide was proven by determined clastogenic effect possibly via being an alkylation agent or stimulating the production of reactive species. 相似文献
