The thyroid hormone agonist/antagonist activities of halogenated derivatives of bisphenol A (BPA) were assessed using a yeast two-hybrid assay incorporating the human thyroid hormone α (TRα), both with and without possible metabolic activation by rat liver S9 preparation. In the absence of the rat liver S9 preparation, 3,3′,5,5′-tetrabromobisphenol A (TBBPA), 3,3′,5,5′-tetrachlorobisphenol A (TCBPA), and 3,3′,5-trichlorobisphenol A (3,3′,5-triClBPA) exhibited agonist activity, whereas 3-chlorobisphenol A (3-ClBPA), 3,5-dichlorobisphenol A (3,5-diClBPA), 3,3′-dichlorobisphenol A (3,3′-diClBPA), and BPA did not. The activities of TBBPA and TCBPA increased markedly (7.6-fold and 3.1-fold, respectively) after their metabolic activation with the rat liver S9 preparation. TBBPA, TCBPA, and 3,3′,5-triClBPA inhibited the binding of triiodothyronine (T3) to TRα at 2 × 10−5 M without rat liver S9 treatment and 4 × 10−6 M with rat liver S9 treatment, demonstrating their T3 antagonist activity. These results revealed that metabolic activation by rat liver S9 significantly increased the agonist/antagonist potential of some halogenated BPAs. 相似文献

为研究石油烃对海洋生物的毒性效应,将栉孔扇贝(Chlamys farreri)暴露于0.08、0.21和0.88mg·L-1石油烃中,采用单细胞凝胶电泳实验(彗星实验)技术检测不同暴露时间扇贝血淋巴细胞的DNA损伤程度,对照组中石油烃背景浓度为0.04mg·L-1。结果显示,低浓度(0.08mg·L-1)的石油烃短期(<7d)内即可导致栉孔扇贝血淋巴细胞的DNA损伤,并且随石油烃浓度的增大和暴露时间的延长,DNA损伤程度增加,石油烃浓度达0.88mg·L-1时,DNA损伤程度已非常严重。3d恢复实验后,各浓度组DNA损伤又均有不同程度的恢复。研究表明,彗星实验是检测石油烃对海洋贝类DNA损伤的一种有效手段,贝类血淋巴细胞DNA损伤有望成为石油烃污染的一种生物标志物,用于海洋污染的早期预警监测。 相似文献

Benzophenones (BPs) are a class of widely used UV filters, which have been frequently detected within multiple environmental matrices. Disinfection is a necessary process in water treatment processes. The transformation behaviors and toxicity changes of 14 BP-type UV filters during chlorination disinfection treatment were investigated in this study. A new index, the acute toxicity formation potential, was proposed to evaluate the toxicity changes and potential risks of BP-type UV filters during chlorination treatment. It was found that 13 of 14 BP-type UV filters exhibited toxicity decreases in the chlorination disinfection process, more or less, while one showed a toxicity increase. The toxicity changes were dependent on substitution effects, such that 2,4-di-hydroxylated or 3-hydroxylated BPs exhibited significant toxicity decreases after chlorination treatment due to the ready cleavage of the aromatic ring. Importantly, the acute toxicity changes could be duplicated in an ambient water matrix. 相似文献

采用GC-MS方法分析了太湖梅梁湾水样的主要有机污染物成分和含量,共检测出15种目标化合物,表明该区域水体已经受到有机物污染;纤细裸藻(Euglena gracilis)在低浓度浓缩水样处理下(1倍组)生长无显著影响,叶绿素a、b含量和类胡萝卜素含量比对照组显著增加,而高浓度下(5倍和10倍组)生长明显受到抑制,叶绿素a和类胡萝卜素含量呈下降趋势;抗氧化酶系统中SOD和POD活性呈上升趋势,显示有机污染物胁迫可诱导抗氧化酶活性;彗星试验中,Olive尾矩和尾动量增加,纤细裸藻细胞DNA损伤程度随着太湖水样污染物浓度增加而加重,呈现出明显的剂量-效应关系,提示太湖梅梁湾水样具有潜在致突变性.结果表明,慧星试验和SOD活性试验结合使用在水环境中的遗传毒性监测方面具有较大的应用价值,适合作为水体有机污染监测的生物标志物. 相似文献

Several factors such as smoking habits, diet, occupational, and environmental exposure to chemical carcinogens influence the overall level of DNA damage. In 69 healthy adult volunteers’ polymorphisms of glutathione S-transferase (GST), an enzyme which participates in the metabolism of a broad range of carcinogens and endogenous compounds were determined. The level of DNA damage was assessed by comet assay and classified according to GSTT1/M1 genotype and smoking habits. GSTM1 null genotype was recognized in 48% of studied subjects and GSTT1 null genotype in only two cases (3%). In subjects carrying GSTT1/M1 alleles a significantly lower degree of DNA damage, determined as % DNA in the comet's tail, than in null individuals was noted. However, the results obtained did not indicate that in studied subjects an elevated endogenous level of DNA damage may be significantly related to smoking habits. 相似文献

Genotoxic effects of ivermectin (IVM) and its commercial formulation ivomec® (IVM 1.0%) were studied on Aedes albopictus larvae (CCL-126?) cells by sister chromatid exchange (SCE) and single cell gel electrophoresis (SCGE) while cytotoxicity was determined by cell-cycle progression (CCP), proliferative rate index (PRI), mitotic index (MI), 3(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT), and neutral red (NR) endpoints within a 1–250 µg mL?1 concentration range. While IVM and ivomec® did not markedly affect SCE frequencies, these agents induced DNA-strand breaks enhancing both slightly damaged and damaged cells at 25–50 and 5–50 µg mL?1 IVM and ivomec®, respectively. Both compounds exerted a delay in CCP and reduction of PRI at 10 µg mL?1. Cytotoxicity was observed at concentrations higher than 25 µg mL?1. A marked reduction of about 98% and 94% of MI compared to controls was noted with 25 µg mL?1 of IVM and ivomec®, respectively. NR and MTT assays revealed that both compounds induced a cell growth inhibition within the 1–250 µg mL?1 concentration range. Data indicated that IVM and ivomec® exert both genotoxicity and cytotoxicity in insect cells in vitro, at least in A. albopictus larvae CCL-126? cells. 相似文献

High copper (Cu) accumulation in medicinal plants might favor lipid peroxidation and hence affect antioxidant responses. This effect was studied by determining antioxidant activities in the water extracts of medicinal plants from Cu mining impact site and compared with control samples. Antioxidant activities of the extracted medicinal plants were assayed by measuring (1) total phenolic and flavonoid contents, (2) diphenyl-1-picrylhydrazyl (DPPH)? free radical scavenging activity, and (3) inhibition capacities of lipid peroxidation. The total phenolic as well as total flavonoid contents of the mining impact samples and control samples are not significantly different. However, inhibition of lipid peroxidation capacities was significantly less (12–60%) for mining impact samples as reflected by the IC50 values. These anomalies were attributed to high concentrations of Cu (2–4-fold higher) in mining impact samples as measured by inductively coupled plasma mass spectrometry. A positive correlation was observed between Cu levels and IC50 values for the inhibition of lipid peroxidation for mining impact samples. 相似文献

AbstractThe in vivo genotoxic potential of bisphenol A using the comet assay in mice and in human sperm cells in vitro without metabolizing enzymes was studied. Male mice were exposed by oral gavage to the following doses of bisphenol A (0 125, 250 and 500?mg/kg body weight). DNA damage was investigated in liver, kidney, testes, urinary bladder, colon and lungs cells. In testicular cells, a significant increase in DNA strand breaks was observed in the lowest, but not in the medium or highest dose groups. Histopathological investigation of the testicular samples did not show any treatment dose-related effects. No DNA strand breaks were observed in any of the other investigated tissues.
