Cobalt and silver are toxic for cells, but mechanisms of this toxicity are largely unknown. Analysis of Corynebacterium glutamicum proteome from cells grown in control and cobalt or silver enriched media was performed by two dimensional gel electrophoresis (2DE) followed by mass spectrometry. Our results indicate that the cell adapted to cobalt stress by inducing five defense mechanisms: Scavenging of free radicals, promotion of the generation of energy, reparation of DNA, reparation and biogenesis of Fe-S cluster proteins and supporting and reparation of cell wall. In response to the detoxification of Ag+ many proteins were up-regulated, which involved reparation of damaged DNA, minimizing the toxic effect of reactive oxygen species (ROS) and energy generation. Overexpression of proteins involved in cell wall biosynthesis (1,4-alpha-glucan branching enzyme and nucleoside-diphosphate-sugar epimerase) upon cobalt stress and induction of proteins involved in energy metabolism (2-methylcitrate dehydratase and 1, 2-methylcitrate synthase) upon silver demonstrate the potential of these enzymes as biomarkers of sub-lethal Ag+ and Co toxicity. 相似文献

双室微生物燃料电池处理硝酸盐废水 总被引:2,自引:1,他引:2
基于双室微生物燃料电池(microbial fuel cell,MFC),针对阴极分别接种活性污泥(A-MFC)和反硝化细菌(D-MFC),研究其产电情况和硝酸盐废水去除效果。结果表明,在产电的同时都可有效去除废水中的硝酸盐污染物。在外接电阻100Ω的情况下,2种MFC均具有良好的产电性能,A-MFC和D-MFC达到的最大输出电压分别为119.6 mV和117.2mV,最大功率密度分别为23.40 mW/m2和26.63 mW/m2;同时两者在阴极室的平均反硝化速率分别为1.86 mg/(L.d)和2.19 mg/(L.d),阳极室的平均COD去除率分别为81.9%和82.4%。另外,通过扫描电镜观察可知,A-MFC和D-MFC阴极碳布表面形貌存在差异,并且阳极与阴极碳布表面形貌差异显著。 相似文献

Nitrate contamination of water sources is a concern where large amounts of nitrogen fertilizers are regularly applied to soils.
Ingested nitrate from dietary sources and drinking water can be converted to nitrite and ultimately to N-nitroso compounds,
many of which are known carcinogens. Epidemiologic studies of drinking water nitrate and cancer report mixed findings; a criticism
is the use of nitrate concentrations from retrospective drinking water data to assign exposure levels. Residential point-of-use
nitrate data are scarce; gaps in historical data for municipal supply finished water hamper exposure classification efforts.
We used generalized linear regression models to estimate and compare historical raw water and finished water nitrate levels
(1960s--1990s) in single source Iowa municipal supplies to determine whether raw water monitoring data could supplement finished
water data to improve exposure assessment. Comparison of raw water and finished water samples (same sampling date) showed
a significant difference in nitrate levels in municipalities using rivers; municipalities using other surface water or alluvial
groundwater had no difference in nitrate levels. A regional aggregation of alluvial groundwater municipalities was constructed
based on results from a previous study showing regional differences in nitrate contamination of private wells; results from
this analysis were mixed, dependent upon region and decade. These analyses demonstrate using historical raw water nitrate
monitoring data to supplement finished water data for exposure assessment is appropriate for individual Iowa municipal supplies
using alluvial groundwater, lakes or reservoirs. Using alluvial raw water data on a regional basis is dependent on region

Statistical methods and a Geographic Information System (GIS) were used to investigate potential indicators of ground water vulnerability to agricultural chemical contamination in a representative area of the Mississippi River alluvial aquifer. A total of 47 wells were sampled for analysis of nitrate, phosphorus, potassium, and 13 pesticides commonly-used in the area. Ten soil and hydrogeologic variables and five ground water vulnerability indices were examined to explain the variations of chemical concentrations. The results showed that no individual soil or hydrogeologic variables or their linear combinations could explain more than 25% of the variation of the chemical concentrations. A quadratic response surface model with the values of confining unit thickness, slope, soil permeability, depth to ground water, and recharge rate accounted for 62% of the variation of nitrate, 43% of P, and 83% of K, suggesting that the interactions among soil and hydrogeologic variables were significant. Observed trends of decreasing nitrate and P concentrations with increasing well depth and/or depth to ground water seemed to correlate with carbonate equilibrium in the aquifer and more reduced environment with depth. In view of uncertainties involved, it was recognized that the limitations associated with input data resolution used in GIS and the formulation of leaching indices limited their use for predicting ground water vulnerability. Misuse of pesticides could be another factor that would complicate the relationships between pesticide concentrations and the vulnerability indices. 相似文献

为了解武汉市秋季PM_(2.5)中硫酸盐、硝酸盐理化特征,2016年9—11月利用热还原法在线连续监测分析系统对此进行了采样分析,并同步收集气象因子和离子色谱方法监测结果。结果表明,硫酸盐、硝酸盐的热还原分析方法与离子色谱法的相关系数分别为0.88、0.94;PM_(2.5)中硫酸盐、硝酸盐的水溶性部分占比达92.5%,难溶性部分为7.5%;空气质量为优、良和轻度污染时,硫酸盐、硝酸盐与PM_(2.5)的占比分别为45%、42%、45%;硫酸盐、硝酸盐在降水日和非降水日平均质量浓度分别为(19.6±18.5)μg/m~3和(31.0±9.1)μg/m~3;硝酸盐与硫酸盐的质量比为1.1,高于国内其他城市,与武汉市机动车保有量大幅增加有关。 相似文献

为评价物料混合对危险化学品热化学行为的影响,采用热分析-红外/质谱联用技术(TA-FTIR/MS)研究硝酸铵、柠檬酸和蔗糖的混合物的热化学行为。通过对硝酸铵及其混合物的分解温度和分解过程中逸出气体的分析,发现硝酸铵、柠檬酸和蔗糖的混合物热分解温度分别降低至135℃和153℃,而硝酸铵与甲基纤维素的混合物分解温度与硝酸铵基本相同为201℃,且各混合体系的气相分解产物均有氮氧化物(NOX),H2O和CO2。结果表明,混合物受热后其中的硝酸铵首先分解为硝酸和氨气;酸性物质和还原性物质由于对硝酸的分解反应有催化作用,使得混合物的热稳定性下降。 相似文献

微生物通过异化性硝酸盐还原成铵(DNRA)途径,硝态氮转化为仍可生物再利用的铵盐。以琥珀酸钠、柠檬酸钠、酒石酸钾钠为碳源,研究碳源的差异对有氧条件下微生物通过DNRA途径产铵的影响。结果显示,以琥珀酸钠和柠檬酸钠为碳源,初始浓度为20 mmol/L是较佳的实验条件,此时C/N约为1.5~2.0,NH4+-N质量浓度30.0~45.0 mg/L,最高产铵率分别为29.9%和27.0%;以酒石酸钾钠为碳源则在初始浓度为30 mmol/L,C/N约为2.0,NH4+-N质量浓度为40.0~45.0 mg/L时,最高产铵率为30.7%。反硝化和DNRA过程是同时存在的,培养液中NO3--N浓度的下降伴随着中间产物NO2--N的积累和NH4+-N浓度的升高。 相似文献

臭氧(O3)被认为是重要的气污染物之一,水稻又是主要的粮食作物,因而准确地评估O3浓度升高对水稻生长发育的影响具有十分重要的意义。采用开顶式气室法模拟研究了O3对水稻叶片可见伤害症状、氮代谢、脯氨酸和谷胱甘肽含量的影响。结果显示,O3污染胁迫会导致水稻叶片产生明显的伤害症状,具体表现为:老叶叶鞘褪绿,有褐斑,直至完全干枯;稻穗小且黄化,籽粒不饱满;水稻成熟期提前等。O3浓度升高对水稻叶片的硝酸还原酶活性有显著影响。当O3浓度为40、80和120nL.L-1时,水稻叶片硝酸还原酶活性与对照组相比均降低,其中,分蘖期分别降低了25.3%、67.4%和86.3%;拔节期分别降低了57.4%、75.7%和97.8%;抽穗期分别降低了91.0%、97.2%和99.3%;乳熟期分别降低了89.5%、89.5%和96.7%。水稻叶片铵态氮和硝态氮含量随着O3浓度的升高而显著地降低,例如当O3浓度为40、80和120nL.L-1时,与对照相比,水稻叶片硝态氮含量分别降低46.3%、52.7%和65.7%,铵态氮含量分别降低6.5%、12.9%和43.4%。O3污染胁迫下水稻叶片脯氨酸含量在不同生长期变化不同,分蘖期、拔节期和抽穗期脯氨酸含量在40nL.L-1浓度O3熏蒸下急剧地提高,但是随着O3浓度的增加,脯氨酸含量又不断地降低。在水稻乳熟期,脯氨酸含量均随着O3浓度的增加而显著地下降。O3污染胁迫导致水稻叶片还原型谷胱甘肽(GSH)含量显著低于对照组,而氧化型谷胱甘肽(GSSG)含量显著高于对照组。当O3浓度为40、80和120nL.L-1时,乳熟期水稻叶片GSH含量分别比对照组降低68.7%、80.2%和78.2%,GSSG含量分别比对照提高494.4%、527.2%和439.8%。研究表明,O3污染胁迫对水稻叶片氮代谢和抗氧化系统产生了极显著的影响。 相似文献

Nanoscale iron particles (nZVI) is one of the most important engineered nanomaterials applied to environmental pollution control and abatement. Although a multitude of synthesis approaches have been proposed, a facile method to screen the reactivity of candidate nZVI materials produced using different methods or under varying synthesis conditions has yet been established. In this study, four reaction parameters were adjusted in the preparation of borohydride-reduced nZVI. The reductive properties of the resultant nanoparticles were assayed independently using two model aqueous contaminants, Cu(II) and nitrate. The results confirm that the reductive reactivity of nZVI is most sensitive to the initial concentration of iron precursor, borohydride feed rate, and the loading ratio of borohydride to ferric ion during particle synthesis. Solution mixing speed, in contrast, carries a relative small weight on the reactivity of nZVI. The two probing reactions (i.e., Cu(II) and nitrate reduction) are able to generate consistent and quantitative inference about the mass-normalized surface activity of nZVI. However, the nitrate assay is valid in dilute aqueous solutions only (50 mg·L−1 or lower) due to accelerated deactivation of iron surface at elevated nitrate concentrations. Additional insights including the structural and chemical makeup of nZVI can be garnered from Cu(II) reduction assessments. The reactivity assays investigated in this study can facilitate screening of candidate materials or optimization of nZVI production parameters, which complement some of the more sophisticated but less chemically specific material characterization methods used in the nZVI research. 相似文献

从研究我国典型集约化蔬菜种植区地下水中硝酸盐的来源和浓度人手,进行富集、培养、分离、纯化,筛选出一株具有反硝化作用的菌株,通过形态学、革兰氏染色结合16SrDNA序列同源性分析鉴定,其鉴定结果为农杆菌(Agrobacterium sp.).该研究为开展地下水硝酸盐污染的生物修复储备宝贵的菌种资源,为地下水中硝酸盐污染的原位微生物修复和相关污水的生物处理提供微牛物基础,对于经济、有效的解决地下水硝酸盐污染和水资源短缺的问题有着十分重要的意义. 相似文献
