Rhodococcus sp. Ns对硝基苯酚的好氧生物降解   总被引:3,自引:2,他引:1  
通过驯化富集培养,从红树林底泥中分离出6株硝基苯酚降解菌,其中Rhodococcus sp. Ns为对硝基苯酚(PNP)与邻硝基苯酚(ONP)的高效降解菌.在好氧条件下该菌可以耐受小于1.8 mmol/L的PNP,能够利用PNP和ONP为唯一碳源、能源和氮源生长并将其完全矿化.研究了Rhodococcus sp. Ns在不同pH、盐度与浓度范围下,PNP的降解特性并探讨了该菌降解PNP的途径.实验得出该菌在盐度<5‰、 pH>5的条件下能较快生长,1.5 mmol/L的PNP在96h内被完全降解,并检测到至少2种中间产物4-硝基儿茶酚(4-nitrocatechol)和1,2,4-苯三酚 (1,2,4-benzenetriol).红树林底泥中固有的细菌对PNP和ONP具有高效降解作用.  相似文献

苯酚降解红球菌的分离鉴定及降解特性研究   总被引:1,自引:0,他引:1  
采用富集培养的方法,从唐山污水处理厂附近的植物根际土中分离得到1株高效苯酚降解菌11-111。该菌株为革兰氏阳性菌,可以在以苯酚为唯一碳源和能源的无机盐培养基上生长,能够耐受最高浓度为2 000 mg/L的苯酚。对该菌株的降解性能研究表明,在温度24~32℃,pH值6.5~8.5,装液量≤20 mL(100 mL摇瓶)范围内,摇床转速为160 r/min,菌株对初始浓度500~1 500 mg/L的苯酚均能有效降解。根据该菌株的形态、生理生化特性和16S rRNA基因序列同源性分析结果,鉴定为红球菌属(Rhodococcus sp.)菌株。该菌株具有较强的环境适应能力和苯酚降解能力,有较高的研究价值及应用前景。  相似文献

红球菌属是有机污染物降解的重要微生物之一。由于红球菌能够适应各种各样的底物环境,具有极强的有机溶剂耐受性和很宽的降解谱,同时它们还能通过产生表面活性剂和改变细胞表面组成结构来提升自身对于疏水性环境的适应能力,因此,红球菌在石油污染物降解及石油污染的生物修复等领域有着极其重要的应用价值。文章基于近年来在红球菌降解石油烃方面的研究进展,从红球菌适应疏水性环境的机制、石油烃中烷烃、环烷烃、芳香烃的降解途径等几个方面进行综述,同时对今后研究的方向进行了展望。  相似文献

Organochlorine pesticides (OCPs) and polychlorinated biphenyls (PCBs) residue levels were determined in 53 unhatched eggs from greater roseus flamingos (Phoenicopterus ruber). Eggs were collected in 1996 from the National Park of Doñana (Guadalquivir marshes, Southwest Spain), immediately after one breeding colony abandoned the nesting site due to predator attacks. The main metabolite of p,p′-DDT, p,p′-DDE, was the OCP residue found at higher concentrations, with a geometric mean of 721 ng/g wet weight. Residues of other pesticides, including some hexachlorocyclohexane isomers, hexachlorobenzene, aldrin, heptachlor, and heptachlor-epoxide, were detected at much lower concentrations. The sum of PCBs was 528 ng/g, with PCB congeners #187 and #153 being the most prominent in eggs. The pattern observed in these compounds of industrial origin corresponded more to Aroclor 1260 than to any other commercial mixture. Levels of organochlorine residues indicate a medium degree of exposure, and they are not considered of any concern for the flamingo population. In particular, neither p,p′-DDE nor PCB levels were found to be correlated with the eggshell thickness.  相似文献

The goals of the present work were as follows: to obtain the dormant forms of R. opacus 1cp; to study the phenotypic variability during their germination; to compare phenotypic variants during the growth on selective and elective media; and to reveal changes in the ability of the strain to destruct xenobiotics that had not been degradable before dormancy. It was shown that Rhodococcus opacus 1cp (the strain degrading chlorinated phenols) became able to utilize a broader spectrum of xenobiotics after storage in the dormant state. Germination of the dormant forms of R. opacus 1cp on an agarized medium was followed by emergence and development of phenotypic variants that could grow on 4-chlorophenol and 2,4,6-trichlorophenol without adaptation. The cells of R. opacus 1cp phenotypic variants also utilized all of the tested chlorinated phenols: 2,3-, 2,5-, and 2,6-dichloro-, 2,3,4- and 2,4,5-trichloro-, pentachlorophenol, and 1,2,4,5-tetrachlorobenzene in concentrations up to 60 mg/L, though at the lower rates than 4-CP and 2,4,6-TCP. The improved degradation of chlorinated phenols by R. opacus strain 1cp exposed to the growth arrest conditions demonstrates the significance of dormancy for further manifestation of the adaptive potential of populations. A new principle of selection of variants with improved biodegradative properties was proposed. It embraces introduction of the dormancy stage into the cell life cycle with subsequent direct inoculation of morphologically different colonies into the media with different toxicants, including those previously not degraded by the strain.  相似文献

The metabolic fate of 14C-phenyl-labeled herbicide clodinafop-propargyl (CfP) was studied for 28 days in lab assays using a soil from Germany (Ap horizon, silt loam, and cambisol). Mineralization amounted to 12.40% of applied 14C after 28 days showing a distinct lag phase until day 7 of incubation. Portions of radioactivity extractable by means of 0.01 M CaCl2 solution (bioavailable fraction) decreased rapidly and were 4.41% after 28 days. Even immediately after application, only 57.31% were extracted with the aqueous solvent. Subsequent extraction using accelerated solvent extraction (ASE; acetonitrile/water 4:1, v/v) released 39.91% of applied 14C with day 0 and 26.16% with day 28 of incubation from the samples. Non-extractable portions of radioactivity thus, increased with time amounting to 11.99% (day 0) and 65.00% (day 28). A remarkable increase was observed between 14 and 28 days correlating with the distinct increase of mineralization. No correlation was found throughout incubation with general microbial activity as determined by DMSO reduction. Analysis of the CaCl2 and ASE extracts by radio-TLC, radio-HPLC and GC/MS revealed that CfP was rapidly cleaved to free acid clodinafop (Cf), which was further (bio-) transformed; DT50 values (based on radio-TLC detection of the parent compound) were far below 1 day (CfP) and about 7 days (Cf). TLC analysis pointed to 2-(4-hydroxyphenoxy)-propionic acid as further metabolite. Due to fractionation of non-extractable residues, most of the 14C was associated with fulvic and humic acids, portions in humin fractions and non-humics were moderate and low, respectively. Using a special strategy, which included pre-incubation of the soil with CfP and then mineralization of 14C-CfP as criterion, a microorganism was isolated from the soil examined. The microorganism grew using CfP as sole carbon source with concomitant evolution of 14CO2. The bacterium was characterized by growth on commonly used carbon sources and by 16S rDNA sequence analysis. The sequence exhibited high similarity with that of Rhodococcus wratislaviensis (99.56%; DSM 44107, NCIMB 13082).  相似文献

在考察了红平红球菌Rhodococcus erythropolis CCRC10900基本生长代谢特性的基础上,研究了营养条件对生物絮凝剂合成的影响。R.erythropolis CCRC10909合成的絮凝剂约80%以上分泌至胞外培养基中,仅20%左右的絮凝活性存在于菌体细胞上,蔗糖是该菌生长及絮凝剂合成的最佳碳源;玉米浆既能刺激菌体生长,又能显著提高絮凝剂的水平,生物絮凝剂的合成与培养基碳氮比密切相关,碳氮比在20-30之间时,絮凝活性达到最大,碳氮比低于10或高于100后,絮凝活性迅速下降,提出了促进絮凝剂合成的控制策略,发酵前期适当提高玉米浆的浓度,发酵后期按碳氮比为20-30,补加一定量的蔗糖和尿素,实验结果表明,采用这一策略,最终发酵液的絮凝活性和细胞生长量分别增长了8%和33.3%。  相似文献

研究了红球菌(Rhodococcus)Chr-9菌株在基础盐培养基中降解吡啶和苯酚的特性,分析了菌株降解苯酚和吡啶间的差异.结果表明,菌株Chr-9能够在72 h内将基础盐培养基中的吡啶(200 mg L-1)和苯酚(200 mg L-1)完全降解,同时利用吡啶和苯酚进行生长.菌株降解吡啶的最适温度为35℃,最适pH为8.0.菌株降解吡啶和苯酚的速度与底物的初始浓度呈负相关;在无其它氮源的基础盐培养基中,菌株能够利用吡啶和苯酚协同生长.图7参12  相似文献

联苯代谢对微生物的生长胁迫及分裂抑制   总被引:1,自引:0,他引:1  
以联苯/多氯联苯降解菌株R04(Rhodococcus sp.R04)和几种模式微生物为研究对象,利用高效液相色谱、荧光显微镜、扫描电子显微镜等分析微生物在联苯及其代谢物培养条件下细胞分裂和形态的变化.结果表明联苯及其代谢产物对红球菌R04和几种模式微生物细胞的分裂有抑制作用,并对部分微生物形态有影响.与前体-联苯及其代谢产物2-羟基-6-酮基-6-苯基-2,4-己二烯酸相比,2,3-二羟基联苯对G+、G-细菌,或是酵母细胞分裂都有较强的抑制和形态的改变.2,3-二羟基联苯导致R.R04和缺陷型R.R04细胞形成不完整隔膜的比例增加;造成96.4%的大肠杆菌BL21细胞表面凹陷,胞质内容物流失,菌体体积缩小;导致枯草芽孢杆菌89.6%的细胞体积明显缩小;导致金黄色葡萄球菌基本没有细胞能形成完整的分裂隔膜;导致红酵母细胞能进行出芽生殖的比例从64.2%降低到19.3%,但对其细胞形态无明显改变.联苯代谢物2,3-二羟基联苯对红球菌R04及其它微生物细胞分裂和增殖的抑制作用比其前体-联苯强,建议在研究环境化合物与微生物互作时,应考虑环境化合物代谢的毒性效应.  相似文献

从岳阳巴陵石化公司环己酮生产车间总出水口的污泥中筛选到一株环己酮降解菌株,菌号为JDM-3-12;该菌能以环己酮为唯一碳源且能忍受5000 mg/L的环己酮,当环己酮的质量浓度为2000 mg/L时,在温度为30℃,转速为150 r/m in,pH=7的条件下,72小时内该菌株对环己酮的降解率达到97.91%。通过形态观察、生理生化特征检测和基于16S rRNA基因序列的系统发育分析,初步鉴定其为赤红球菌(Rhodococcus ruber)中的一个菌株,该菌最适生长温度为35℃,最适生长pH 7。  相似文献
