为了研究运动对2,3,7,8-四氯二苯并二恶英(2,3,7,8-TCDD)急性暴露大鼠肝组织酶活性的影响,将40只雄性Wistar大鼠随机分为正常对照组(NC)、染毒组(NT)、运动对照组(EC)、运动染毒组(ET)。染毒组(NT组与ET组)腹腔注射10μg·kg-1(以单位体重计)的TCDD,对照组(NC组与EC组)腹腔注射等量的玉米油;NT、NC组静养4周,ET、EC组运动(尾部负重5%游泳30分钟)4周。4周后,称重并宰杀大鼠,分离肝组织,称重后-80℃保存待测7-乙氧基异吩恶唑酮脱乙基酶(EROD)、7-乙氧基香豆素-O-脱乙基酶(ECOD)及芳香烃羟化酶(AHH)的活性。将数据进行多因素方差分析(MAVONA)处理,结果表明,染毒可降低大鼠体重,增加肝湿重和肝相对重量、增加EROD、ECOD活性;运动可增加大鼠肝相对重量、增加AHH的活性;染毒后运动可降低EROD、ECOD的活性。结论:急性10μg·kg-1(以单位体重计)TCDD染毒后4周可增加大鼠肝相对重量;4周的运动能有效降低TCDD对EROD、ECOD活性的激活作用。  相似文献

Effects of artemether administration on liver and selected biochemical parameters were evaluated. Eighty albino mice were divided into four equal groups. Group 1 was given water which served as control, while groups 2, 3, and 4 were given 1.2, 2.4, or 4.8 mg kg?1 body weight artemether intramuscularly for five consecutive days. On day 6 all mice were sacrificed by cervical dislocation and blood was collected for analysis of alanine and aspartate transaminases, alkaline phosphatase, copper, and total proteins. Liver tissues were prepared for histological studies. It was found that the serum alanine and aspartate transaminase and alkaline phosphatase activities were higher in groups treated with artemether compared to control. The serum concentrations of copper and total proteins were lower than control. The histological features of liver tissues after administration of artemether showed histopathological alterations. These findings showed that artemether administration may have reversible adverse effects on mouse hepatocytes.  相似文献

火箭推进剂单推-Ⅲ对大鼠肝、肾的毒性效应   总被引:1,自引:1,他引:0  
为探讨新型火箭推进剂单推-Ⅲ(主成分为肼)对大鼠肝、肾的毒性效应及其作用机制,将24只雄性Wistar大鼠随机分为4组:生理盐水对照组,低剂量组、中剂量组和高剂量组,采用灌胃方式染毒,单次染毒剂量分别为0、0.70、2.35、7.05mg·kg-1(以单推-Ⅲ中的肼计算),每天1次,连续染毒7d,眼眶静脉丛取血后腹主动脉放血处死大鼠,称肝、肾重量计算肝、肾的脏器系数,测定大鼠血清中反映肝、肾功能的生化指标,并对肝、肾进行病理学观察.结果表明,1)单推-Ⅲ染毒可导致大鼠出现后肢运动障碍,并显著提高大鼠肝、肾脏器系数及血糖水平;2)病理学观察发现,单推-Ⅲ染毒可导致大鼠肝脏灶性及汇管区少量炎症细胞浸润,肝细胞脂肪变性,肾脏则无明显改变;3)单推-Ⅲ染毒可导致大鼠肝功能异常,部分肝功能指标如谷丙转氨酶(ALT)、谷草转氨酶(AST)、碱性磷酸酶(ALP)、直接胆红素(D-BIL)等出现显著升高或降低;4)单推-Ⅲ染毒可导致大鼠肾功能异常,部分肾功能指标如尿素氮(BUN)、尿酸(UA)、肌酐(CREA)等出现显著升高或降低.综合以上实验结果可以看出火箭推进剂单推-Ⅲ对大鼠肝、肾组织可产生一定的毒性效应,并可造成一定程度的中枢神经损伤。  相似文献

Increasing environmental pollution in the tropics is creating a breeding ground for mosquitoes, leading to increasingly frequent use of insecticides to combat home infestation of mosquitoes, the vector of malarial parasites. Household members are therefore more prone to aerosol exposure and subsequent health effects. We assessed the hepatotoxic and clastogenic effects in rats exposed to different levels of aerosols of one of the insecticides most commonly in used in Nigeria. There were significant (p?2 in the groups of rats exposed to the aerosols when compared with the control group. Gender differences in oral cocaine hepatotoxicity have been observed and lipopolysaccharide (LPS) was shown to potentiate cocaine hepatotoxicity in male CF-1 mice. Since cocaine and LPS inflict hepatocellular damage through the production of harmful reactive species, the role of cyclooxygenase-2 (COX-2) in gender-dependent cocaine hepatotoxicity was investigated. COX-2 increases nitric oxide (NO) synthesis and pro-inflammatory mediator production in response to hepatocellular insult. Male and female CF-1 mice were orally administered 20 mg kg?1 cocaine hydrochloride once daily for 7 days. Four hours after the last cocaine administration, the mice received 12 × 106 EU LPS intraperitoneally (cocaine + LPS). Mice receiving meloxicam (7.5 mg kg?1, p.o.), a selective COX-2 inhibitor, were pretreated 1 h before each cocaine administration. Meloxicam prevented liver damage in males produced by cocaine and cocaine + LPS as evidenced by microscopic appearance and biochemical indices equivalent to saline treatment. Acetaminophen (APAP) was administered orally as a positive control which shows no gender dependency and elicited a hepatotoxic response in both males and females. Altered biochemical indices suggest that COX-2 is activated, possibly to a greater extent in females versus males in APAP-induced hepatotoxicity. Meloxicam prevented all oxidative stress changes induced in males following APAP. The results suggest that the induction of COX-2 is responsible, in part, for cocaine hepatotoxicity with and without LPS exposure in male mice.  相似文献

Trichlorfon is an organophosphate insecticide that is widely used in aquaculture and agriculture against parasitic infestations and has caused aquatic toxicity to non-target organisms. To evaluate the effects of low doses of trichlorfon on the oxidative stress and hepatotoxicity in amphibians, Chinese brown frogs (Rana chensinensis) were exposed to trichlorfon at concentrations of 0, 0.01, 0.1, and 1.0 mg/L for 2 and 4 weeks. Then, the activities of superoxide dismutase (SOD), catalase (CAT), glutathione S-transferase (GST), and the content of malondialdehyde (MDA) in hepatic tissue were examined to evaluate the effects of oxidative stress and lipid peroxidation. The histopathological alternations to the liver were observed through light and transmission electron microscopy (TEM). The results showed that SOD and CAT activities were increased in the livers of frogs exposed to various concentrations of trichlorfon. The GST activity showed no significant changes at any concentration after 2 weeks of exposure, whereas there was an initial increase after exposure to 0.1 mg/L of trichlorfon at 4 weeks. The content of MDA revealed a significant decrease after exposure. Histopathological and ultrastructural studies showed that trichlorfon induced hyalinization, vacuolation, nucleus necrosis, and cellular swelling in hepatocytes. These results suggest that low doses of trichlorfon could induce oxidative stress, lipid peroxidation, and hepatic lesions in frogs, which shows that even lower, non-lethal doses of trichlorfon are potentially toxic to amphibians.  相似文献

The main purpose of this study was to ascertain whether occupational exposure to asphalt fumes, under normal working conditions, is associated with any hepatotoxic, nephrotoxic, or hematotoxic response. This was a cross-sectional study in which 210 subjects (80 exposed and 130 reference subjects) were investigated. Using standard methods, atmospheric concentrations of total particulate and benzene-soluble fractions of asphalt fumes were measured. Additionally, urine and blood samples were taken from subjects for complete blood count, white blood cell differential test, urinalysis, and routine biochemical tests of kidney and liver function. Both groups were similar as far as their demographic variables and smoking habits were concerned. The levels of exposure to total particulate and benzene-soluble fractions were estimated to be 0.9?±?0.2 and 0.32?±?0.09 mg m?3, respectively (mean?±?SD), both of which being lower than the existing threshold limit values. Statistically, significant associations between exposure to asphalt fumes and changes in most liver and kidney function tests and complete blood count parameters provided circumstantial evidence in favor of the notion that exposure to sub-threshold limit value levels of total particulate and benzene-soluble fractions is associated with early liver and kidney dysfunction as well as hematological disorders.  相似文献

Acrylamide (ACR) exerts its toxicity through stimulation of the oxidative stress; yet, its effect on neurotransmitter catabolic enzymes has not been elucidated. We investigated the effects of ACR exposure on brain and hepatic tissues antioxidant enzymes activities and different markers such as, acetylcholinesterase (AChE), nitric oxide (NO), monoamine oxidase (MAO), and lipid profile, and to evaluate the protective effects of garlic against ACR toxicity. Male Sprague-Dawley rats were exposed to ACR (1 mg kg?1 body weight) with or without diet containing 1.5% of garlic powder for 40 days. ACR administration showed a decrease in AChE activity associated with an increase in MAO activity in both brain and hepatic tissues. In addition, ACR administration increased the lipid peroxidation and NO levels of both tissues while decreased the activities of glutathione (GSH), superoxide dismutase, and glutathione-S-transferase (GST). On the other hand, the activities of glutathione peroxidase (GPx) and catalase activities increased as a consequence of GSH depletion after ACR exposure. Finally, ACR exposure increased the brain and liver lipid profile of cholesterol, triglycerides and total lipid, while phospholipids level was decreased. Coadministration of garlic powder with ACR significantly attenuated oxidative stress, MAO activity, and inflammation in brain and hepatic tissues but did not ameliorate AChE activity. In conclusion, our results emphasized the role of garlic as a potential adjuvant therapy to prevent ACR neurotoxicity and hepatotoxicity.  相似文献

Medicinal herbs are steadily increasing in complementary use for chronic and alterative therapies. The health risks associated with herbal supplements have therefore been identified as a top research priority. Tecoma stans (Tronadora), a Bignoniaceae plant, is a herbal drug traditionally consumed as tea in South America for the control of diabetes. It contains the alkaloids of tecomine and tecostanine which were shown as functional compounds responsible for hypoglycemic activity. However, the side effects of aqueous extracts of this herbal tea have not been reported. In this study, studies sought to evaluate the cytotoxicity of water extracts from T. stans in human hepatoblastoma (HepG2). Toxic effects of T. stans were concentration- and time-dependent in the presence and absence of fetal bovine serum (FBS). Cells were incubated for up to 72-h with varying concentrations of herbal extracts (60–100%), cytotoxicity was determined spectrophotometrically by MTT and reported in terms of % cell viability. For IC50 assay (24 h exposure), cytotoxicity was found at concentration of 60–100%. Evaluation of the effects of T. stans and Brickellia cavanillesi (Asteraceae) in combination revealed that extracts of both herbal extracts' nontoxic levels of T. stans significantly reduced cell viability in a time-dependent manner. Further studies are needed regarding the identification, toxicity, and molecular mechanism of action of active compounds, as well as their cytotoxicity to other cell lines following exposure to both single and combined herbal extracts of these plants with anti-diabetic properties.  相似文献

研究离子液体溴化1-十四烷基-3-甲基咪唑盐([C14mim]Br)对小鼠的肝脏毒性及其作用机制。实验设立3个剂量的染毒组(1/16 LD50、1/8 LD50、1/4 LD50)和1个空白对照组,考察昆明种小鼠染毒14 d后,[C14mim]Br对小鼠血清生化指标、肝脏抗氧化酶活性及脂质过氧化产物的影响,并观察肝脏组织的病理变化。与对照组相比,小鼠染毒后,血清中ALT、AST、DBIL和γ-GT明显升高;肝组织受到不同程度的损伤,HSI增大,蛋白质含量降低。当染毒剂量为1/4 LD50时,肝脏中SOD活性和GSH-Px活性显著降低,MDA的含量则明显增加。实验结果表明,[C14mim]Br可损伤小鼠的肝脏功能,破坏机体的抗氧化防御系统,造成氧化损伤和脂质过氧化。  相似文献
