Herlitz junctional epidermolysis bullosa (HJEB) is a severe blistering disorder which usually results in death during infancy. We have previously shown that the anchoring filament protein laminin-5 (kalinin/nicein), which mediates keratinocyte attachment and dermal–epidermal cohesion, is abnormally expressed in individuals with HJEB. Laminin-5 was detected by Western blot analysis in amniotic fluid from 44 consecutive normal secondtrimester control pregnancies, but was undetectable in second-trimester amniotic fluid from four pregnancies with fetuses affected by HJEB. In one case of severe non-Herlitz JEB, laminin-5 was detected in both amniotic fluid and skin. In human amniotic fluid, the laminin-5 a3 subunit was processed to a major 165 kD species and a minor 145 kD species and the β2 subunit was partially processed to 105 kD. Although laminin-5 was covalently associated with laminin-6 (K-laminin) in amniotic membrane, no covalent interaction was detected in amniotic fluid. Laminin-5 from amniotic fluid strongly supported keratinocyte attachment. These results suggest that Western blot analysis of second-trimester amniotic fluid is useful in determining the prenatal diagnosis of HJEB and that laminin-5 may serve a physiologically important function in amniotic fluid. 相似文献

Recent landscape changes in a farmed landscape are analysed and related to farm and farmer characteristics. It is assumed that farm and farmer characteristics serve as mediators of large scale or macro driving forces of change-in the present case, a changing farming context including demands for a more environmentally friendly farming practise and a reduced output. The results are based on multivariate analyses of data collected from structured interviews of 160 farmers in a case study area, in central Jutland, measuring 5000 ha. The analysis shows that farmers are highly involved in landscape changes. The investigated landscape changes include creation and removal of landscape elements as well as certain management changes. The most common activity was creation of elements: hedgerows, small woodlands and conversion of rotational arable land to permanent grassland, whereas removal of elements, mainly hedgerows and semi-natural grasslands, were seen less frequently. Management changes like abandonment of permanent grassland were widespread. The results indicate a general extensification of the land use and the authors interpret the results partly as an indication of a change from productivism to a more multifunctional agricultural regime. The observed landscape changes at the farm level show a low, but structured relationship with the current farm and farmer characteristics, meaning that landscape changes were undertaken by various farmers and on various farms. On a general level, however, the age of the farmer and the duration of farm ownership seem to have a major influence on the landscape changes. 相似文献

Most flax (Linum usitatissimum) varieties are described as tolerant to high concentrations of Cd. The aim of the present paper was to better characterize this tolerance, by studying the responses of flax plantlets, cv Hermes, to 18d growth on 0.5mM Cd. In Cd-treated seedlings, the majority of Cd was compartmentalized in the roots. Analysis of other elements showed that only Fe concentration was reduced, while Mn increased. Growth parameters of Cd treated flax were only moderately altered, with similar mass tolerance-indices for roots and shoots. Tissue anatomy was unaffected by treatment. The effect on lipid peroxidation, protein carbonylation and antioxidative activities appeared low but slightly higher in roots. The most important impacts of Cd were, in all organs, cell expansion, cell-wall thickening, pectin cross-linking and increase of cell-wall enzymatic activities (pectin methylesterase and peroxidase). Thus, the role of the cell wall in Cd tolerance might be important at two levels: (i) in the reinforcement of the tissue cohesion and (ii) in the sequestration of Cd. 相似文献

Summary. Several species of Longitarsus take up, metabolize and
store pyrrolizidine alkaloids (PAs) from their host plants. In feeding experiments using
radioactively labeled PAs of different types we examined the time course of the sequestration
process in L. jacobaeae and
L. aeruginosus. We found that adapted species efficiently
store PAs for at least two weeks without major losses. During that time, there is virtually
no change in the ratio of tertiary alkaloids to stored non-toxic
N-oxides, regardless of chemical form fed to the beetles. This
implies a transient N-oxidation process where the alkaloids are
only temporarily accessible to the enzyme. A dissection experiment with
L. aeruginosus six days after uptake of labeled PAs
showed that the tertiary alkaloids are not found in the hemolymph but are stored in the
elytra and other body compartments. This conforms with earlier experiments that localized the
enzymes site of action in the hemolymph. Furthermore we show that different total alkaloid
doses in the diet of L. jacobaeae and the potentially less
adapted L. succineus do not affect the ratio of recovered
N-oxides to tertiary molecules. Thus, the efficiency of the
N-oxidizing enzyme is not dependent on the concentration of

Summary. Sequestration and processing of pyrrolizidine alkaloids
(PAs) by leaf beetles of the genus Platyphora were investigated. Tracer
experiments with labeled alkaloids were performed with P. eucosma
feeding on Koanophyllon panamense (Asteraceae, tribe Eupatorieae). P.
eucosma catalyzes the same reactions previously demonstrated for P.
boucardi specialized to Prestonia portobellensis (Apocynaceae): (i)
epimerization of rinderine to intermedine; (ii) esterification of
retronecine yielding insect-specific PAs; (iii) efficient transport of
the PAs as free bases into the defensive secretions. P. bella feeding on
Tournefortia cuspidata (Boraginaceae) shows the same sequestration
behavior and ability to synthesize the specific retronecine esters. P.
ligata, a species phylogenetically closely related to the PA adapted
species and clustering in the same clade, but feeding on a host plant
devoid of PAs, feeds easily on PA treated host-plant leaves, but does
not sequester or metabolize PAs. P. kollari a species clustering outside
the PA clade refused to feed on its food-plant leaves painted with PAs.
The results are discussed in relation to host-plant selection of the PA
adapted species and the role of PAs in chemical defense.

Summary. Field collected exocrine defensive secretions of nine neotropical Platyphora species were analyzed for the presence of plant acquired pyrrolizidine alkaloids (PAs) and pentacyclic triterpene saponins.
All species secrete saponins. In addition, five species feeding on Tournefortia (Boraginaceae), Koanophyllon (Asteraceae, tribe Eupatorieae) and Prestonia (Apocynaceae) were shown to sequester PAs of the lycopsamine type, which are characteristic for species of the three plant
families. The PA sequestering species commonly store intermedine, lycopsamine and their O3′-acetyl or propionyl esters as well as O7- and O9-hydroxyisovaleryl esters of retronecine. The latter as well as the O3′-acyl esters were not found in the beetles’ host plants, suggesting the ability of the beetles to esterify plant derived retronecine
and intermedine or its stereoisomers. Despite the conformity of the beetles’ PA patterns, considerable inconsistencies exist
regarding the PA patterns of the respective host plants. One host plant was devoid of PAs, while another contained only simple
necines. Since the previous history of the field collected beetles was unknown this discrepancy remains obscure. In contrast
to the Palearctic chrysomeline leaf beetles, e.g. some Oreina species which ingest and store PAs as their non-toxic N-oxides, Platyphora leaf beetles absorb and store PAs as the toxic free base (tertiary PA), but apparently avoid to accumulate PAs in the haemolymph.
This suggests that Chrysolina and Platyphora leaf beetles developed different lines of adaptations in their parallel evolution of PA mediated chemical defense.

Both evolutionary ecologists and wildlife managers make inference based on how fitness and demography vary in space. Spatial variation in survival can be difficult to assess in the wild because (1) multisite study designs are not well suited to populations that are continuously distributed across a large area and (2) available statistical models accounting for detectability less than 1.0 do not easily cope with geographical coordinates. Here we use penalized splines within a Bayesian state-space modeling framework to estimate and visualize survival probability in two dimensions. The approach is flexible in that no parametric form for the relationship between survival and coordinates need be specified a priori. To illustrate our method, we study a game species, the Eurasian Woodcock Scolopax rusticola, based on band recovery data (5000 individuals) collected over a > 50 000-km2 area in west-central France with contrasted habitats and hunting pressures. We find that spatial variation in survival probability matches an index of hunting pressure and creates a mosaic of population sources and sinks. Such analyses could provide guidance concerning the spatial management of hunting intensity or could be used to identify pathways of spatial variation in fitness, for example, to study adaptation to changing landscape and climate. 相似文献
