This study re-evaluated the colorimetric assay for cytidine deaminase (CTD), and showed that the optimum conditions were pH 7·5, 37°C, and up to 24 h. In addition, this method was found to require protein precipitation. Following these modifications, intra-assay and inter-assay coefficients of variation were below 5 per cent, indicating that the assay was highly reliable. CTD activity was determined in 282 serum samples from 206 normal pregnant women by the incubation of 100 μl of serum and 400 μl of 1·4 mmol/l cytidine substrate for 16 h at 37°C. Following protein precipitation, the ammonia liberated during conversion was measured by a colorimetric procedure. The mean (±SD) CTD activity was 7·31 ± 2·50 U at 3–12 weeks of gestation, 8·70 ± 2·12 U at 13–24 weeks, 7·59 ± 2·25 U at 25–36 weeks, and 7·29 ± 2·16 U at 37–42 weeks. High levels of CTD activity were found in patients with abruptio placentae and amnionitis associated with intrauterine fetal death (IUFD). The increase in CTD activity was noted from 3 days to 1 week before the confirmation of IUFD. The placenta contains extremely high levels of CTD, but cord serum does not. Thus, the excessive elevation of CTD activity was probably derived from progressive placental damage. This modified CTD assay was concluded to be simple and reliable, and may perhaps be useful in detecting pregnancy disorders.  相似文献

Lake Baikal is the deepest and oldest freshwater lake in the world containing about 20% of the world's fresh water. The present state of its contamination with organic pollutants was studied during 1997–1998 by collecting samples of sediments, phytoplankton, Zooplankton, sponges, aquatic plants, algae, muscles of various fish species, eggs of various birds and blubber of seals. Samples were prepared and analysed using GC‐MS for semivolatile priority pollutants on the US EPA list, for the presence of organochlorine pesticides and for PCBs. The results showed that there was pronounced bioaccumulation of persistent organochlorine compounds along the trophic chain, leading to substantial residues of organochlorine compounds in seals (e.g. 0.87–25.3 ppm of p,p'‐DDT in blubber).  相似文献

Chain-extension reactions were carried out using titanium-iso-propoxide (TIP) as a catalyst for a series of polyesters or copolyesterethers with low molecular weights (M n =1500–10,000) synthesized by the ring-opening copolymerization of succinic anhydride (SA) with ethylene oxide (EO). The copolymers having aM n from 25,000 to 50,000 of different properties were obtained. Both the melting point (T m ) and the fusion heat (H), which indicate the crystallinity of the copolymers, rose with an increase in SA content in the copolymers. Semitransparent films were prepared by compression molding of the copolymers. The biodegradation of the copolymer films was evaluated by enzymatic hydrolysis by lipases and by an aerobic gas evolution test in standard activated sludge. The hydrolyzability of these copolymers by three kinds of lipases was affected by their copolymer composition SA/EO, form, andM n . The copolyesterether (SA/EO=43/57,M n =48,900) was more easily biodegraded by standard activated sludge compared to the polyester (SA/EO=47/53,M n =36,300).Presented at the Pacifichem-95, December 17–22, 1995, Honolulu, Hawaii.  相似文献

Two fractions containing the oxidase activity toward 2,2'-azinobis (3-ethylbenzothiazoline-6-sulfonate) (ABTS) were obtained using ion-exchange DEAE-Sepharose column chromatography of the culture fluid of white-rot fungus, Trametes versicolor. These two fractions can reduce the level of coplanar PCB congeners (Co-PCBs). The ABTS oxidase in the first fraction passed through the DEAE-Sepharose column. An international exercise to directly assess consistency of standards for ground-level ozone in East Asia was conducted as part of the East Asian Regional Experiment 2005 (EAREX 2005) in the framework of the Atmospheric Brown Clouds (ABC) project. Ten organizations collaboratively participated in the intercomparison. Four groups representing Japan, Korea, Hong Kong, and Taiwan made comparisons at the Gosan super observatory, Jeju Island, Korea, in March 2005, with ozone instruments calibrated to their national standards, and four Japanese groups made off-site comparisons with laboratory-level standards. All comparisons generally indicated good agreement with the standard reference photometer (SRP) 35, built by the National Institute of Standards and Technology (USA) and maintained by the National Institute for Environmental Studies (Japan). The assessment was expanded to measurement networks contributing to the World Meteorological Organization's Global Atmospheric Watch (WMO/GAW) program as part of off-site comparisons, and excellent agreement was achieved. These efforts contribute to propagating traceability of the national metrology standards among the atmospheric science community, to ensuring comparability of the existing ozone measurements, and to establishing an integrated network of air quality monitoring in Asia.  相似文献

This study aimed at clarifying the impact of deforestation and afforestation on the quality of life in a village in Sichuan Province, China. We devised a conceptual model of bioresource production and use based on quantified energy flow. The basic structure of the model has three sectors: production, use, and externals. We developed comprehensive methodology to quantify the model. Bioresource use per person in 1997 was 3.7 GJ for food, 10.2 GJ for fodder, 0.2–0.4 GJ for building material, 12.8 GJ for fuel, and 1.8 GJ for fertilizer, totaling 28.6–28.8 GJ.We used four environmental indicators to evaluate bioresource production and use: a biological productivity indicator, a use-efficiency indicator, a supply–demand balance indicator, and a self-sufficiency indicator. Use of these indicators showed that supply-demand balance of fuel was dramatically improved from 30% to 85% by afforestation, but 99% of bioresource use still depends on domestic products. Thus, it is necessary to improve biological productivity and promote the efficient use of bioresources to achieve sustainable living in the area. Massive deforestation in the 1950s caused a direct shortage of building material and fuel wood. The shortage of wood led to a stagnation in the rebuilding of houses, and fuel wood was substituted with crop residues. Because crop residues had been used for fertilizer and fodder, their use as fuel caused a shortage of fertilizer and fodder. This was an indirect impact of deforestation on peoples quality of life.  相似文献
