Pseudomonas (PI2) capable of degrading pyridine was isolated from the mixed population of the activated sludge unit which was being used for treating complex effluents, the strain was characterized. Aerobic degradation of pyridine was studied with the isolated strain and the growth parameters were evaluated. Pyridine degradation was further conformed by chromatography (HPLC) analysis. The process parameters like biomass growth and dissolved oxygen consumption were monitored during pyridine degradation. In order to conform with the plasmid capability to degrade pyridine, the requisite plasmid was isolated and transferred to DH 5alpha Escherichia coli. The subsequent biodegradation studies revealed the ability of the transformed plasmid capability to degrade the pyridine.  相似文献

The optimal design of reserve networks and fisheries closures depends on species occurrence information and knowledge of how anthropogenic impacts interact with the species concerned. However, challenges in surveying mobile and cryptic species over adequate spatial and temporal scales can mask the importance of particular habitats, leading to uncertainty about which areas to protect to optimize conservation efforts. We investigated how telemetry-derived locations can help guide the scale and timing of fisheries closures with the aim of reducing threatened species bycatch. Forty juvenile speartooth sharks (Glyphis glyphis) were monitored over 22 months with implanted acoustic transmitters and an array of hydrophone receivers. Using the decision-support tool Marxan, we formulated a permanent fisheries closure that prioritized areas used more frequently by tagged sharks and considered areas perceived as having high value to fisheries. To explore how the size of the permanent closure compared with an alternative set of time-area closures (i.e., where different areas were closed to fishing at different times of year), we used a cluster analysis to group months that had similar arrangements of selected planning units (informed by shark movements during that month) into 2 time-area closures. Sharks were consistent in their timing and direction of migratory movements, but the number of tagged sharks made a big difference in the placement of the permanent closure; 30 individuals were needed to capture behavioral heterogeneity. The dry-season (May–January) and wet-season (February–April) time-area closures opened 20% and 25% more planning units to fishing, respectively, compared with the permanent closure with boundaries fixed in space and time. Our results show that telemetry has the potential to inform and improve spatial management of mobile species and that the temporal component of tracking data can be incorporated into prioritizations to reduce possible impacts of spatial closures on established fisheries.  相似文献

Phosphogypsum (PG) is produced as a solid waste from phosphatic fertilizer plants. The waste slurry is disposed off in settling ponds or in heaps. This solid waste is now increasingly being used as a calcium supplement in agriculture. This study reports the effectof PG amendmenton soil physico chemical properties, bacterial and fungal count and activities of soil enzymes such as invertase, cellulase and amylase over an incubation period of 28 days. The highest mean percent carbon loss (55.98%) was recorded in 15% PG amended soil followed by (55.28%) in 10% PG amended soil and the minimum (1.68%) in control soil. The highest number of bacterial colonies (47.4 CFU g(-1) soil), fungal count (17.8 CFU g(-1) soil), highest amylase activity (38.4 microg g(-1) soil hr(-1)) and cellulase activity (38.37 microg g(-1) soil hr(-1)) were recorded in 10% amended soil. Statistically significant difference (p<0.05) has been recorded in the activities of amylase and cellulase over the period of incubation irrespective of amendments. Considering the bacterial and fungal growth and the activities of the three soil enzymes in the control and amended sets, it appears that 10% PG amendment is optimal for microbial growth and soil enzyme activities.  相似文献

Miscibility characteristics of Xanthan gum and Poly (vinyl pyrrolidone) (PVP) in common solvent water were studied by viscometry, ultrasonic velocity and density techniques at 30 and 50 °C. Blend films of Xanthan gum/PVP were prepared by solution casting method and characterized by scanning electron microscopic (SEM) and differential scanning calorimeter (DSC) techniques. Using the viscosity data, interaction parameters of Chee’s (μ) and Sun’s (α) were computed to determine their miscibility. The values obtained revealed that blends were miscible when PVP content is up to 70% in blend at 30 °C. Xanthan gum/poly (vinyl pyrrolidone) blends showed miscibility in all composition at 50 °C. The results were then confirmed by ultrasonic velocity, density, and DSC techniques. Compatibility in the above compositions may be due to the formation of hydrogen bonding between the carbonyl group in PVP and hydroxyl group in Xanthan gum. Further, the results revealed that change in temperature had significant effect on the miscibility of Xanthan gum/Poly (vinyl pyrrolidone) blends.  相似文献

Miscibility studies of Sodium alginate (NaAlg)/Pullulan have been carried out in different percentage of blend components. The ultrasonic velocity, viscosity, density and refractive index were measured at 30 and 40 °C. Further the blend films of NaAlg and Pullulan were prepared by solution casting method and characterized by differential scanning calorimetric (DSC), fourier transition infrared spectroscopic (FTIR), and scanning electron microscopic (SEM) methods. Using the viscosity data, interaction parameters were computed to determine the miscibility. The data suggest that the blend is miscible in the entire composition range. The change in temperature had no significant effect on the miscibility of NaAlg/Pullulan blends. The miscibility is confirmed by SEM, DSC, ultrasonic velocity, density, and refractive index methods. The specific interactions of hydrogen bonding type of the blends were investigated by FTIR.  相似文献
