Organic solvents, such as dimethylsulfoxide (DMSO) and methanol are widely used as vehicles to solubilise lipophilic test compounds in toxicity testing. However, the effects of such solvents upon innate detoxification processes in aquatic organisms are poorly understood. This study assessed the effect of solvent exposure upon cytochrome P450 (CYP)-mediated xenobiotic metabolism in Daphnia magna and zebrafish larvae (4 d post fertilisation). Adult D. magna were demonstrated to have a low, but detectable, metabolism of ethoxyresorufin in vivo and this activity was not modulated by pre-exposure to DMSO or methanol (24 h, up to 0.1% and 0.05% v/v, respectively). In contrast, the metabolism of ethoxyresorufin in zebrafish larvae was significantly reduced by both solvents (0.1% and 0.05% v/v, respectively) after 24 h of exposure. In zebrafish, these observed decreases in activity towards ethoxyresorufin were accompanied by decreased expression of a variety of genes coding for drug metabolising enzymes (corresponding to CYP1, CYP2, CYP3 and UDP-glucuronyl transferase [UGT] family enzymes), measured by quantitative PCR. Reduction of gene expression and CYP1 enzyme activities by methanol (0.05% v/v) in zebrafish larvae was partially reversed by co-exposure with Aroclor 1254 (100 μg L−1). Overall this study suggests that relatively low concentrations of organic solvents can impact upon the biotransformation of certain xenobiotics in zebrafish larvae, and that this warrants consideration when assessing compounds for metabolism and toxicity in this species.  相似文献

Introduction

The purpose of the current study was to examine differences in factors associated with self-reported collision involvement of three age groups of drivers based on a large representative sample of Ontario adults. Method: This study was based on data from the CAMH Monitor, an ongoing cross-sectional telephone survey of Ontario adults 18 years and older from 2002 to 2005. Three age groups were examined: 18-34 (n = 1,294), 35-54 (n = 2,428), and 55+ (n = 1,576). For each age group sample, a logistic regression analysis was conducted of self-reported collision involvement in the last 12 months by risk factor measures of driving exposure (kilometers driven in a typical week, driving is stressful, and driving on busy roads), consuming five or more drinks of alcohol on one occasion (past 12 months), cannabis use (lifetime, and past 12 months), and driving after drinking among drinkers (past 12 months), controlling for demographics (gender, region, income, and marital status). Results: The study identified differences in factors associated with self-reported collision involvement of the three age groups of adult drivers. The logistic regression model for the youngest group revealed that drivers who reported that driving was stressful at least some of the time, drank five or more drinks on an occasion, and drove after drinking had an increased risk of collision involvement. For the middle age group, those who reported using cannabis in the last 12 months had significantly increased odds of reporting collision involvement. None of the risk factor measures showed significant associations with collision risk for older drivers (aged 55+). Impact: The results suggest potential areas for intervention and new directions for future research.  相似文献

Drowsiness has been recognized as a pervasive problem for drivers, with effects comparable to alcohol. Alcohol, however, has a clear legal limit for impairment; there are no comparable criteria to suggest sleepiness. Drowsiness has been associated with light and circadian rhythm. To investigate the joint effects of these factors on crash occurrence, along with other factors, single vehicle crashes reported in Ontario (1999–2004) were analyzed. Crashes occurring at four times of day, when light varies and circadian rhythm is low (2–5 a.m. and 2–4 p.m.) and with similar light conditions and higher circadian rhythm (9–11 p.m. and 10 a.m.–12 noon). Logistic regression was used to see how light and other factors are associated with single vehicle crashes occurring at times of low circadian rhythm, when drowsiness is more likely.Initial results indicated many circumstances associated with occurrence at these times: the age and sex of the driver and reported driver condition as well as weather. There is, however, an interaction between light and presumed alertness. In separate analyses for daytime and night time crashes most variables were significant for nighttime crashes but not for daytime events. The effects of alcohol and youth remained. A lack of light may exacerbate the effects of other factors at times of low alertness; this should be further investigated in controlled environments such as sleep laboratories and/or driving simulators.  相似文献

Carbon baking process involves evolution of fumes containing hydrocarbons and soot particles which cannot be discharged directly into the atmosphere. An incinerator can be used to clean these fumes. However, length of the baking cycle, nature of the fumes and variations in fume volume and temperature may result in excessive auxiliary fuel usage and inefficient incineration, if the incinerator is not designed properly. This paper describes the application of fundamental knowledge of aerodynamics, reaction kinetics and combustion, together with clear understanding of the process, in design of a highly efficient, fully automated incinerator. The design incorporates a unique but simple control system which results in reduction of auxiliary fuel usage without endangering the safety and efficiency of the incineration process. Operations and economics of the incinerator are described by illustrating a typical baking cycle and comparing actual fuel usage with the thermal ratings of the incinerator. Operating experience from a number of installations in the U. S. and Canada is also noted.  相似文献

Many traffic crashes are side impact collisions resulting in significant death and injury. A review was conducted of the evidence of driver, road, and vehicle characteristics affecting either the risk of occurrence or the severity of injury in such crashes for papers published from 1996 to early 2003. For drivers, evidence was found of increased crash risk or injury severity only for age and age-related medical conditions (e.g., dementia). Traffic roundabouts and other traffic control devices--stop signs, traffic lights, and so on--had mixed results; traffic controls were better than no controls, but their effectiveness varied with circumstance. Most vehicle characteristics have had little or no effect on crash occurrence. Antilock braking systems (ABS) in the striking vehicle had been anticipated to reduce the risk of crashes, but so far have demonstrated little effect. The primary emphasis in vehicle design has been on protective devices to reduce the severity of injury. Disparity in the size of the two vehicles, especially when the struck vehicle is smaller and lighter, is almost a consistent risk factor for occupant injury. The occupants of light trucks, however, when struck by passengers cars on the opposite side, were at higher risk of injury. Wearing seat belts had a consistently protective effect; airbags did not, but there were few studies, and no field studies, of lateral airbags found. Of all the characteristics examined, vehicle design, including occupant restraints, is the most easily modified in the short term, although road design, traffic control, and the monitoring of older drivers may also prove effective in reducing side impact crashes in the longer term.  相似文献

Mussels were collected from the urban/industrialized site of New Brighton, Merseyside and the relatively non-industrial site of Llandudno, North Wales. All mussels were identified as Mytilus edulis by PCR amplification of Mefp1. DNA single strand breaks and 8-oxo-7,8-dihydro-2'-deoxyguanosine were measured in gill within 24h of collection, using the COMET assay, both with and without formamidopyrimidine glycosylase. Gill lipid peroxidation was also measured within 24h. No difference between sites was found for frank SSB and malonaldehyde levels, however 8-oxo-dG and 4-hydroxynonenal were significantly greater in New Brighton mussels compared to Llandudno mussels. After 1-month laboratory maintenance, lipid peroxidation and 8-oxo-dG levels were lower. In contrast, frank SSB were higher. This could reflect enhanced DNA repair excision, though we cannot exclude the possibility of other non-oxidative DNA damage. The results suggest that laboratory maintenance allows recovery from environmentally induced oxidative damage, which was more extensive at Merseyside.  相似文献
