The potential for xenobiotic compounds to bioconcentrate is typically expressed through the bioconcentration factor (BCF), which has gained increased regulatory significance over the past decade. Due to the expense of in vivo bioconcentration studies and the growing regulatory need to assess bioconcentration potential, BCF is often calculated via single-compartment models, using K(OW) as the primary input. Recent efforts to refine BCF models have focused on physiological factors, including the ability of the organism to eliminate the compound through metabolic transformation. This study looks at the ability of in vitro biotransformation assays using S9 fractions to provide an indication of metabolic potential. Given the importance of the fish gill and liver in metabolic transformation, the metabolic loss of ibuprofen, norethindrone and propranolol was measured using rainbow trout (Oncorhynchus mykiss) and channel catfish (Ictalurus punctatus) gill and liver S9 fractions. Metabolic transformation rates (k(M)) were calculated and integrated into a refined BCF model. A significant difference was noted between BCF solely based on K(OW) and BCF including k(M). These studies indicate that the inclusion of k(M) in BCF models can bring predicted bioconcentration estimates closer to in vivo values.  相似文献

The effects of residing in a polycyclic aromatic hydrocarbon (PAH) contaminated environment on the cytometric characteristics of hemocytes from the American oyster Crassostrea virginica (collected from the Rappahannock River, Virginia, USA in Spring 1991) were analyzed using a multichannel Coulter counter (10000 hemocytes oyster-1). The percentage and relative volume of small-sized hemocytes (>2.5 to 5.1 m) was higher (P<0.01) in oysters from Hospital Point (HP, a PAH contaminated site) in the Elizabeth River (ER), Virginia, compared to oysters from the relatively unpolluted Rappahannock River (RR). On the contrary, a decrease (P<0.001) in the percentage and volume of larger hemocytes (>6.2 to 10 m) was observed in the HP-oysters. Maintaining the RR-oysters at the HP site for 8 wk induced statistically significant increases in the number and relative volume contribution of the >2.5 to 5.1 m hemocytes and sharp decreases in the occurrence and relative volume of >6.2 to 13 m cells (P<0.01). By depurating the HP-oysters in the York River (the control site), the number of, and volume contributed by the small hemocytes (>2.5 to 5.1 m) showed a significant decrease over time as compared to the baseline values. On the other hand, the relative number of >6.2 to 13 m hemocytes increased sharply within 8 wk (P<0.001). This indicates that these changes are both inducible and reversible and, at least in part, due to exposure to the ER-sediment. Based on these studies, oyster hemocyte cytometric characteristics could be developed as a sensitive biomarker of exposure to PAH.  相似文献

Aqueous 7-d germination and growth experiments were performed to compare responses of T. latifolia to exposures of atrazine (2-chloro-4-ethylamino-6-isopropylamine-s-atrazine) and paraquat dichloride (1,1′-dimethyl-4,4′-bipyridinium dichloride). T. latifolia seed germination was < 50 % in concentrations ≥ 1.0 mg/L of paraquat dichloride. No observed effect concentration (NOEC) and lowest observed effect concentration (LOEC) for paraquat and root growth were 0.001 and 0.01 mg/L, respectively, while NOEC and LOEC for paraquat and shoot growth were 0.01 and 0.1 mg/L, respectively following 7-d exposures. Greater than 72 % of seeds germinated in each concentration up to 30 mg/L atrazine. After 7-d exposure, NOEC and LOEC for atrazine and root growth were 0.1 and 1.0 mg/L, while atrazine and shoot growth NOEC and LOEC values were 15 and 30 mg/L, respectively. This research provides data concerning relative sensitivity of T. latifolia seedlings to the herbicides atrazine and paraquat, as well as the potential use of T. latifolia as a representative plant test species.  相似文献

Beta adrenergic receptor antagonists (beta-Blockers) are frequently prescribed medications in the United States and have been identified in European municipal wastewater effluent, however no studies to date have investigated these compounds in United States wastewater effluent. Municipal wastewater effluent was collected from treatment facilities in Mississippi, Texas, and New York to investigate the occurrence of metoprolol, nadolol, and propranolol. Propranolol was identified in all wastewater samples analyzed (n = 34) at concentrations < or = 1.9 microg/l. Metoprolol and nadolol were identified in > or = 71% of the samples with concentrations of metoprolol < or = 1.2 microg/l and nadolol < or = 0.36 microg/l. Time course studies at both Mississippi plants and the Texas plant indicate that concentrations of propranolol, metoprolol, and nadolol remain relatively constant at each sampling period. This study indicates that beta-Blockers are present in United States wastewater effluent in the ng/l to microg/l range.  相似文献

Pharmaceutical products and their metabolites are being widely detected in aquatic environments and there is a growing interest in assessing potential risks of these substances to fish and other non-target species. Ibuprofen is one of the most commonly used analgesic drugs and no peer-reviewed laboratory studies have evaluated the tissue specific bioconcentration of ibuprofen in fish. In the current study, fathead minnow (Pimephales promelas) were exposed to 250 μg L−1 ibuprofen for 28 d followed by a 14 d depuration phase. In a minimized bioconcentration test design, channel catfish (Ictalurus punctatus) were exposed to 250 μg L−1 for a week and allowed to depurate for 7 d. Tissues were collected during uptake and depuration phases of each test and the corresponding proportional and kinetic bioconcentration factors (BCFs) were estimated. The results indicated that the BCF levels were very low (0.08-1.4) implying the lack of bioconcentration potential for ibuprofen in the two species. The highest accumulation of ibuprofen was observed in the catfish plasma as opposed to individual tissues. The minimized test design yielded similar bioconcentration results as those of the standard test and has potential for its use in screening approaches for pharmaceuticals and other classes of chemicals.  相似文献

Copper sulfate (CuSO4) is applied periodically to commercial channel catfish (Ictalurus panctatus) ponds as an algicide or parasiticide. Current understanding of the chemistry of copper in soil-water systems suggests that copper may accumulate in pond sediments, although the forms and potential bioavailability of copper in catfish pond sediments are not known. This study investigated the accumulation and distribution of copper in the sediment of catfish ponds receiving periodic additions of CuSO4.5H2O. All ponds were constructed in Sharkey (very-fine, smectitic, thermic Chromic Epiaquert) soil. Nine 0.40-ha ponds received 59 applications of 2.27 kg CuSO4.5H2O per application per pond over 3 yr; no CuSO4.5H2O applications were made to nine additional ponds. Total Cu concentration in the sediments of CuSO4.5H2O-amended catfish ponds (172.5 mg kg(-1)) was four to five times higher than that in the sediments of nonamended ponds (36.1 mg kg(-1)). Copper accumulated in catfish pond sediments at a rate of 41 microg kg(-1) dry sediment for each 1 kg ha(-1) of CuSO4. 5H2O applied to ponds. Copper in the sediments of amended ponds was mainly in the organic matter-bound (30.7%), carbonate-bound (31.8%), and amorphous iron oxide-bound (22.1%) fractions with a considerable fraction (3.4%; 3 to 8 mg kg(-1)) in soluble and exchangeable fractions. This indicates that Cu accumulates differentially in various fractions, with proportionally greater initial accumulation in potentially bioavailable forms. However, toxicity bioassays with amphipods (Hyallela azteca) and common cattail (Typha latifolia L.) indicated that the effect of exposure to amended or nonamended pond sediments was not different.  相似文献

Surfaces from the habitat of adult Haliotis rubra were tested as inducers of larval settlement to determine the cues that larvae may respond to in the field. Settlement was high on the green algal species Ulva australis and Ulva compressa (Chlorophyta), the articulated coralline algae Amphiroa anceps and Corallina officinalis, and encrusting coralline algae (Rhodophyta). Biofilmed abiotic surfaces such as rocks, sand and shells did not induce settlement. Ulvella lens was also included as a control. Treatment of U. australis, A. anceps and C. officinalis with antibiotics to reduce bacterial films on the surface did not reduce the settlement response of H. rubra larvae. Similarly, treatment of these species and encrusting coralline algae with germanium dioxide to reduce diatom growth did not significantly reduce larval settlement. These results suggest that macroalgae, particularly green algal species, may play an important role in the recruitment of H. rubra larvae in the field and can be used to induce larval settlement in hatchery culture.  相似文献

Ecological effects of weathered and unweathered Louisiana crude oil spilled in eastern coastal plain estuaries were evaluated at all trophic levels and the major chemical components of the oil were followed through selected components of the ecosystem for 36 months.The study site consisted of a natural estuarine marshcreek habitat off the York River, Virginia, modified by the construction of retaining walls to form aquatic and marsh areas in each experimental unit. Each system was closed on all sides with the exception of an opening below the level of lowest tide to allow communication with tidal flow. Five experimental units were constructed, each having a surface area of about 810 m2, containing 695 m2 of marsh, 100 m2 of open water and 15 m2 of intertidal mud flat. The most upstream unit served as the control and the four downstream units received dosages of fresh and weathered crude oil. In addition, biological sampling in the open marsh and creek areas was conducted to provide an estimate of the effects of containing the marsh.Five hundred and seventy liters of fresh South Louisiana crude were added to each of the two downstream experimental enclosures approximately three hours into flood tide and were completed in 1 h. The weathered oil was similarly applied to the upstream units 3 days later.The weathered oil was not visible one week after the spill except as coatings on marsh grass blades, while the fresh crude remained for over three weeks. Plankton populations recovered within a week, and fish mortalities, which were most pronounced in the weathered oil unit, ceased after 10 days.Reductions in standing crops of marsh grasses were most pronounced the year following the spills, when peak biomass in the control exceeded the spill units by a factor of three. Recovery progressed further in the second year with peak biomass in the oiled units being about 70% of the control. The third year following the spill, recovery was nearly complete.Benthic populations were dominated numerically by oligochaetes. Significant reductions in populations of this dominant organisms and in populations of polychaetes and amphipods were demonstrable for a period of three years although most of the populations have shown definite signs of recovery.  相似文献

Sediments were collected from a stream (upstream, outfall and downstream) receiving copper laden catfish pond effluent to assess toxicity to non-target biota. No significant reduction in Hyalella azteca survival or growth (10 d), or Typha latifolia germination and root and shoot growth (7 d) were observed after exposure to upstream and outfall sediments. A significant reduction in H. azteca survival was observed after exposure to the downstream sediment sample; however, no reduction in T. latifolia germination or seedling growth was detected. Bulk sediment copper concentrations in the upstream, outfall and downstream samples were 29, 31, and 25 mg Cu/kg dry weight, respectively. Interstitial water (IW) concentrations ranged from 0.053 to 0.14 mg Cu/l with 10 d IW toxicity units > or = 0.7. Outfall samples were amended with additional concentrations of copper sulfate so that bulk sediment measured concentrations in the amended samples were 172, 663, 1245, and 1515 mg Cu/kg dry weight. Survival was the most sensitive endpoint examined with respect to H. azteca with a no observed effects concentration (NOEC) and lowest observed effects concentration (LOEC) of 1245 and 1515 mg Cu/kg, respectively. NOEC and LOEC for T. latifolia root growth were 663 and 1245 mg Cu/kg, respectively. IW copper concentrations were > or = 0.86 mg Cu/l with H. azteca intersitial water toxicity unit (IWTU) concentrations > or = 1.2. Sequential extraction qualitatively revealed the carbonate and iron oxide fractions which accounted for a majority of the copper binding. In this instance, the copper which was applied to catfish ponds does not appear to be adversely impacting the receiving stream system.  相似文献

Mercury in sediment and fish from North Mississippi Lakes   总被引:7,自引:0,他引:7  
Sediments and/or fish were collected from Sardis, Enid and Grenada Lakes, which are located in three different watersheds in North Mississippi, in order to assess mercury contamination. The mean total mercury concentration in sediments from Enid Lake in 1997 was 0.154 mg Hg/kg, while 1998 sediment concentrations in Sardis, Enid and Grenada Lakes were 0.112, 0.088 and 0.133 mg Hg/kg, respectively. Sediment mercury concentrations in 1999 were similar in all three lakes but, generally lower than 1998. Mean total mercury concentrations in edible fillets of fish collected from Enid Lake in 1998 were above the human health FDA action level (>1.0 mg Hg/kg) for bass (1.40), crappie (1.69) and gar (1.89); however, tissue concentrations were less than 1.0 mg Hg/kg in carp (0.63) and catfish (0.82). Human hazard indexes for each species was 1 for both adults and children, indicating that there is a potential for toxic effects to occur. In addition, calculated consumption limits indicate that adults may consume 4–12 oz. of fish per month, depending on the species consumed. For children, 2 oz. per month may be consumed. Further studies are needed to determine the exact environmental consequences and human health impacts associated with mercury contamination in North Mississippi and the Southeastern United States.  相似文献
