The most common mutation in alpha-1-antitrypsin deficiency, conversion of a G to an A at base 9989 (PI-Z), was detected with the chemical cleavage of mismatch method, demonstrating the power of the method for prenatal diagnosis. Exon V of the gene was amplified using the polymerase chain reaction and heteroduplexes were formed to test for the presence of the mutation. The predicted C mismatch was readily detectable with hydroxylamine, and by making the probe from the chorionic villus sample it was possible to determine that the fetus was heterozygous, not homozygous, for the mutation. 相似文献

Providing an accurate estimate of the dry component of N deposition to low N background, semi-natural habitats, such as bogs and upland moors dominated by Calluna vulgaris is difficult, but essential to relate nitrogen deposition to effects in these communities. To quantify the effects of NH3 inputs to moorland vegetation growing on a bog at a field scale, a field release NH3 fumigation system was established at Whim Moss (Scottish Borders) in 2002. Gaseous NH3 from a line source was released along of a 60 m transect, when meteorological conditions (wind speed >2.5 m s?1 and wind direction in the sector 180–215°) were met, thereby providing a profile of decreasing NH3 concentration with distance from the source. In a complementary study, using a NH3 flux chamber system, the relationships between NH3 concentrations and cuticular resistances were quantified for a range of NH3 concentrations and micrometeorological conditions for moorland vegetation. Cuticular resistances increased with NH3 concentration from 11 s m?1 at 3.0 μg m?3 to 30 s m?1 at 30 μg m?3. The NH3 concentration data and the concentration-dependent canopy resistance are used to calculate NH3 deposition taking into account leaf surface wetness. The implications of using an NH3 concentration-dependent cuticular resistance and the importance for refining critical loads are discussed. 相似文献

Relationships between riparian land cover, in-stream habitat, water chemistry, and macroinvertebrates were examined in headwater streams draining an agricultural region of Illinois. Macroinvertebrates and organic matter were collected monthly for one year from three intensively monitored streams with a gradient of riparian forest cover (6, 22, and 31% of riparian area). Bioassessments and physical habitat analyses were also performed in these three streams and 12 other nearby headwater streams. The intensively monitored site with the least riparian forest cover had significantly greater percent silt substrates than the sites with medium and high forest cover, and significantly higher very fine organics in substrates than the medium and high forested sites. Macroinvertebrates were abundant in all streams, but communities reflected degraded conditions; noninsect groups, mostly oligochaetes and copepods, dominated density and oligochaetes and mollusks, mostly Sphaerium and Physella, dominated biomass. Of insects, dipterans, mostly Chironomidae, dominated density and dipterans and coleopterans were important contributors to biomass. Collector-gatherers dominated functional structure in all three intensively monitored sites, indicating that functional structure metrics may not be appropriate for assessing these systems. The intensively monitored site with lowest riparian forest cover had significantly greater macroinvertebrate density and biomass, but lowest insect density and biomass. Density and biomass of active collector-filterers (mostly Sphaerium) decreased with increasing riparian forest. Hilsenhoff scores from all 15 sites were significantly correlated with in-stream habitat scores, percent riparian forest, and orthophosphate concentrations, and multiple regression indicated that in-stream habitat was the primary factor influencing biotic integrity. Our results show that these "drainage ditches" harbor abundant macroinvertebrates that are typical of degraded conditions, but that they can reflect gradients of conditions in and around these streams. 相似文献

Elemental concentrations and bioaccessibility were determined in background soils collected in Canada as part of the North American Geochemical Landscapes Project. The concentrations of As, Cr, Cu, Co, Ni and Zn were higher in the C-horizon (parent material) compared to 0–5 cm (surface soil), and this observation along with the regional distribution suggested that most of the variability in concentrations of these elements were governed by the bedrock characteristics. Unlike the above-stated elements, Pb and Cd concentrations were higher in the surface layer reflecting the potential effects of anthropogenic deposition. Elemental bioaccessibility was variable decreasing in the order Cd > Pb > Cu > Zn > Ni > Co > As > Cr for the surface soils. With the exception of As, bioaccessibility was generally higher in the C-horizon soils compared to the 0–5 cm soils. The differences in metal bioaccessibility between the 0–5 cm and the C-horizon and among the provinces may reflect geological processes and speciation. The mean, median or 95th percentile bioaccessibility for As, Cr, Cu, Co, Ni and Pb were all below 100 %, suggesting that the use of site-specific bioaccessibility results for these elements will yield more accurate estimation of the risk associated with oral bioavailability for sites where soil ingestion is the major contributor of human health risk. 相似文献

Environmental nongovernmental organizations (NGOs) in Australia have struggled to generate and sustain public concern about climate change. If debates about climate policy can be viewed as sites of contestation between competing actors, Australia’s environmental NGOs have found it difficult to compete against countervailing forces that have sought to shape public attitudes to climate action and the contours of policy responses. While to a significant degree this reflects the power of those forces and the sentiments of the government of the day, there is also a case to be made that some of Australia’s most prominent environmental NGOs have appeared wedded to strategies inconsistent with building or sustaining public support for action or guiding policy responses. How have Australia’s largest environmental NGOs engaged climate politics, and why has this engagement taken that form? Pierre Bourdieu’s political sociology provides unique insight for coming to terms with the multifaceted nature of the constraints, opportunities, and drivers of political action, from the context of climate politics to the forces behind Australian NGOs’ engagement with that politics, and the limits of that engagement. Bourdieu’s work also suggests possible avenues for more effective forms of political communication on climate change in the Australian context. 相似文献

In this paper, the island of Tasmania is considered as a government jurisdiction. Management at this scale can conflict with sustainability in local environments. The case used to make this argument is the Tasmanian commercial rock lobster fishery. Within this fishery, Tasmania-wide management paradoxically intensifies differences at the same time as it effaces them. These differences contrast with Tasmania-wide management claims regarding sustainability. Actor-network theory is used to help approach these tensions. Land application of manure is a common practice in the Upper Midwest of the United States. Recently, there have been concerns regarding the effect of this practice on water quality, especially when manure is applied during winter over frozen soils. A study undertaken on a Rozetta silt loam (fine-silty, mixed, superactive, mesic Typic Hapludalfs) at Lancaster, WI, evaluated the effects of tillage and timing of manure application on surface and subsurface water quality. The daily scrape and haul liquid dairy manure was applied either in the fall (before snow) or in winter (over snow with frozen soil underneath) to be compared with no manure under two tillage systems (no-till and chisel-plowing). In this paper, we report results on the effects of the above treatments on mineral N leaching. Percolation and mineral N leaching during the nongrowing season were, respectively, 72 and 78% of the annual losses, mainly because of the absence of plant water and N uptake. Percolation was generally higher from no-till compared with chisel-plow but there was no significant effect of tillage on mineral N concentration of the leachate or mineral N losses via leaching. Mineral N leaching was statistically higher from the manure-applied vs. no-manure treatment, but there was no difference between winter-applied manure and no-manure treatments. There were significant tillage by manure interactions with fall manure application followed by chisel-plowing resulting in highest N leaching losses. Averaged over the two years, N leaching rates were 52, 38, and 28 kg N ha(-1) yr(-1) from fall-applied, winter-applied, and no-manure treatments, respectively. These results show that there is substantial N leaching from these soils even when no fertilizer or manure is applied. Furthermore, fall-applied manure followed by fall tillage significantly increases N leaching due to enhanced mineralization of both soil and manure organic N. 相似文献

Quality and Quantity of Suspended Particles in Rivers: Continent-Scale Patterns in the United States 总被引:2,自引:0,他引:2
Suspended solids or sediments can be pollutants in rivers, but they are also an important component of lotic food webs. Suspended sediment data for rivers were obtained from a United States–wide water quality database for 622 stations. Data for particulate nitrogen, suspended carbon, discharge, watershed area, land use, and population were also used. Stations were classified by United States Environmental Protection Agency ecoregions to assess relationships between terrestrial habitats and the quality and quantity of total suspended solids (TSS). Results indicate that nephelometric determinations of mean turbidity can be used to estimate mean suspended sediment values to within an order of magnitude (r2 = 0.89). Water quality is often considered impaired above 80 mg TSS L–1, and 35% of the stations examined during this study had mean values exceeding this level. Forested systems had substantially lower TSS and somewhat higher carbon-to-nitrogen ratios of suspended materials. The correlation between TSS and discharge was moderately well described by an exponential relationship, with the power of the exponent indicating potential acute sediment events in rivers. Mean sediment values and power of the exponent varied significantly with ecoregion, but TSS values were also influenced by land use practices and geomorphological characteristics. Suspended fine particles (seston) are an important component of energy and nutrient cycling in streams, but they can also be pollutants. We examined seston dynamics and filter-feeding macroinvertebrate communities in sites representing headwaters to large rivers in the Kansas River drainage, northeastern KS. Seston samples were collected at least seasonally during low to moderate flows for one year beginning in the summer of 1999, and quality was assessed by determining organic content and C to N ratio. A rapid bioassessment approach was used to examine filter-feeders. Relationships between stream size and seston concentrations were markedly influenced by anthropogenic activities. There was no relationship between total seston concentration and stream size across all sites (r = 0.14, p > 0.05), but a significant, positive relationship was evident when impounded and suburban sites were excluded (r = 0.73, p < 0.01); this same trend was evident for organic and inorganic components. Seasonal patterns of C to N ratio were evident, with generally lower values during winter and highest values in summer. However, seasonal patterns were dampened in suburban sites and virtually absent below impoundments. Filter-feeder richness was correlated with average organic seston concentrations (r = 0.8, p < 0.01), but this relationship was also obscured by impoundments and suburban development. In particular, impoundments had a dramatic, negative effect on richness. Abundance of most hydropsychid caddisfly taxa was positively correlated with organic seston concentration. Results indicate there are significant patterns regarding seston, filter-feeders, and stream size in this Great Plains river system, but patterns are strongly influenced by human activities. Stream Invertebrate Communities, Water Quality, and Land-Use Patterns in an Agricultural Drainage Basin of Northeastern Nebraska, USA 总被引:2,自引:0,他引:2
We used invertebrate bioassessment, habitat analysis, geographic information system analysis of land use, and water chemistry
monitoring to evaluate tributaries of a degraded northeast Nebraska, USA, reservoir. Bimonthly invertebrate collections and
monthly water chemistry samples were collected for two years on six stream reaches to identify sources contributing to reservoir
degradation and test suitability of standard rapid bioassessment methods in this region. A composite biotic index composed
of seven commonly used metrics was effective for distinguishing between differentially impacted sites and responded to a variety
of disturbances. Individual metrics varied greatly in precision and ability to discriminate between relatively impacted and
unimpacted stream reaches. A modified Hilsenhoff index showed the highest precision (reference site CV = 0.08) but was least
effective at discriminating among sites. Percent dominance and the EPT (number of Ephemeroptera, Plecoptera, and Trichoptera
taxa) metrics were most effective at discriminating between sites and exhibited intermediate precision. A trend of higher
biotic integrity during summer was evident, indicating seasonal corrections should differ from other regions. Poor correlations
were evident between water chemistry variables and bioassessment results. However, land-use factors, particularly within 18-m
riparian zones, were correlated with bioassessment scores. For example, there was a strong negative correlation between percentage
of rangeland in 18-m riparian zones and percentage of dominance in streams (r

= 0.90, P < 0.01). Results demonstrate that standard rapid bioassessment methods, with some modifications, are effective for use in this agricultural
