Metals have been reported to inhibit organic pollutant biodegradation; however, widely varying degrees and patterns of inhibition have been reported. To investigate the roles of medium composition and metal bioavailability on these different degrees and patterns of inhibition, we assessed the impact of cadmium on naphthalene biodegradation by a newly isolated strain of Comamonas testosteroni in three chemically-defined minimal salts media (MSM): Tris-buffered MSM, PIPES-buffered MSM, and Bushnell-Haas medium. Cadmium (total concentrations of 100 and 500 microM) inhibited biodegradation in each medium. Degrees of inhibition were different in each medium. Cadmium was most inhibitory in PIPES-buffered MSM and least inhibitory in Bushnell-Haas. For example, in Bushnell-Haas medium, 100 microM cadmium reduced the cell yield more than 4-fold compared to controls not containing cadmium. The same concentration of cadmium completely inhibited growth in PIPES-buffered MSM. No difference in inhibition was observed in any medium when cadmium was added 24 h before inoculation rather than when added within one minute of inoculation. Two patterns of inhibition were observed. Inhibition occurred in a dose dependent pattern in Tris- and PIPES-buffered MSM and in a non-dose dependent pattern in Bushnell-Haas. Specifically, in Bushnell-Haas, 100 microM total cadmium extended the lag phase by 23+/-8.66 h, whereas 500 microM did not extend the lag phase. Soluble, ionic cadmium (Cd2+) concentrations were measured and modeled in each medium to assess cadmium bioavailability. In media containing 500 microM total cadmium, bioavailability was highest in Tris- and PIPES-buffered MSM and lowest in Bushnell-Haas. In Bushnell-Haas, cadmium bioavailability was initially higher in the 500 microM treatments (196+/-21.2 microM) than in the 100 microM treatments (78.2+/-2.04 microM); however, after 12 h, bioavailability was higher in the 100 microM treatments (56.4+/-24.8 micro) than the 500 microM treatments (13.3+/-1.2 microM). These data suggest that the type of medium determines the degrees and patterns by which metals inhibit biodegradation and emphasize the importance of coupling metal toxicity and bioavailability data.  相似文献

Biodiversity conservation has emerged within the past two decades as one of the most important global challenges confronting national planners, world bodies, professionals and academics. Governments faced with increasing biodiversity loss as a result of human activities have resorted to the creation of protected areas as a strategy to both slow down habitat loss and/or degradation and eventually mitigate species extension and reduction rates. However, the creation of a protected area can have strong implications on the livelihoods of people inhabiting the forest and depending on it, especially those caught within its borders. The involvement of such inhabitants in the management process of the protected area can be profitable. This paper discusses the case of the Korup National Park, Cameroon, considered in the late 1980s by some to be a flagship of conservation and development efforts, and later on in the late 1990s by others as a catastrophic failure as an example of integrated conservation and development. As a means of updating the program's management information base, an in-depth participatory and socio-ecological survey was conducted by some of the program's technical staff. This study aimed at appraising the extent to which the Park's human community of 4200 inhabitants continued to relate to its resources and depend on them for their livelihood. The aim was to evaluate the potential links between the communities' livelihood and the long-term management and survival of the Park as the important biodiversity conservation zone it had been found to be. Results indicate that the successful management of a Park like Korup may well depend on the involvement of the local communities; and that successful management through approaches that minimize the potential contributions and aspirations of the local people is difficult to achieve.  相似文献

Costal sediment samples from Great Kwa River as well as adjoining termite nest and soil samples were analyzed for quantitative determination of polycyclic aromatic hydrocarbons (PAHs) and sterols using gas chromatography–mass spectrometry (GC–MS) in order to access the possibility of transport of biologically produced PAHs/sterols from termite nest to the sediments. The total PAH concentrations (sum of parent and alkyl) for the sediments ranged between 131.96 and 139.35 ng/g dry weight (dw) while those for the nest and soil samples were in the range 9.51–9.71 and 71.85–77.26 ng/g dw, respectively. These levels of PAHs in sediments were relatively low compared to other urban/industrial Asian and American countries. No evidence of the usual biologically produced PAHs was found, thus reducing the likelihood of transport of these compounds from the nest to the sediments. The absence of parent and alkyl PAHs in central compartment of the nest may reflect the selective fern leaves feeding pattern of the dominant termite species prevalent in the vicinity of the study site. Utilization of six selected PAH ratios such as Fla/(Fla + Pyr) (0.4–0.5), Ant/(Ant + Phe) (0.25–0.90), BaA/(BaA + Chrys) (0.45–0.61), MP/P (0.05–6.81), 1,7/(1,7?+?2,6)-DMP (0.61–0.95), and LPAH/HPAH ( 2.80–3.80) allows discrimination of PAH sources for the samples to be made with a mixed source dominance observed. Examination of sterol distributions in the samples shows relatively high abundance of cholest-5-en-3β-ol in central compartment of the nest, considered here as a consequence of metabolic conversion of phyto-/fungi sterols in the tissues of the termite species. The relatively reduced levels of stanol compounds in central compartment of the nest may be associated with their utilization by the termites for growth and development.  相似文献

The inventory of profile distribution of total iron(Fe),zinc(Zn), manganese(Mn) and copper(Cu) were determined in three different soil horizons each of the wetland soils selected form Mbiabet(MB),Nkari(NK)and Nkana(NA) in Ini Local Government Area of Akwa Ibom State.Total Fe ranged between 3.25 and 4.15ppm.The average contents were 3.72,3.91 and 3.62ppm in Mbiabet(MB),Nkari(NK) and Nkana(NA)soils respectively.The total value of Zn also ranged between 2.4 and 4.9 ppm with the average content in each soil being 28.27,17.73and 36.53 ppm respectively.The amount of Fe and Zn in these soil profiles were strongly correlated with the clay content and high levels of organic matter of 3.70%,2.47%and 2.5% respectively.The content clearly reflected a poor drainage conditions.In all the soil profiles Mn and Cu were detected in at least one of the soil horizons.However, Mn and Cu were not detected in the soil horizons at Nkari.Generally , the relative inventory of these micronutrients appeared to be influenced by pH,drainage pattern,organic matter and clay contents of these soils.The inventory of total values of the wetland soils considered are assessed in the light of establishing a baseline information.  相似文献
