Some of the people living in the Chukotka Peninsula of Russia depend heavily on marine mammals, but little is known of the exact dietary patterns and plasma levels of POPs among these populations. In this study, POPs levels in plasma from 50 participants from the isolated community of Uelen (Bering Strait) were determined and related to dietary information obtained through a food frequency questionnaire. The intake of marine mammals was high and the combined intake of blubber from walrus, seal and whale was a significant predictor (p < 0.01) of plasma concentrations of sum PCBs and borderline for sum CDs (p = 0.02) and sum DDTs (p = 0.04). There was a significant gender difference in the levels of POPs, and among women there was a significant increase with age. Extensive breastfeeding and lower blubber intake among women could be possible explanations for this gender difference. Despite the high intake of blubber the plasma levels of PCBs and DDTs were lower than some of those reported for the East Coast of Greenland. The geometric mean values for sum PCBs (17 congeners) and sum DDTs were 1316 ng g(-1) lipids and 563 ng g(-1) lipids, respectively. PCB 163, which partly co-eluted with PCB 138, was found in high concentrations (40% of PCB 138). This raises questions regarding the validity of using PCB 138 and PCB 153 to calculate the level of Arochlor 1260. The geometric mean of sum CDs was 518 ng g(-1) lipids. Concentrations of beta-HCH (geometric mean; 410 ng g(-1) lipids) were higher than observed for other native populations depending on marine mammals. Transportation of beta-HCH by ocean currents through the Bering Strait into the Arctic Ocean or regional point sources might explain these elevated levels.  相似文献

Divergent scientific and regulatory agency perspectives about contaminants in fish have lead to contradictory advice and confusing public messages about its consumption. The evidence for the protective effect of eating fish on cardiovascular outcomes is considered to be convincing. It has long been attributed to n-3 unsaturated long-chain fatty acids. Persistent organic pollutants (POPs) are compounds that are lipid soluble and accumulate in the aquatic food chain. Despite a considerable decrease in their levels in fish during the last two decades, there is still significant concern about potential negative health effects and an ongoing debate exists about what type of fish consumption advisories are most suitable. In this review our aim is twofold, namely to explore: (1) the underlying causes for the conflicting recommendations by discussing the strengths and limitations of risk assessment and epidemiological evidence; and (2), the role of risk management in formulating public dietary advisories. It is our view that the latter advice is most appropriately formulated in the context of risk management, of which both epidemiologic evidence and risk assessment are essential components.  相似文献

The traditional northern Norwegian fish dish "m?lje", consisting of boiled cod, cod liver, cod liver oil and hard roe, is still consumed frequently during the winter months January to March. The liver of the cod is rich in lipids and the levels of persistent organic pollutants (POPs) are relatively high. To better understand the short-term consequences of this traditional meal on the plasma levels of PCBs and p,p'-DDE, individual intake of liver and cod liver oil during one meal was measured. Blood samples were collected from 33 participants before the meal, and then 4 h, 12 h and 5 days after it. Lipid-weight and wet-weight levels of 10 PCB congeners and p,p'-DDE were determined in the plasma samples and the food. The plasma levels of p,p'-DDE was found to increase significantly from 0 to 4 h, both when expressed as wet-weight (35% change) and lipid-weight (20% change). The corresponding changes (0-4 h) in wet-weight levels of the most prevalent PCB congeners were non significant. By contrast, PCB congeners with low levels in the food showed a significant drop in lipid-weight levels during the first 4 h. The observed changes were independent of amount consumed. Significant differences in fasting and non-fasting samples were found for most PCBs and p,p'-DDE. For the lipid weight levels of sum PCBs there was a significant decrease of 16% from non-fasting to fasting samples. To obtain reliable data on human levels of POPs it is, on the basis of these findings, recommended that blood samples should be collected from fasting individuals and both wet-weight and lipid-weight levels should be reported.  相似文献

Long-range transport of pollutants towards circumpolar regions emphasizes the need for up-to-date and reliable biological monitoring data. This paper explores the use, reliability and availability of maternal blood (MB) and plasma (MP), cord blood (CB) and plasma (CP) and mother's milk (MM) in terms of assessing exposure to persistent toxic substances (PTSs). It is concluded that MP has the best combination of availability, sensitivity in terms of number of PTSs, their detection frequency and concentrations, and physiological relevance. The study group consisted of 48 pregnant women of indigenous origin from the Chuchki district in the eastern Russian arctic. Blood, CB and MM specimens were collected from all women and MP, CP and MM were analyzed for the Arctic Monitoring and Assessment Programme (AMAP) suite of organochlorines (OCs) and metals (Pb and Cd in MB and CB). Generally speaking, the levels of PTSs coincided with those indicated in several AMAP publications from Chukotka and other areas of northern Russia. The correlations of PTS concentrations between the three body fluid compartments exceeded the minimum statistical requirements of alpha = 0.05 and beta = 0.20 for most of the compounds, with r > 0.46 except for Cd (r = 0.05); lipid adjustments for the OCs did not affect the r-values to any significant extent. The majority of the inter-OC correlations within compartments also fulfilled the indicated statistical condition. Careful consideration is given to the replacement of concentrations below the detection limit, OC detection frequency, the criteria for log-transformation of the data, analytical uncertainty, and biological variability. Practical implications of the findings are explored.  相似文献

The aim of the study was to investigate the hypothesis that consumption of fish liver increases cancer risk in humans due to increased intake of persistent organic pollutants (POPs). This study is based on data from the Norwegian Women and Cancer Study (NOWAC). The study has a prospective cohort design with questionnaire data from 64 285 randomly selected Norwegian women (aged 40-70 at baseline) and linkage to the Norwegian Cancer Registry. Cox proportional hazards regression was used to calculate risk ratios associated with consumption of fish liver and total cancer and cancer in breast, uterus, and colon. Fish liver consumption was, after adjusting for known risk factors, associated with a significant reduced risk for total cancer (RR = 0.92, 95% CI: 0.85, 0.99), and non-significant reduced risk for breast cancer (RR = 0.90, 95% CI: 0.78, 1.04), and colon cancer (RR = 0.82, 95% CI: 0.63, 1.07). Relative risk for uterus cancer was 0.82 (95% CI: 0.61-1.12). No significant dose-response effect was found for frequency of fish liver consumption (when divided into three intake groups) and total cancer, nor for any of the other cancer sites.We have concluded that in Norwegian women, fish liver consumption was not associated with an increased cancer risk in breast, uterus, or colon. A trace analytical method is presented for the analysis of hydroxylated polychlorinated biphenyl metabolites (HO-PCBs) and pentachlorophenol (PCP) in human plasma. The described methodology is a modification of a previously validated method used for PCB and organochlorine pesticide analysis. The modified method enables the combined analysis of phenolic and neutral halogenated compounds. A tandem Florisil column is used for separating the HO-PCBs and PCP from the neutral fraction, instead of the more common chemical partitioning. In the same step the neutral fraction is purified for GC analysis. The extraction of the HO-PCBs and PCP was found to be highly dependent on sufficient acidification of the sample and the polarity of the extracting solvent. Analysis of plasma samples gave recovery rates for (13)C(6)-PCP and (13)C(12)-4-HO-CB 187 of 64 and 72%, respectively. The limit of detection ranged between 2-20 pg g(-1) plasma for the HO-PCBs and 5 pg g(-1) plasma for PCP. No matrix interferences were observed in the chromatograms. In plasma samples (n = 15) from the native Chukchi people in Uelen (Russian Arctic), a population with high PCB exposure, the median ratio of sum HO-PCBs to sum PCBs was as high as 0.4 and the sum HO-PCBs and PCBs were significantly correlated (r(2) > 0.7, p < 0.01). The median sum HO-PCBs (10 congeners) was 5920 pg g(-1) plasma with 4-HO-CB 107 as the dominating congener (median: 1670 pg g(-1) plasma). The median PCP level was measured at 642 pg g(-1) plasma.  相似文献

Increased cancer incidence and mortality have been found among humans exposed to high levels of organochlorines (OCs), either accidentally or as industrial workers. In order to assess levels of OCs in Norwegian women north of the Arctic Circle and validate self-reported fish consumption as a surrogate measure of organochlorine body burden, concentrations of seven polychlorinated biphenyl (PCB) congeners [IUPAC Nos. CB-105, CB-118, CB-138 (+ CB-163), CB-153, CB-180, CB-183, CB-187], beta-hexachlorocyclohexane (beta-HCH), 2,2'-bis(p-chlorophenyl)-1,1-dichloroethylene (p,p'-DDE) and cis- and trains-chlordane (c-CD and t-CD) were examined in plasma samples of middle-aged women attending for health screening. Altogether, 47 of those invited (81%) completed a questionnaire and donated a suitable blood sample. The ability of questionnaire data to predict plasma levels of OCs was tested in linear and logistic regression analyses. Measured plasma concentrations were in the range reported for the general female population of other Western countries and the relative amounts of PCBs were similar to the circumpolar pattern. Intake of seagulls' eggs was a predictor of PCB congeners CB-138 (+CB-163) (p<0.05) and CB-153 (p<0.01). No other food category was positively associated with any compound. In contrast, duration of residence in the study municipality, body mass index (BMI) and lifetime lactation (months) were the best univariate predictors. There was an increase in beta-HCH, p,p'-DDE and most of the PCBs (p<0.05 for all) with increasing length of time a subject had lived in the municipality. BMI was a positive predictor for beta-HCH (OR=3.10, 95% CI 1.50-6.43, per 5 kgm(-2)), chlordane (OR=2.13, 95% CI 1.12-4.05, per 5 kgm(-2)) and CB-105 and CB-153 (p<0.05 for both). Lactation was negatively associated with all OCs (p<0.05), except chlordane and two of the PCB congeners. Time living in the municipality and lactation explained 34%, of the variance in concentration of total PCB in a multivariate model (p<0.001). The results indicate that regular consumption of fish (mostly lean species) from the Norwegian waters is not associated with an increased body burden of OCs (e.g., of importance to cancer development), although they confirm that lactation is the most important elimination route of these contaminants in women.  相似文献

The objective of this study was to investigate changes in the levels of toxic and essential elements in maternal blood during pregnancy and subsequently. A subset of 211 pregnant women from the North Norwegian Mother-and-Child Study was included. Blood samples were collected during the 2(nd) trimester (P1) and postpartum (day 3, P2; and 6 weeks, P3) in different regions of northern Norway, and were analyzed for a suite of 10 selected elements. The latter feature three general but distinct concentration patterns across the three collection periods, namely: progressive increase [Group 1: As, Cd (non-smokers only), Mn, Pb and Zn]; V-shaped, with a minimum at P2 [Group 2: Cd (smokers), Hg, Mo and Se]; and downward V, with a maximum at P2 [Group 3: Co and Cu]. These trends are interpreted in the context of underlying metabolic, hematological and physiological changes that occur in mothers, as well the biochemistry and accumulation preferences of these elements within the whole blood compartment and breast milk. Implications for biomonitoring strategies are discussed. In a multivariable analysis of the P1 data, fish consumption was a robust positive predictor for Hg (p < 0.02), As (p < 0.01) and Se (p ≤ 0.001) blood concentrations, multivitamin intake for Se (p < 0.001), and parity for Co (p < 0.002); age reached significance only for Hg (p < 0.001). An inverse association was observed between parity and Hg (p < 0.05). For the other elements, predictor patterns were not evident.  相似文献

In low-income countries, the use of some organochlorine pesticides is still common in order to increase food production. Monitoring the chemical exposure is an important step in risk-reducing strategies. This is the first study to report concentrations of organochlorines in breast milk of women from Bangladesh where farming is the main income source.Organochlorines such as p,p′-DDT, o,p′-DDT, p,p-DDE, p,p-DDD (i.e., ∑DDT), HCB, α-, β- and γ-HCH, trans-chlordane, cis-chlordane, oxy-chlordane, trans-nonachlor, cis-nonachlor, mirex and polychlorinated biphenyls (CB 28, 52, 99, 101, 105, 114, 118, 123, 128, 138, 141, 149, 153, 156, 157, 163, 167, 170, 180, 183, 187, 189, 194) were analyzed in breast milk collected in 2002 from 72 first-time mothers (median age 20 years) living in the rural area Matlab, Bangladesh.While the concentrations of PCBs and many of the pesticides were low, the concentrations of p,p′-DDT and its metabolite p,p′-DDE were high (median 349 and 1645 ng g−1 lipid, respectively) in comparison to other countries. The median value of ∑DDT was 2123 ng g−1 lipid. The estimated daily exposure to p,p′-DDT, p,p′-DDE and ∑DDTs was 10, 30 and 42 μg kg−1 body weight, respectively, in 3 months old infants. The p,p′-DDE/p,p′-DDT ratio ranged from 1 to 23, where 58% of the mothers had a ratio below 5 indicating recent or ongoing DDT exposure.This study reports infant exposure and maternal body burden of organochlorines through breast milk. Although the findings give no reason to limit breast-feeding, it is essential to identify the main exposure sources and find means to decrease the exposure.  相似文献

Recent research has again raised the issue regarding the potential health effects of long term exposure to persistent organic pollutants (POPs) in the context of a risk-benefit analysis. There remains clear evidence of the beneficial effects of fish consumption on public health and in particular fatty fish with its essential fatty acids. In addition to providing fatty acids, fish fat constitutes an essential source to vitamin D for the northern Norwegian population. Norwegian Food Control Authorities have recommended that children and women of childbearing age should not consume fish liver due to the risk associated with high intake of POPs. The aim of this study was to assess the influence of fish liver intake on the levels of PCBs and chlorinated pesticides in a rural coastal community (n = 31) in northern Norway, applying a cross-sectional design. The geometric mean plasma levels of sum of PCBs (14 congeners) and p,p'-DDE was found to be 558 microg kg(-1) lipids and 179 microg kg(-1) lipids, respectively. These levels were not significantly affected by the reported intake of fish liver, when age and gender were considered. Age in particular, but also gender were significant predictors for most compounds. When merging the dataset with an urban population with considerable lower intake of fish liver, the intake of cod liver still did not significantly affect the levels of PCBs and p,p'-DDE when adjusted for age and gender. Age and gender remain the strongest predictors of plasma levels of POPs in this study whereas fish liver intake was not significantly associated with the POPs.  相似文献
