The aim of the present study was to demonstrate induction ofvitellogenin in the common carp (Cyprinus carpio) as abiomarker for monitoring freshwater ecosystems. Sexually undifferentiated specimens of common carp were treated experimentally with 17-estradiol and increasing doses of 4-nonylphenol and levels of plasma Vtg were measured in orderto: 1) validate an ELISA assay for plasma Vtg in the common carpusing the polyclonal rabbit anti-salmon Vtg antibody AA1 (Biosense, Norway); 2) check the sensitivity of carp juvenilesin producing Vtg in response to estrogen stimulation. The grouptreated with 17-estradiol showed high induction (156%)with respect to controls, also groups treated with 4-nonylphenolshowed induction of Vtg. The group treated with 100 mg kg-1b.wt showed an induction of 61%. A statistically significantcorrelation was found between dose and response. This preliminary study demonstrate a response to the ELISA assay for Vtg in the common carp using rabbit anti-salmon antibody AA1. With further evidence the tested biomarker might be proposed for large scale monitoring of estrogenic effects caused by pollution in urban and industrial effluents.  相似文献

The biomarker approach has been used for 25 years to study the environmental quality of marine, brackish and freshwater ecosystems. Biomarkers may indicate health status and can be applied to organisms of all zoological phyla by destructive or non destructive methods. For 5 years we have been using this approach in zooplankton to detect ecotoxicological alterations at low levels of the food chain due to contaminants. Here we review our approach to validate and apply biomarker techniques in zooplankton. We discuss advantages, limitations, some results and future research. We indicate that biomarkers in zooplankton can be used as new indices of trophic status and ecological integrity of Italian marine coastal and lagoon environments, to be included among the tools specified by Italian law D.Lgs. 152/2006.  相似文献

A biomonitoring study was performed to evaluate the human impact on two small coves adjacent to the Italian Scientific Station at Terra Nova Bay in November 1995. The study used the fish species Trematomus bernacchii as a bioindicator organism for a biomarker analysis based on porphyrin levels, and BPMO (Benzo(a)pyrene MonoOxygenase) and EROD (Ethoxyresorufin-O-deethylase) activities. Porphyrin levels and EROD and BPMO activities were found to be generally low. In contrast to previous years, no statistically significant difference was found between the potentially contaminated cove and the control cove after the Italian expedition had been active nearby for a period of one month. This indicates a marked decrease in certain types of contaminants such as organochlorines and trace metals, mainly due to improvements in waste disposal.  相似文献

The aim of this study was to validate a multi-trial biomarker approach for the evaluation of toxicological risk due to benzo(alpha)pyrene. Carcinus aestuarii, exposed to increasing concentrations of B(alpha)P in the water, was used as the bioindicator organism. A set of biomarkers were tested in order to: identify biological materials for biomarker and residue analysis; determine a group of sensitive techniques for the assessment of PAH contamination; investigate correlation between responses at different levels of biological organisation. The results underlined that BPMO activities in hepatopancreas and gills were a good biomarker of exposure to PAH-type compounds. B esterases activities in hemolymph and porphyrin patterns in excreta could be proposed as a non-destructive approach for evaluating chemical exposure in this species.  相似文献

Many studies document the chemical stress related to organochlorine (OC) xenobiotics in Mediterranean cetaceans. The aim of this study was to establish a theoretical model to evaluate the hazard to Mediterranean striped dolphins (Stenella coeruleoalba) due to HCB, DDTs and PCB congeners. Differences in OC levels in blubber of stranded and free-ranging specimens enabled us to evaluate the hazard associated with different chlorinated xenobiotics, taking the live population as control sample, assumed to be in good health. For the most toxic compounds, with teratogenic, mutagenic, carcinogenic and endocrine disrupting capacity, we indicate levels beyond which there can be toxicological hazard for the striped dolphin. Using a mathematical formula derived from knowledge of the length and age of 62 stranded specimens, the age of dolphins was estimated and sexual maturity was identified at nine years. This evaluation was important for understanding differences in contaminant burden between males and females.  相似文献

The aim of this study is to propose a bioindicator organism, the lizard Gallotia galloti, and a nondestructive biomarker assay, utilising serum butyrylcholinesterase, for the assessment of the toxicological impact of organophosphorus (OP) insecticides in the Canary Islands. Laboratory and field studies were performed using the OP insecticide Trichlorphon. In the laboratory study, experimental groups of Gallotia galloti were treated with 5, 50 and 100 mg/kg of Trichlorphon, respectively, and after 24 h the following enzyme activities were assayed: brain acetylcholinesterase (AChE), serum butyrylcholinesterase (BChE), microsomal carboxylesterase (CbE) and microsomal 7-ethoxyresorufin dealkylation (EROD). BChE activity was monitored in two groups of lizards treated with 50 and 100 mg/kg of Trichlorphon, respectively, for a period of 21 and 31 days after treatment. In the field study, BChE activity was detected in Gallotia galloti specimens, 24 and 48 h after treatment of an experimental area with 10 kg/ha of Dipterex sp80 (80% Trichlorphon). Three conclusions can be drawn. (1) Gallotia galloti has the features of an ideal bioindicator: high sensitivity to OPs and extremely slow recovery of serum BChE with respect to other vertebrate species; this property extends the temporal application of this biomarker in field studies. (2) A high correlation was found between the destructive biomarker brain AChE and the nondestructive biomarker serum BChE, 24 h after treatment. (3) The results of the field study show the relative 'non-toxicity' of Trichlorphon for nontarget organisms, such as lizards, at the average concentrations used in agriculture.  相似文献

The main aims of the present study, conducted in the framework of the MONIQUA-Egadi Scientific Project, were twofold: first, to make the first step in the development and validation of an ecotoxicological approach for the assessment of marine pollution in coastal environments on the basis of a set of biomarker responses in new sentinel species; and second, to obtain preliminary information on environmental quality in an Italian marine protected area, the Egadi Islands (Sicily). Several cytochrome P450-dependent mixed-function oxidase activities were measured in the following sentinel species: rainbow wrasse Coris julis, gastropod limpet Patella caerulea, and sea urchin Paracentrotus lividus. The results suggest that specimens from the Favignana Harbor may be exposed to P450 inducers, whereas most of the other sites seem to share similar environmental quality. The proposed approach has potential for assessment of environmental quality in marine protected areas.  相似文献

Human pharmaceutical residues are a serious environmental concern. They have been reported to have eco, geno, and human toxic effects, and thus their importance as micropollutants cannot be ignored. These have been studied extensively in Europe and North America. However, African countries are still lagging behind in research on these micropollutants. In this study, the wastewaters of the University Teaching Hospital of Yaoundé (UTHY) were screened for the presence of active pharmaceutical ingredients and their metabolites. The screening was carried out using two methods: high-performance liquid chromatography coupled to a triple quadrupole analyzer (LC/QQQ) and high-performance coupled to a mass spectrometer with a time of flight analyzer (LC/Q-TOF). A total of 19 active pharmaceutical ingredients and metabolites were identified and quantified. The compounds identified include paracetamol (211.93 μg/L), ibuprofen (141 μg/L), tramadol (76 μg/L), O-demethyltramadol (141 μg/L), erythromycinanhydrate (7 μg/L), ciprofloxacin (24 μg/L), clarinthromycine (0.088 μg/L), azitromycine (0.39 μg/L), sulfamethoxazole 0.16 μg/L), trimetoprime (0.27 μg/L), caffeine (5.8 μg/L), carnamaeepine (0.94 μg/L), atenolol (0.43 μg/L), propranolol (0.3 μg/L), cimetidine (34 μg/L), hydroxy omeprazole (5 μg/L), diphenhydramine (0.38 μg/L), metformine (154 μg/L), and sucralose (13.07 μg/L).  相似文献
