Pentachlorotoluene (PCT) and pentabromotoluene (PBT) are environmental contaminants detected in the Great Lakes ecosystem. In view of the paucity of toxicity data and the potential for human exposure, a subacute (28 day) and a subchronic (91 day) study were conducted in the rat. In the subacute study, groups of 10 male and 10 female rats were fed the diet containing PCT or PBT at 0, 0.5, 5.0, 50 or 500 ppm for 28-days. In the subchronic study, the group size was increased to 15, the dose levels were 0, 0.05, 0.5, 5.0, 50 and 500 ppm in the diet and the exposure period was 91 days. Growth rate and food consumption were not affected by exposure to either chemical in the subacute and subchronic study. Clinical observations revealed no abnormalities. Decreased hemoglobin was observed in female rats fed 5.0 ppm PCT and higher levels in the subacute (28 day) study. In the same study the hematocrit value and erythrocyte numbers of females fed 5.0 or 500 ppm PCT diets were significantly lower than the control. In both subacute and subchronic studies mild dose-dependent histopathological changes were observed in the thyroid, liver and kidney of rats fed PCT and PBT diets. In general male rats were more susceptible than females to the treatment of PCT and PBT. Based on these data, it was concluded that the no observable adverse effect level for PCT was 50 ppm in the diet (3.5 mg/kg b.w./day) and that of PBT was 5.0 ppm (0.35 mg/kg b.w./day).  相似文献

Incineration is the main option for residual Municipal Solid Waste treatment in France. This study compares the environmental performances of 110 French incinerators (i.e. 85% of the total number of plants currently in activity in France) in a Life Cycle Assessment perspective, considering 5 non-toxic impact categories: climate change, photochemical oxidant formation, particulate matter formation, terrestrial acidification and marine eutrophication. Mean, median and lower/upper impact potentials are determined considering the incineration of 1 tonne of French residual Municipal Solid Waste. The results highlight the relatively large variability of the impact potentials as a function of the plant technical performances. In particular, the climate change impact potential of the incineration of 1 tonne of waste ranges from a benefit of ?58 kg CO2-eq to a relatively large burden of 408 kg CO2-eq, with 294 kg CO2-eq as the average impact. Two main plant-specific parameters drive the impact potentials regarding the 5 non-toxic impact categories under study: the energy recovery and delivery rate and the NOx process-specific emissions. The variability of the impact potentials as a function of incinerator characteristics therefore calls for the use of site-specific data when required by the LCA goal and scope definition phase, in particular when the study focuses on a specific incinerator or on a local waste management plan, and when these data are available.  相似文献

Abstract

The objective of the present study was to assess the potential interactive effects of two Great Lakes chemical contaminants, hexachlorobenzene (HCB) and mercury (HgCl2). Groups of 10 female Sprague‐Dawley rats were administered by gavage single doses of HCB (400, 600 mg/kg b.w. in corn oil), HgCl2 (10.0, 12.5 mg/kg b.w. aqueous) or combinations of both followed by observation for clinical signs of toxicity for 14 days. Five animals from treatment groups died before the termination of the study; one animal each in 600 mg HCB, 400 mg HCB + 10 mg HgCl2, and 600 mg HCB + 10 mg HgCl2, and two animals in 600 mg HCB + 12.5 mg HgCl2. The surviving animals were necropsied at the termination of the study, and hematological, clinical chemistry, histopathological and tissue residue analyses were performed. Relative liver weights were increased in both low and high dose groups of HCB but not in animals treated with HgCl2 alone. Co‐administration of HgCl2 did not alter the HCB effects on the liver weight of the animals. Serum cholesterol levels were increased in all the groups receiving HCB but not HgCl2. No interactive effects on other serum parameters were seen in animals administered with both chemicals. Mild to moderate morphological changes occurred in the liver, thyroid, thymus, ovary and bone marrow of rats exposed to HCB or HCB + HgCl2, and in the kidney of HgCl2 or HgCl2 + HCB treated animals. More severe histological changes occurred in the groups receiving both chemicals. The histological effects appeared to be additive. It was concluded that co‐administration with HCB and HgCl2 resulted in additive effects in some of the endpoints measured but no synergism or antagonism was observed.  相似文献

Abstract

Lake Ontario coho salmon were known to contain a mixture of chemical contaminants. A previous study demonstrated that rats fed the Lake Ontario fish‐supplemented diet for 28 days exhibited mild biochemical and histological changes. The purpose of the present study was to determine the effects due to a longer term of exposure and the reversibility of these effects. Growth rate and food consumption were not affected by feeding the animals with Lake Ontario or Pacific fish‐supplemented diets for 13 weeks. No deaths were observed. Decreased spleen weights were observed in groups of males fed 1.45%, 5.8% Lake Ontario and 2.9% Pacific diet. After a 13 week recovery the spleen weights returned to normal. Decreased serum potassium was observed in male rats fed 2.9% Lake Ontario diet, and all levels of Pacific diet for 13 weeks, and was not evident following maintenance on normal diet. Serum glucose was not affected by the 13‐week period of treatment, however; a reduction in this parameter occurred in male rats fed the two highest doses of Lake Ontario diet and all doses of Pacific diet following the 13‐week recovery period. Minor hematological changes occurred only in the male rats fed either Lake Ontario or Pacific diet following a 13 week recovery period and included reduced marrow myeloid cells and myeloid/erythroid ratio. Hepatic microsomal ethoxyresorufin deethylase activity was significantly increased in rats ingesting Lake Ontario diet. Mild histological changes occurred in the liver and thyroid of the treated males, and in the liver and kidney of the treated females. These changes were attributed to the chemical residues and/or the fish diet. Data presented here indicated that the Lake Ontario fish‐supplemented diet can cause mild biochemical, hematological and histological changes but most of these were reversible when exposure was terminated.  相似文献

From 1988 to 2003, the St. Lawrence Action Plan, a Canada – Quebec cooperation agreement on the St. Lawrence River, helped to mobilize a still-growing number of stakeholders in the conservation and protection of this great river and generated tangible results in a number of areas of intervention. The successes enjoyed in the area of industrial, agricultural and urban clean-up, in protecting plant and animal species and their habitats, and in acquiring new knowledge on the state and trends of the ecosystem reflect the sustained efforts of government and non-government partners over the course of those 15 years. This study employed mechanism-specific in vitro bioassays to help characterize the occurrence and distribution of dioxin-like and estrogenic contaminants in sediment from Yeongil Bay, Korea. Approximately 85% of the sediments tested induced significant dioxin-like activity in the H4IIE-luc bioassay, while approximately 50% induced significant estrogenic activity in the MVLN bioassay. Instrumentally-derived estimates of 2,3,7,8-tetrachlorodibenzo-p-dioxin and 17beta-estradiol equivalents tended to underestimate the magnitude of response observed in the bioassays, suggesting that compounds detected by chemical analysis did not account for all the activity associated with Yeongil Bay sediments, or that non-additive interactions were occurring. The greatest dioxin-like and estrogenic activity was associated with the mid-polarity Florisil fractions (F2) expected to contain polycyclic aromatic hydrocarbons (PAHs) as well as chlorinated dioxins and furans. As in previous studies of Korean coastal sediment, more polar fractions (F3) generated more modest responses both in terms of magnitude and the number of samples responding.  相似文献

4-Nonylphenol (NP) has been shown to elicit estrogenic responses both in vivo and in vitro. The mechanism by which NP exerts estrogenic and other endocrine-modulating effects in vivo remains unclear, however. The goal of this study was to evaluate the ability of NP to elicit estrogenic responses through indirect mechanisms of action involving the modulation of endogenous steroid hormone concentrations. Sexually mature male common carp (Cyprinus carpio) were exposed to aqueous NP concentrations ranging from <0.05 to 5.4 microg NP/l for 28-31 d. Approximately 0.5-3.5 ppm of NP was detected in pooled plasma samples or tissue samples from the carp studied. NP exposure did not significantly increase plasma concentrations of 17beta-estradiol (E2), testosterone (T) or vitellogenin (VTG). Excluding outliers, plasma E2 concentrations ranged from <175 to 700 pg E2/ml. T concentrations ranged from 940 to 24,700 pg T/ml plasma. The greatest VTG concentration detected was 52 microg/ml. One-third of the plasma samples tested contained <1 microg VTG/ml. Overall, the results of this study did not support the hypothesis that exposure to waterborne NP can modulate concentrations of steroid hormones in the plasma of sexually mature male carp. The results did, however, raise a number of questions regarding the utility of estradiol equivalent (EEQ) estimates as a means of predicting in vivo effects of estrogenic substances. Furthermore, they provide information regarding the concentrations and variability of E2, T, and VTG in the plasma of sexually mature male carp, which may aid in design and interpretation of future studies.  相似文献

A teratological assessment of four trihalomethanes in the rat   总被引:7,自引:0,他引:7  
Four trihalomethanes were administered by gavage to Sprague-Dawley rats from day 6 to day 15 of gestation. Chloroform (Ch) was administered at levels of 100, 200 and 400 mg/kg and bromoform (Br), bromodichloromethane (BDCM) and chlorodibromomethane (CDBM) were administered at levels of 50, 100 or 200 mg/kg/day. A separate control was used for each compound. Maternal weight gain was depressed in all groups receiving Ch and at the highest dose levels of BDCM and CDBM. Ch administration caused decreased maternal hemoglobin and hematocrit values at all dose levels and also produced increased serum inorganic phosphorus and cholesterol at the highest dose. Liver enlargement was observed at all dose levels of Ch but in no other treatment groups. Evidence of a fetotoxic response was observed with Ch, CDBM and Br but not BDCM. No dose-related histopathological changes were observed in either mothers or fetuses as a result of treatment. None of the chemicals tested produced any teratogenic effects.  相似文献
