Abstract

Microtox and Ames bioassays were employed to assess acute toxicity and mutagenicity of water soluble components of class–fractionated oils extracted from one creosote–and four petroleum–contaminated soils. Microtox results revealed that potential acute toxicity resides mainly in the polar class fractions at three sites and indicated potential synergistic and antagonistic effects between compounds in the total extracts at two sites. Ames Salmonella/microsome testing indicated that the polyaromatic fractions at two sites exhibit weak mutagenicity with enzymatic activation, while the polar fractions at two sites are weakly mutagenic without enzyme activation. Further chemical characterization of the polar and polyaromatic fractions is required to fully assess the potential of health and ecological risks at the creosote–and petroleum–contaminated sites exhibiting these toxic responses.  相似文献

An extended analytical characterisation of weathered hydrocarbons isolated from historically contaminated sites in Alberta is presented. The characterisation of soil extracts, chromatographically separated into component classes, by GC-simulated distillation and nitrous oxide (N2O) chemical ionisation (CI) GC-MS provides new insights into the composition of the residual oil at these sites, the principal partition medium for risk critical components. The combined polar and asphaltene content of representative soil extracts ranged from 40% w/w to 70% w/w of the oils encountered. An abundance of C14-C22 2-4 ring alicyclics, alkylbenzenes and benzocycloparaffins is prevalent within the saturate fractions of site soils. Implications for the partitioning of risk critical compounds at sites with weathered hydrocarbons and the practical application of bioremediation technologies are discussed.  相似文献

A broad range of organic compounds are known to exist in drinking water sources and serve as precursors of disinfection byproducts(DBPs).Epidemiological findings of an association of increased risk of bladder cancer with the consumption of chlorinated water has resulted in health concerns about DBPs.Peptides are thought to be an important category of DBP precursors in water.However,little is known about the actual presence of peptides and their DBPs in drinking water because of their high sample complexity and low concentrations.To address this challenge and identify peptides and non-chlorinated/chlorinated peptide DBPs from large sets of organic compounds in water,we developed a novel high throughput analysis strategy,which integrated multiple solid phase extraction(SPE),high performance liquid chromatography(HPLC)separation,and non-target identification using precursor ion exclusion(PIE)high resolution mass spectrometry(MS).After MS analysis,structures of candidate compounds,particularly peptides,were obtained by searching against the Human Metabolome Database(HMDB).Using this strategy,we successfully detected 625 peptides(out of 17,205 putative compounds)and 617 peptides(out of 13,297)respectively in source and finished water samples.The source and finished water samples had 501 peptides and amino acids in common.The remaining 116 peptides and amino acids were unique to the finished water.From a subset of 30 putative compounds for which standards were available,25 were confirmed using HPLC-MS analysis.By analyzing the peptides identified in source and finished water,we successfully confirmed three disinfection reaction pathways that convert peptides into toxic DBPs.  相似文献

Millions of people die every year around the world from diarrheal diseases much of which is caused by contaminated drinking water. By contrast, drinking water safety is largely taken for granted by many citizens of affluent nations. The ability to drink water that is delivered into households without fear of becoming ill may be one of the key defining characteristics of developed nations in relation to the majority of the world. Yet there is well-documented evidence that disease outbreaks remain a risk that could be better managed and prevented even in affluent nations. A detailed retrospective analysis of more than 70 case studies of disease outbreaks in 15 affluent nations over the past 30 years provides the basis for much of our discussion [Hrudey, S.E. and Hrudey, E.J. Safe Drinking Water--Lessons from Recent Outbreaks in Affluent Nations. London, UK: IWA Publishing; 2004.]. The insights provided can assist in developing a better understanding within the water industry of the causes of drinking water disease outbreaks, so that more effective preventive measures can be adopted by water systems that are vulnerable. Residents of affluent nations are remarkably lucky to have high-quality, safe drinking water supplies that most residents of modem cities enjoy, particularly when considered in contrast to the toll of death and misery that unsafe drinking water causes for most of the world's population. Some may presume that drinking-water disease outbreaks are a thing of the past, but complacency can easily arise. A review of drinking water outbreaks in developed countries over the past 3 decades reveals some of the reasons why drinking water outbreaks keep occurring when society clearly has the means to prevent them. Prevention of future outbreaks does not demand perfection, only a commitment to learn from past mistakes and to act on what has been learned.  相似文献
