Objectives: In this article, we evaluate the sensitivity to cognitive load of 3 versions of the Detection Response Task method (DRT), proposed in ISO Draft Standard DIS-17488.

Methods: We present a user study with 30 participants in which we compared the sensitivity to cognitive load of visual, audio, and tactile DRT in a simulated driving environment. The amount of cognitive load was manipulated with secondary n-back tasks at 2 levels of difficulty (0-back and 1-back). We also explored whether the DRT method is least sensitive to cognitive load when the stimuli and secondary task are of the same modality. For this purpose, we used 3 forms to present the n-back task stimuli: visual, audio, and tactile. Responses to the task were always vocal. The experiment was based on a between-subject design (the DRT modalities) with 2 levels of within-subject design study (modalities and difficulty of the secondary n-back tasks). The participants' primary task in the study was to drive safely, and a second priority was to answer to DRT stimuli and perform secondary tasks.

Results: The results indicate that all 3 versions of the DRT tested were sensitive to detecting the difference in cognitive load between the reference driving period and driving and engaging in the secondary tasks. Only the visual DRT discriminated between the 0-back and 1-back conditions on mean response time. Contrary to expectations, no interaction was observed between DRT modality and the stimuli modality used for presentation of the secondary tasks.

Conclusions: None of the 3 methods of presenting DRT stimuli showed a consistent advantage in sensitivity in differentiating multiple levels of cognitive load if all response times, hit rates, and secondary task performance are considered. If only response time is considered, the visual presentation of the DRT stimulus used in this study showed some advantages. In interpreting these data, it should be noted that the methods of DRT stimulus presentation varied somewhat from the currently proposed draft ISO standard and it is possible that the relative salience level of the visual DRT stimulus influenced the findings. It is further suggested that more than 2 levels of difficulty of the n-back task should be considered for further investigation of the relative sensitivity of different DRT stimuli modalities. Parameters that indicate change in cognitive load (response time, hit rate, task performance) should be analyzed together in assessing the overall impact on the driver and not individually, in order to obtain a fuller insight of the assessed cognitive load.  相似文献

We studied the degradation of carbazole (CAR) and 2,3-dichlorodibenzo-p-dioxin (2,3-DCDD) in soils inoculated with carbazole- and dioxin-degrader Pseudomonas resinovorans strain CA10. By using Tn5-based transposon delivery systems, this bacterium was chromosomally marked with a tandem green fluorescent protein (gfp) gene. Real-time competitive PCR and direct counting using the (gfp) marker were employed to monitor the total number of carbazole 1,9a-dioxygenase gene (carAa) and survival of CA10 cells in the soil and soil slurry microcosms. Bioaugmentation studies indicated that the survival of the marked CA10 cells in soil microcosms was strongly influenced by pH and organic matter. While the number of the marked CA10 cells decreased rapidly in pH 6 with low organic matter, a high cell density was maintained in pH 7.3 with 2.5% organic matters up to 21 days after inoculation. In pH 7.3 soil, the period needed for complete degradation of CAR (100 microg kg(-1)) was markedly shortened from 21 to 7 days by the inoculation with the CA10 cells. Single inoculation of CA10 cells into the soil slurry system of 2,3-DCDD-contaminated soil enhanced the degradation of 2,3-DCDD from 25.0% to 37.0%. In this system, the population density of CA10 cells and the total number of carAa gene were maintained up to 14 days after inoculation. By repeated inoculation (every 2 days) with CA10 cells each at a density of 10(9) CFU g(-1) of soil, almost all of the 2,3-DCDD (1 microg kg(-1)) was degraded within 14 days. Results of these experiments suggest that P. resinovorans strain CA10 may be an important resource for bioremediation of CAR and chlorinated dibenzo-p-dioxin in contaminated soils.  相似文献

Objectives: There are 3 standardized versions of the Detection Response Task (DRT), 2 using visual stimuli (remote DRT and head-mounted DRT) and one using tactile stimuli. In this article, we present a study that proposes and validates a type of auditory signal to be used as DRT stimulus and evaluate the proposed auditory version of this method by comparing it with the standardized visual and tactile version.

Methods: This was a within-subject design study performed in a driving simulator with 24 participants. Each participant performed 8 2-min-long driving sessions in which they had to perform 3 different tasks: driving, answering to DRT stimuli, and performing a cognitive task (n-back task). Presence of additional cognitive load and type of DRT stimuli were defined as independent variables. DRT response times and hit rates, n-back task performance, and pupil size were observed as dependent variables.

Results: Significant changes in pupil size for trials with a cognitive task compared to trials without showed that cognitive load was induced properly. Each DRT version showed a significant increase in response times and a decrease in hit rates for trials with a secondary cognitive task compared to trials without. Similar and significantly better results in differences in response times and hit rates were obtained for the auditory and tactile version compared to the visual version. There were no significant differences in performance rate between the trials without DRT stimuli compared to trials with and among the trials with different DRT stimuli modalities.

Conclusions: The results from this study show that the auditory DRT version, using the signal implementation suggested in this article, is sensitive to the effects of cognitive load on driver's attention and is significantly better than the remote visual and tactile version for auditory–vocal cognitive (n-back) secondary tasks.  相似文献

The mechanisms of plant defence against cadmium toxicity have been studied by short-term exposure of Lemna minor L. (common duckweed) to concentrations of CdCl2 ranging from 0 to 500microM. High accumulation of cadmium was observed (12,320+/-2155microgg(-1) at 500microM CdCl2), which caused a gradual decrease of plant growth, increased lipid peroxidation, and weakened the entire antioxidative defence. Total glutathione concentration decreased significantly; however, the concentration of oxidized glutathione remained stable. The responses of four antioxidant enzymes showed that catalase was the most inhibited after CdCl2 exposure, ascorbate peroxidase and guaiacol peroxidase moderately, and glutathione reductase least. The total antioxidative potential revealed an induced antioxidative network at 0.1microM CdCl2 (137+/-13.2% of the control) and its reduction to only 47.4+/-4.0% of the control at higher cadmium concentrations. The possible application of the examined biomarkers in ecotoxicological research is discussed.  相似文献

Objectives: The detection response task (DRT) is a method for measuring attentional effects of secondary tasks on a driver's cognitive load by measuring response times and hit rates to different types of stimuli as indirect indicators of increased cognitive load. ISO 17488 (International Organization for Standardization 2016) only provides guidelines for the technical implementation and measurement methods for the visual and tactile versions (use of visual and tactile stimuli) of the DRT method. This article presents a study with the goal of finding the most appropriate auditory stimulus for the implementation of an auditory version of the DRT method.

Methods: This article presents the results of an experiment in which responses to 7 different auditory DRT stimuli—varying in frequency—were compared while inducing users' cognitive load with a modified n-back task. The experiment was conducted in a surrogate driving environment and in a within-subject design. Response times, hit rates, and secondary task performances were observed as indicators of increased cognitive load.

Results: Significantly shorter response times were found for the white noise signal compared to single-frequency signals. However, the largest differences in response times, for trials without and with a cognitive task, were found for 4- and 8-kHz single-frequency signals. No significant differences were found for hit rates and secondary task performances between the different stimuli.

Conclusions: Consistent significant differences in response times for all tested stimuli prove that the auditory DRT variant is also sensitive to changes in cognitive load. The mean increase in response times of more than 25% for 4- and 8-kHz signals for trials with a cognitive task compared to trials without one indicates that one of these signals could be used as a potential auditory stimulus for the auditory DRT variant.  相似文献

Goal, Scope and Background

Elevated concentrations of copper in the environment result in accumulation of the metal in plants and cause an increase in reactive oxidative species (ROS). The first response to elevated amounts of ROS is increased levels of enzymatic and non-enzymatic antioxidants that reduce oxidative stress. The aim of our study was to evaluate the early stages of antioxidative responses to the low copper concentrations usually present in moderately polluted environments. In addition, some other parameters were examined to evaluate the effect of copper on plants.

Methods

Duckweed (Lemna minor L.) was exposed to different concentrations of copper sulphate for up to 24 hours. Glutathione concentration and enzymatic activities of catalase, guaiacol peroxidase and glutathione reductase were measured spectrophotometrically. Additionally, delayed and prompt chlorophyll fluorescence was measured by luminometry and fluorometry, respectively. The accumulation of copper in plants exposed for 24 hours to various concentrations of copper sulphate was measured by flame atomic absorption spectrophotometry.

Results

The treatment of plants with copper sulphate resulted in an immediate decrease of the glutathione pool, which was replenished after 24 hours at CuSO4 concentrations lower than 2 μM. Higher CuSO4 concentrations caused a decrease of reduced glutathione. The responses of the antioxidant enzymes glutathione reductase, guaiacol peroxidase and catalase to CuSO4 differed during the first six hours of exposure, but their enzyme activities all increased after 24 hours of exposure. All these enzymes displayed biphasic activity curves with maximum values between 0.5 μM and 1 μM CuSO4. The response of guaiacol peroxidase was the most pronounced and statistically significantly specific and that of catalase the least. Delayed chlorophyll fluorescence decreased after exposure to 1 μM CuSO4, but no significant effect on maximum quantum yield of photosystem II (Fv/Fm) was observed. L. minor accumulated relatively high concentrations of copper. The accumulation rate was higher at lower concentrations of copper in the test medium (up to 2 μM CuSO4) than at concentrations above 2 μM CuSO4.

Discussion

One of the most pronounced antioxidative responses to copper exposure was modified levels of oxidized and reduced forms of glutathione. The decrease of the glutathione pool is most probably coupled with induced production of phytochelatins. Antioxidative enzymes showed the biphasic enzyme activity characteristic of stress response. Guaiacol peroxidase exhibited the greatest significant increase of activity, even at higher CuSO4 concentrations at which the activity of catalase and glutathione reductase dropped. The intensity of delayed chlorophyll fluorescence decreased, indicating reduced photosynthesis of plants under stress. All the measured parameters showed that plants respond to even low copper concentrations very soon after exposure. The accumulation rate of copper in duckweed tissues indicates that L. minor is an accumulator species.

Conclusions

The synchronized and prompt inducibility of antioxidants indicates their involvement in a general plant defence strategy for coping with metal-induced oxidative stress. Glutathione concentration and guaiacol peroxidase activity were found to be the most sensitive of the early indicators of exposure to copper concentrations present in polluted water bodies.

Recommendation and Perspectives

The experimental design of the present study allowed us to compare the sensitivity of various methods and parameters for detecting plant responses to heavy metal-induced oxidative stress. The level of glutathione and the enzyme activities of guaiacol peroxidase and glutathione reductase could be used as a rapidly determined early warning system in toxicity studies.
