The availability of genomic data for an increasing number of species makes it possible to incorporate evolutionary processes into conservation plans. Recent studies show how genetic data can inform spatial conservation prioritization (SCP), but they focus on metrics of diversity and distinctness derived primarily from neutral genetic data sets. Identifying adaptive genetic markers can provide important information regarding the capacity for populations to adapt to environmental change. Yet, the effect of including metrics based on adaptive genomic data into SCP in comparison to more widely used neutral genetic metrics has not been explored. We used existing genomic data on a commercially exploited species, the giant California sea cucumber (Parastichopus californicus), to perform SCP for the coastal region of British Columbia (BC), Canada. Using a RAD-seq data set for 717 P. californicus individuals across 24 sampling locations, we identified putatively adaptive (i.e., candidate) single nucleotide polymorphisms (SNPs) based on genotype–environment associations with seafloor temperature. We calculated various metrics for both neutral and candidate SNPs and compared SCP outcomes with independent metrics and combinations of metrics. Priority areas varied depending on whether neutral or candidate SNPs were used and on the specific metric used. For example, targeting sites with a high frequency of warm-temperature-associated alleles to support persistence under future warming prioritized areas in the southern coastal region. In contrast, targeting sites with high expected heterozygosity at candidate loci to support persistence under future environmental uncertainty prioritized areas in the north. When combining metrics, all scenarios generated intermediate solutions, protecting sites that span latitudinal and thermal gradients. Our results demonstrate that distinguishing between neutral and adaptive markers can affect conservation solutions and emphasize the importance of defining objectives when choosing among various genomic metrics for SCP.  相似文献

Increases in the extent and severity of spruce budworm (Choristoneura fumiferana Clem.) outbreaks over the last century are thought to be the result of changes in forest structure due to forest management. A corollary of this hypothesis is that manipulations of forest structure and composition can be used to reduce future forest vulnerability. However, to what extent historical forest management has influenced current spatial patterns of spruce budworm host species is unknown. To identify landscape-scale spatial legacies of forest management in patterns of spruce budworm host species (i.e., Abies balsamea and Picea spp.), we analyzed remotely sensed forest data from the Border Lakes landscape of northern Minnesota and northwestern Ontario. Our study area contains three regions with different management histories: (1) fine-scale logging patterns in Minnesota, (2) coarse-scale logging patterns in Ontario, and (3) very limited logging history in the Boundary Waters Canoe Area and adjacent Quetico Provincial Park. We analyzed forest basal-area data using wavelets and null models to identify: (1) at which scales forest basal area is structured, (2) where those scales of pattern are significantly present, and (3) whether regions of local significance correspond to regional boundaries that separate the study area. Results indicate that spatial patterns in host basal area are created by nonstationary processes and that these processes are further constrained by lakes and wetlands. Wavelet analysis combined with significance testing revealed a bimodal distribution of scale-specific wavelet variance and separate zones of host species basal area that partially correspond with regional boundaries, particularly between Minnesota and the Wilderness region. This research represents one of the first comparisons of forest spatial structure in this region across an international border and presents a novel method of two-dimensional wavelet analysis that can be used to identify significant scale-specific structure in spatial data.  相似文献

The society demands safer products with a better ecological profile. Regulatory criteria have been developed to prevent risks for human health and the environment, for example, within the framework of the European regulation REACH (Regulation (EC) No 1907, 2006). This has driven industry to consider the development of high throughput screening methodologies for assessing chemical biodegradability. These new screening methodologies must be scalable for miniaturisation, reproducible and as reliable as existing procedures for enhanced biodegradability assessment. Here, we evaluate two alternative systems that can be scaled for high throughput screening and conveniently miniaturised to limit costs in comparison with traditional testing. These systems are based on two dyes as follows: an invasive fluorescent dyes that serves as a cellular activity marker (a resazurin-like dye reagent) and a noninvasive fluorescent oxygen optosensor dye (an optical sensor). The advantages and limitations of these platforms for biodegradability assessment are presented. Our results confirm the feasibility of these systems for evaluating and screening chemicals for ready biodegradability. The optosensor is a miniaturised version of a component already used in traditional ready biodegradability testing, whereas the resazurin dye offers an interesting new screening mechanism for chemical concentrations greater than 10 mg/l that are not amenable to traditional closed bottle tests. The use of these approaches allows generalisation of high throughput screening methodologies to meet the need of developing new compounds with a favourable ecological profile and also assessment for regulatory purpose.  相似文献

Recent ecosystem and fire management research aims to quantify, model and understand historical fire disturbances focusing on their frequency, size and distribution. Less attention, however, has been paid to fire boundaries and their location on the landscape. Our study presents a spatial method to quantify the location, pattern and persistence of historical fire boundaries using tree ring fire scar data in the lower Stein watershed (British Columbia). Data from Pinus ponderosa and Pseudotsuga menziesii collected in 35 one-hectare plots over a 412-hectare study area were analyzed for the period between 1879 and 1947 using local spatial statistics and boundary detection techniques. Results of the analysis using local spatial statistic Moran’s I showed significant clustering of boundaries near topographic breaks. To determine and test whether fire boundaries between plots were persistent, we used boundary detection methods and developed a spatially restricted randomization test. The results revealed that out of 86 possible boundary links, 8 were significantly persistent (P < 0.025) while another 8 were significantly rare (P < 0.025). These spatial methods can help determine the historical spatial configuration of persistent boundaries and can be used to maintain natural forest dynamics.

Graph theory, network theory, and circuit theory are increasingly being used to quantify multiple aspects of habitat connectivity and protected areas. There has been an explosive proliferation of network (connectivity) measures, resulting in over 60 measures for ecologists to now choose from. Conceptual clarification on the ecological meaning of these network measures and their interrelationships is overdue. We present a framework that categorizes network measures based on the connectivity property that they quantify (i.e., route-specific flux, route redundancy, route vulnerability, and connected habitat area) and the structural level of the habitat network to which they apply. The framework reveals a lack of network measures in the categories of "route-specific flux among neighboring habitat patches" and "route redundancy at the level of network components." We propose that network motif and path redundancy measures can be developed to fill the gaps in these categories. The value of this framework lies in its ability to inform the selection and application of network measures. Ultimately, it will allow a better comparison among graph, network, and circuit analyses, which will improve the design and management of connected landscapes.  相似文献
