Endotoxin exposure is associated with wheeze and asthma morbidity, while early life exposure may reduce risk of allergy and asthma. Unfortunately, it is difficult to compare endotoxin results from different laboratories and environments. We undertook this study to determine if lipopolysaccharide (LPS) extraction efficiency could account for differences among laboratories. We generated and collected aerosols from chicken and swine barns, and corn processing. We randomly allocated side-by-side filter samples to five laboratories for Limulus assay of endotoxin. Lyophilized aliquots of filter extracts were analyzed for 3-hydroxy fatty acids (3-OHFAs) as a marker of LPS using gas chromatography-mass spectrometry. There were significant differences in endotoxin assay and GC-MS (LPS) results between laboratories for all dust types (p < 0.01). Patterns of differences between labs varied by dust type. Relationships between assay and GC/MS results also depended on dust type. The percentages of individual 3-OHFA chain lengths varied across labs (p < 0.0001) suggesting that each lab recovered a different fraction of the LPS available. The presence of large amounts of particle associated LPS and absence of a freezing thawing cycle were associated with lower correlations between LPS and bioactivity, consistent with an absence of Limulus response to cell-bound endotoxin. These data suggest that extraction methods affect endotoxin measurements. The LAL methods may be most suitable when comparing exposures within similar environments; GC-MS offers additional information helpful in optimizing sample treatment and extraction. GC-MS may be of use when comparing across heterogeneous environments and should be considered for inclusion in future studies of human health outcomes.  相似文献

Bio-economic farm models are tools to evaluate ex-post or to assess ex-ante the impact of policy and technology change on agriculture, economics and environment. Recently, various BEFMs have been developed, often for one purpose or location, but hardly any of these models are re-used later for other purposes or locations. The Farm System Simulator (FSSIM) provides a generic framework enabling the application of BEFMs under various situations and for different purposes (generating supply response functions and detailed regional or farm type assessments). FSSIM is set up as a component-based framework with components representing farmer objectives, risk, calibration, policies, current activities, alternative activities and different types of activities (e.g., annual and perennial cropping and livestock). The generic nature of FSSIM is evaluated using five criteria by examining its applications. FSSIM has been applied for different climate zones and soil types (criterion 1) and to a range of different farm types (criterion 2) with different specializations, intensities and sizes. In most applications FSSIM has been used to assess the effects of policy changes and in two applications to assess the impact of technological innovations (criterion 3). In the various applications, different data sources, level of detail (e.g., criterion 4) and model configurations have been used. FSSIM has been linked to an economic and several biophysical models (criterion 5). The model is available for applications to other conditions and research issues, and it is open to be further tested and to be extended with new components, indicators or linkages to other models.  相似文献

Geomorphologically appropriate rehabilitation measures were proposed to enhance the in-stream environment of the lowland River Idle, north Nottinghamshire, UK. However, the River Idle has multi-functional management requirements including those of flood defence so environmental enhancement must be pursued without significantly increasing the flood risk. Hydraulic testing of rehabilitation proposals is complicated because of the stringent assumptions about flow and morphology in ‘traditional’ hydraulic models. While new generation two- and three-dimension hydraulic models may overcome some of these problems, they are extremely data intensive, require advanced modelling capabilities and are, therefore, very expensive to apply. Also, they do not yet predict morphology-flow interactions adequately. As an alternative, several simple hydraulic models were applied to test the rehabilitation proposals, based on a fitness-for-purpose criterion.BENDFLOW was applied to fine tune the optimal siting of measures and to estimate the additional near-bank scour generated by proposed bend re-profiling. HMODEL2 and the FCFA method were used to test the impact on local channel conveyance capacities and HECRAS was applied to simulate the impact of the proposals on regional flood defence. Indicative results from the testing suggested a maximum increase in near-bank scour of 0·15 m in re-profiled bends, a loss of approximately 10% in flood conveyance locally due to deflector installation or reed and tree planting, and a 0·12 m increase in flood stage within the reach for a 15 year flood. The modelling results were acceptable to the management authority as an indication of an acceptable compromise between flood defence and conservation interests, and construction of the measures followed in 1996. It is clearly that it will require the results of post-project monitoring to indicate whether compromises made to the rehabilitation initiatives in order to satisfy flood defence requirements have unduly reduced their environmental enhancement potential but, for assessing the proposed methods, the models are recommended for use other lowland river environments.  相似文献

In post-closure radiological safety assessments of repositories for solid radioactive wastes, transfers of radionuclides to animal products are typically characterised using Transfer Factors (TFs), defined as the ratio of the concentration of the radionuclide in the animal product of interest to the rate of intake in diet. Such transfer factors can be measured directly in experimental studies, but they can also be estimated by use of biokinetic models for uptake and retention of radionuclides in animals. Based on a review of the literature, biokinetic models have been developed for the uptake and retention of iodine, technetium, selenium and uranium. These biokinetic models allow TF values to be estimated for different types of animals and for different animal lifetimes.For each radionuclide considered, reference values and ranges of TF values are estimated. Reclaimed Appalachian surface mined lands have difficulty in sustaining native deciduous forest communities. Establishing prairie communities could increase ecosystem function; however, a native model system does not exist. We evaluated establishment of 15 North American prairie grasses as monocultures on reclaimed mine soil in southeast Ohio in four randomized complete blocks planted May 2005 and 2006. Population density was assessed 30 d after planting (30 DAP) and in October of the planting year (YR1) and second year following planting (YR2) and expressed as percentage of viable seeds sown (PVSS). Canopy cover of nonnative species reestablishing in the plots was measured in 2007. Eastern gamagrass ( L.) population was >50 PVSS in all censuses. Western wheatgrass [ (Rydb.) A. L?ve] was initially 7 PVSS at 30 DAP, but increased to 154 PVSS by YR2 from rhizomes spreading into gaps. Big bluestem ( Vitman) was 7 PVSS at 30 DAP and 4 PVSS at YR2. Blue grama [ (Willd. ex Kunth) Lag. ex Griffiths] and sideoats grama [ (Michx.) Torr.] did not survive past YR1. Gaps left from poor stand establishment were primarily recolonized by nonnative Kentucky bluegrass ( L.) in the 2005 planting and birdsfoot trefoil ( L.) in the 2006 planting, but was least in eastern gamagrass and tall dropseed [ (P. Beauv.) Kunth]. This research demonstrates the potential for increasing diversity and species richness on mine soil habitats with regionally native grasses that could increase functional quality through ecological resilience.  相似文献

Flood risk to the economy, society and the environment reflects the cumulative effects of environmental and socio-economic change over decades. Long-term scenarios are therefore required in order to develop robust and sustainable flood risk management policies. Quantified national-scale flood risk analysis and expert appraisal of the mechanisms causing change in flood risk have been used to assess flood risk in England and Wales over the period 2030–2100. The assessment involved the use of socio-economic and climate change scenarios. The analysis predicts increasing flood risk unless current flood management policies, practices and investment levels are changed—up to 20-fold increase in economic risk by the 2080s in the scenario with highest economic growth. The increase is attributable to a combination of climate change (in particular increasing precipitation and relative sea level rise in parts of the UK) and increasing socio-economic vulnerability, particularly in terms of household/industrial contents and infrastructure vulnerability. The policy implications of these findings are discussed.  相似文献

Safety assessment is a tool which, by means of an iterative procedure, allows the evaluation of the performance of a disposal system and its potential impact on human health and the environment. Radionuclides from a deep geological disposal facility may not reach the surface environment until many tens of thousands of years after closure of the facility. The BIOMASS Programme on BIOsphere Modelling and ASSessment developed Examples of "Reference Biospheres" to illustrate the use of the methodology and to demonstrate how biosphere models can be developed and justified as being fit for purpose. The practical examples are also intended to be useful in their own right. The Example Reference Biosphere 2B presented here involves the consideration of alternative types of geosphere-biosphere interfaces and calculation of doses to members of hypothetical exposure groups arising from a wide range of exposure pathways within agricultural and semi-natural environments, but without allowing for evolution of the corresponding biosphere system. The example presented can be used as a generic analysis in some situations although it was developed around a relatively specific conceptual model. It should be a useful practical example, but the above numerical results are not intended to be understood as prescribed biosphere 'conversion factors'.  相似文献
