A mercury biosensor was constructed by integrating biosensor genetic elements into E. coli JM109 chromosome in a single copy number, using the attP/attB recombination mechanism of λphage. The genetic elements used include a regulatory protein gene (merR) along with operator/promoter (O/P) derived from the mercury resistance operon from pDU1358 plasmid of Serratia marcescens. The expression of reporter gene gfp is also controlled by merR/O/P. Integration of the construct into the chromosome was done to increase the stability and precision of the biosensor. This biosensor could detect Hg(II) ions in the concentration range of 100-1700 nmol/L, and manifest the result as the expression of GFP. The GFP expression was significantly different (P ≤ 0.05) for each concentration of inducing Hg(II) ions in the detection range, which reduces the chances of misinterpretation of results. A model using regression method was also derived for the quantification of the concentration of Hg(II) in water samples. 相似文献

Atrazine is a persistent organic pollutant in the environment which affects not only terrestrial and aquatic biota but also human health. Since its removal from the environment is needed, atrazine biodegradation is achieved in the present study using the bacterium Rhodococcus sp. BCH2 isolated from soil, long-term treated with atrazine. The bacterium was capable of degrading about 75 % atrazine in liquid medium having pH 7 under aerobic and dark condition within 7 days. The degradation ability of the bacterium at various temperatures (20–60 °C), pH (range 3–11), carbon (glucose, fructose, sucrose, starch, lactose, and maltose), and nitrogen (ammonium molybdate, sodium nitrate, potassium nitrate, and urea) sources were studied for triumph optimum atrazine degradation. The results indicate that atrazine degradation at higher concentrations (100 ppm) was pH and temperature dependent. However, glucose and potassium nitrate were optimum carbon and nitrogen source, respectively. Atrazine biodegradation analysis was carried out by using high-performance thin-layer chromatography (HPTLC), Fourier transform infrared spectroscopy (FTIR), and liquid chromatography quadrupole time-of-flight (LC/Q-TOF-MS) techniques. LC/Q-TOF-MS analysis revealed formation of various intermediate metabolites including hydroxyatrazine, N-isopropylammelide, deisopropylhydroxyatrazine, deethylatrazine, deisopropylatrazine, and deisopropyldeethylatrazine which was helpful to propose biochemical degradation pathway of atrazine. Furthermore, the toxicological studies of atrazine and its biodegraded metabolites were executed on earthworm Eisenia foetida as a model organism with respect to enzymatic (SOD and Catalase) antioxidant defense mechanism and lipid peroxidation studies. These results suggest innocuous degradation of atrazine by Rhodococcus sp. BCH2 in nontoxic form. Therefore the Rhodococcus sp.BCH2 could prove a valuable source for the eco-friendly biodegradation of atrazine pesticide. 相似文献

Eighteen sites impacted by abandoned mine drainage (AMD) in Pennsylvania were sampled and measured for pH, acidity, alkalinity, metal ions, and sulfate. This study compared the accuracy of four acidity calculation methods with measured hot peroxide acidity and identified the most accurate calculation method for each site as a function of pH and sulfate concentration. Method E1 was the sum of proton and acidity based on total metal concentrations; method E2 added alkalinity; method E3 also accounted for aluminum speciation and temperature effects; and method E4 accounted for sulfate speciation. To evaluate errors between measured and predicted acidity, the Nash-Sutcliffe efficiency (NSE), the coefficient of determination (R 2), and the root mean square error to standard deviation ratio (RSR) methods were applied. The error evaluation results show that E1, E2, E3, and E4 sites were most accurate at 0, 9, 4, and 5 of the sites, respectively. Sites where E2 was most accurate had pH greater than 4.0 and less than 400 mg/L of sulfate. Sites where E3 was most accurate had pH greater than 4.0 and sulfate greater than 400 mg/L with two exceptions. Sites where E4 was most accurate had pH less than 4.0 and more than 400 mg/L sulfate with one exception. The results indicate that acidity in AMD-affected streams can be accurately predicted by using pH, alkalinity, sulfate, Fe(II), Mn(II), and Al(III) concentrations in one or more of the identified equations, and that the appropriate equation for prediction can be selected based on pH and sulfate concentration. 相似文献

urban and industrial areas. Heavy metal components associated with air pollutants have far reaching effects with respect to
economic and ecological importance of pollens. The pollens are male reproductive organs of the plant and travel through air
from flower to flower for pollination purpose. During this period they are exposed to air pollutants. Present investigation
thus pertains to study of effect of air pollutants on pollens especially biosorption and bioaccumulation of heavy metals.
The pollens of three commonly occurring plants namely Cassia siamea, Cyperus rotundus, Kigelia pinnata have been studied from the NH-6 of Nagpur city, India. The pollens exposed to polluted air showed the presence of higher
concentrations of Ca, Al and Fe as compared to unexposed pollens. Higher concentration of these metals was observed in Cyperus rotundus followed by Cassia siamea and Kigelia pinnata. These results indicate that pollens act as good indicator of air pollution giving results in short time of exposure of 5–10 h.
Apart from this, it is also reported that some of these metals play crucial role in the metabolic activity in pollens for
example Calcium is necessary for growth of pollen tube and other metabolic activities in pollens. The presence of these metals
in pollens may also enhance the allergenicity of the pollens. Similarly accumulation of heavy metals may also deteriorate
the quality of pollen for their economical use. The viability of pollen is also affected by these pollutants in sensitive

The electrocoagulation (EC) process is an electrochemical means of introducing coagulants and removing suspended solids, colloidal material, and metals, as well as other dissolved solids from water and wastewaters. The EC process has been successfully employed in removing pollutants, pesticides, and radionuclides. This process also removes harmful microorganisms. More often during EC operation, direct current is applied and electrode plates are sacrificed (dissolved into solution). The dissolution causes an increased metal concentration in the solution that finally precipitates as oxide precipitates. Due to improved process design and material of construction, the EC process is being widely accepted over other physicochemical processes. Presently, this process has gained attention due to its ability to treat large volume and for its low cost. Avifaunal study was carried out in Panipat refinery area before and after commissioning. A total of 63 birds were observed during the study, of which 39 and 49 birds were present during 1992 and 2001 respectively. Of these, 25 birds were common during both the study period. Blue rock pigeon was most dominant during 1992 whereas house sparrow was dominant during 2001. Species diversity index was found to be 5.26 and 6.64 in respective years. Jaccard's and Sorenson similarity indices were 0.40 and 0.57 respectively. Majority of birds were insectivorous in habits followed by granivorous birds in 1992 and piscivorous birds in 2001. The study reveals that less diversity was observed in 1992 when Panipat refinery was commissioned, but more birds especially aquatic ones were observed after the commissioning of the refinery. This has been attributed to increase in birds habitats due to developments of wetlands in and outside refinery, development of green belt around refinery and increase in green cover of area due to boosting of agroforestry and social forestry. 相似文献

The effects of pesticides on blood characteristics and histological changes in erythrocytes of the fish species Cyprinus carpio and Puntius ticto were studied. The fishes were exposed to sub lethal concentrations of different chlorinated pesticides namely aldrin, dieldrin, DDT, BHC and chlordane for 10, 20 and 30 d in continuous flow-through test. The LCso values were calculated based on acute toxicity tests and the sublethal doses were arrived at for chronic bioassay studies. Results showed an increase in haemoglobin content of both Cyprinus carpio and Puntius ticto in case of aldrin and dieldrin. Haemoglobin content reduced from an initial 13 g/100 ml to 8.07 and 10.15 g/100 ml in case of Cyprinus at the end of ten days exposure to aldrin and dieldrin respectively, and gradually increased to 8.7 g/100 ml and 10.15 g/100 ml after 20 d of exposure. The haemoglobin content after 30 d exposure to aldrin and dieldrin was 10.15 g/100 ml and 11.6 g/100 ml respectively. In case of Puntius ticto, the haemoglobin content in control fishes recorded was 12.8 g/100 ml while in case of fish exposed to aldrin, the haemoglobin content reduced initially on ten days exposure to 10.15 g/100 ml and increased to 11.6 g/100 ml and 13.0 g/100 ml during twenty days and thirty days exposure respectively. This trend was also observed with dieldrin in both the fishes studied. Red blood cells were also counted in case of all the pesticides and exposure periods with respect to Cyprinus carpio and Puntius ticto. Irrespective of the species and pesticide, the RBC counts uniformly showed decreasing trend with the increase in exposure period, while packed cell volume, PCV(%) showed increasing trend with respect to increase in exposure period in case of aldrin and dieldrin in both the fishes. But DDT, BHC and chlordane showed decreasing trend in PCV(%) values with increasing periods of exposure. 相似文献
