Amniotic fluid (AF) levels of 17-hydroxyprogesterone (17OHP) and testosterone (T) were determined at 16–17 weeks in 17 pregnancies at risk for CAH and results compared to 75 normal controls. The fetus was predicted to be unaffected in 12 cases on the findings of normal AF levels of both 17OHP and T and the latter allowed a correct prediction of fetal sex in all instances. HLA typing confirmed normality in 12 cases revealing 5 carriers, 5 homozygous normal and 2 indeterminate. Steroid levels of the 2 groups were similar. Three fetuses were predicted to be CAH affected on unambiguously high levels of 17OHP and T (in female only). HLA typing was in agreement, and the diagnosis was confirmed in 2 abortuses and a female newborn by physical and hormonal studies. In the last 2 cases AF levels of OHP and T were normal but HLA (A/B/C) genotypes were identical to the CAH affected siblings. Normal physical and hormonal findings in the 2 aborted fetuses would exclude the possibility of an in utero virilizing form of CAH. The discrepancy could be explained on the basis that the fetuses had an allelic form of 21-hydroxylase deficiency or on the basis of recombination (not fully tested). It is concluded that a fully informative prenatal diagnosis of CAH should not rely entirely on HLA typing but on hormonal studies.  相似文献

We studied the validity of mid-trimester amniotic fluid testosterone and FSH in the diagnosis of fetal sex in abnormal fetuses and in twins. It was found that the testosterone/FSH ratio, but not the level of the individual hormones, was an excellent criterion for fetal sex diagnosis. In a control group of 32 female and 34 male fetuses, the testosterone/FSH ratio was diagnostic. In two cases the values were indeterminate and fetal sex could not be predicted, but there were no false diagnoses. The abnormalities of the fetuses were diagnosed by karyotype analyses or raised levels of α-fetoprotein. It was found that neither autosomal trisomies nor X-chromosome aneuploidy or neural tube defect interfered with the diagnostic value of the hormonal approach. The twin study included 14 pairs, three of whom were discordant for sex. In all twins the fetal sex diagnosis was appropriate. In co-twins of the same sex the hormone levels were very close to each other.  相似文献

Abstract

The objective of this study was to determine the effect of either 2.5 mg/kg Body Weight or 5 mg/kg Body Weight (BW) doses of isoflavones on semen quality, testosterone levels, lipid peroxidation and semen biochemistry of male New Zealand White rabbits. Animals were given both 2.5 mg/kg BW and 5 mg/kg BW doses of isoflavones. The tested doses were given to rabbits orally every other day for 13 weeks. Treatment with isoflavones caused an increase (p < 0.05) in libido (by decreasing the reaction time), sperm concentration, sperm motility (%), total motile sperm per ejaculate (TMS), packed sperm volume (PSV), total functional sperm fraction (TFSF), total sperm output, initial fructose concentration and normal sperm, while dead sperm was reduced compared to control animals. On the other hand, ejaculate volume, initial hydrogen ion concentration (pH) and plasma testosterone levels did not change in treated animals with both doses of isoflavones as compared to control. Concentrations of thiobarbituric acid-reactive substances (TBARS), total lipids, and low density lipoprotein were significantly (p < 0.05) reduced in seminal plasma of rabbits treated with either 2.5 mg/kg BW or 5 mg/kg BW doses of isoflavones. While, the activities of glutathione S-transferase (GST), lactate dehydrogenase (LDH), aspartate aminotransferase (AST), alanine aminotransferase (ALT), acid phosphatase (AcP), and alkaline phosphatase (AlP) were significantly (p < 0.05) increased in seminal plasma of treated animals. Also, total cholesterol, percentage cholesterol (out of total lipids), and high density lipoprotein were significantly (p < 0.05) increased, while triglyceride did not change in seminal plasma of treated animals. Supplementation at either level of isoflavones did not cause changes in live body weight (LBW), dry matter intake (DMI), and relative weights of testes and epididymis. The present results showed that either 2.5 mg/kg BW or 5 mg/kg BW doses of isoflavones caused an improvement of some semen characteristics and did not have negative effects on male fertility.  相似文献

Previous studies have suggested that testosterone (T) profiles of male birds reflect a trade-off between mate attraction behaviours (requiring high T levels) and parental care activities (requiring low T levels). In this study, we experimentally elevated T levels of monogamous males in the facultatively polygynous European starling (Sturnus vulgaris), and compared mate attraction and paternal behaviour of T-treated males with those of controls (C-males). T-males significantly reduced their participation in incubation and fed nestlings significantly less often than C-males. Females paired to T-treated males did not compensate for their mate’s lower paternal effort. The observed reduction in a male’s investment in incubating the eggs was accompanied by an increased investment in typical female-attracting behaviours: T-males spent a significantly higher proportion of their time singing to attract additional females. They also occupied more additional nestboxes than C-males, although the differences just failed to be significant, and carried significantly more green nesting materials into an additional nestbox (a behaviour previously shown to serve a courtship function). T-males also behaved significantly more aggressively than C-males. During the nestling period, the frequency of mate-attracting behaviours by T-treated and control males no longer differed significantly. Despite the reduced paternal effort by T-males and the lack of compensation behaviour by females, hatching and breeding success did not differ significantly between T- and C-pairs. Fitness tradeoffs are difficult to examine because many fitness variables are correlated and vary in the same direction. Phenotypic manipulation circumvents many of these difficulties, and here we used this technique to examine mechanisms for tradeoffs between increased aggression (territorial defense) and survivorship. The behavioral phenotype of male mountain spiny lizards (Sceloporus jarrovi) was manipulated with testosterone to increase territorial defense, a sexually selected trait. We previously demonstrated that increased territorial defense results in a decrease in survival caused by a lower ratio of energy intake to energy expenditure. Here we measured energy consumption of increased territorial aggression using the doubly labeled water technique in the field and compared males with and without testosterone implants (Fig. 1). In a supplementary study we measured standard metabolic rate using captive lizards given similar testosterone implants to examine if an increase in energy expenditure was a result of only an increase in standard metabolic rate (Fig. 3). Our results indicated that a primary contribution to tradeoffs between increased territorial defense and survivorship could be made by a 31% increase in energy expenditure in the field that is not due to an increase in standard metabolic rate.  相似文献

For males of socially polygynous avian species like the spotless starling, there may exist a trade-off between investing in paternal care and controlling several nests. To determine how the intensity of paternal care affects reproductive success per brood sired or expressed as the total number of young raised in all nests controlled by the same male, it is necessary to manipulate paternal care. Testosterone (T) has been shown to depress the tendency for males to care for their young, and induces them to acquire more mates. The effects of paternal care on reproductive success were studied by treating certain male starlings with exogenous T and others with the antiandrogen cyproterone acetate (CA), and comparing the parental behavior of T- and CA-males throughout the breeding season with that of controls. CA-males fed their chicks more during the first week after hatching than T-males, with controls feeding at intermediate rates, both on a per nest basis and as total effort for all nests controlled by the same male. Paternal feeding rates during the first week of chick life had a significant positive effect on the number of fledged young. The hormone treatment significantly affected the number of chicks raised per nest, CA-males having a higher breeding success per nest than T-males, and controls showing intermediate levels of success. There was no significant effect of treatment on total reproductive success attained by males throughout the season. In the polygonous spotless starling, the intensity of paternal care of young affects reproductive success per nest positively but not on a seasonal basis. In the European starling, Sturnus vulgaris, optimal mating systems differ between males and females. Males gain from polygyny, whereas monogamy increases female fitness. The cost of polygyny to females lead to intense female–female competition, and it has previously been shown that the intensity of female aggression during the pre-breeding period can predict the realised mating system. The physiological regulation of such female aggression in starlings is not yet known. This study examines the role of testosterone in mediating aggressive behaviours involved in intra-specific reproductive competition in female starlings. Testosterone levels were experimentally elevated with testosterone implants in females during the pre-laying period. To simulate a situation in which an additional female tried to mate with the focal female’s mate, a caged female was presented close to a nest-site to which the male could attract a secondary female. Testosterone was significantly related to several behaviours involved in female–female interactions. Females with testosterone implants spent significantly more time close to the caged female and produced more song bouts than control females. In contrast, male behaviour was unrelated to the experimental status of the mate. Females mated to males that attracted a secondary female were less aggressive towards the caged female than those that remained monogamously mated. The effect of exogenous testosterone in this study indicates that androgens may mediate social behaviours in female starlings during the breeding season.  相似文献

The deposition of androgens into the yolks of eggs can have long-lasting effects on the growth and development of young birds. It has been proposed that female birds and reptiles deposit yolk androgens according to the differential allocation hypothesis (DAH), which posits the allocation of more resources to offspring sired by more attractive, higher-quality males. We examined deposition patterns of yolk androgens in relation to mate attractiveness in the house finch Carpodacus mexicanus. Contrary to the predictions of the DAH, female house finches deposited significantly more androgens into eggs sired by less attractive males. We propose that, rather than serving as resources, androgens are used as mediators in a compensatory distribution strategy, enabling females to improve the quality of young produced with less attractive males.  相似文献

This study investigated hormonal and demographic processes underlying unimale and multimale mating systems in primates. Reproductive skew and challenge models of male competition provide conflicting predictions of the relationship of male residence to group composition and androgen regulation. These predictions were tested using endocrine and socioecological data from Kenyan vervet monkeys (Cercopithecus aethiops). Serum samples from 57 adult male monkeys, drawn from 19 separate groups and 4 populations, were assayed for testosterone by radioimmunoassay. Male ability to respond to conspecific challenge was assessed by their testosterone response to the capture protocol.Analyses showed that reproductive skew models were useful predictors of intergroup and interpopulation variation in male residence and T profiles. The Limited Control model of male residence was supported by positive correlations of the number of males per group with the number of females without dependent offspring, demonstrating that monopolization potential was a key determinant of male residence. Testosterone concentrations under conditions that elevated serum levels were positively correlated with infanticide risk, supporting the Concession model. Population comparisons provided evidence for increased T responsiveness where groups were predominantly unimale. Unimale populations were from sites with higher rainfall, suggesting that ecological factors contributed to population differences in male residence and T regulation.For species name, we follow the 2000 taxonomy of the IUCN/SSC Primate Specialist Groups workshop (Grubb et al. 2003)  相似文献

Mothers may profoundly affect offspring phenotype and performance by adjusting egg components, including steroid hormones. We studied the effects of elevated prenatal testosterone (T) exposure in the ring-necked pheasant on the expression of a suite of male and female traits, including perinatal response to stress, immune response, growth, and secondary sexual traits. Prenatal T levels were increased by injecting the yolk of unincubated eggs with physiological doses of the hormone. Yolk T injection resulted in a reduced length of male tarsal spurs, a trait which positively predicts male success in intra- and intersexual selection and viability, whereas no direct effect on male wattle characteristics or plumage traits of either sex was observed. Female spur length was also negatively affected by T, but to a lesser extent than in males. In addition, the covariation between male secondary sexual traits, which are reliable quality indicators, differed between T and control males, suggesting that the manipulation may have altered the assessment of overall male quality by other males and females. In conclusion, the negative effects of elevated yolk T on spur length, a trait which positively predicts male fitness, coupled with the lack of effects on growth or other traits in both sexes, provided limited evidence for mothers being subjected to a trade-off between positive and negative consequences of yolk T deposition on offspring traits and suggest that directional selection for reduced yolk T levels may occur in the ring-necked pheasant.  相似文献
