针对食品微生物学的课程考核方式进行了初步研究,采取对照试验的方法,对传统的期末考试方法与“当堂课考核需要掌握的关键点”的新方法进行了比较。食品微生物学课程的理论部分和实验部分采用“当堂课考核需要掌握的关键点”的考试方法后,不仅大大提高了学生学习本课程的积极性,而且还提高了学习成绩,特别是学生对“必需掌握的关键点”掌握得特别牢固。  相似文献

The CO2SINK pilot project at Ketzin is aimed at a better understanding of geological CO2 storage operation in a saline aquifer. The reservoir consists of fluvial deposits with average permeability ranging between 50 and 100 mDarcy. The main focus of CO2SINK is developing and testing of monitoring and verification technologies. All wells, one for injection and two for observation, are equipped with smart casings (sensors behind casing, facing the rocks) containing a Distributed Temperature Sensing (DTS) and electrodes for Electrical Resistivity Tomography (ERT). The in-hole Gas Membrane Sensors (GMS) observed the arrival of tracers and CO2 with high temporal resolution. Geophysical monitoring includes Moving Source Profiling (MSP), Vertical Seismic Profiling (VSP), crosshole, star and 4-D seismic experiments. Numerical models are benchmarked via the monitoring results indicating a sufficient match between observation and prediction, at least for the arrival of CO2 at the first observation well. Downhole samples of brine showed changes in the fluid composition and biocenosis. First monitoring results indicate anisotropic flow of CO2 coinciding with the “on-time” arrival of CO2 at observation well one (Ktzi 200) and the later arrival at observation well two (Ktzi 202). A risk assessment was performed prior to the start of injection. After one year of operations about 18,000 t of CO2 were injected safely.  相似文献

• OBS inhibited the growth of P. stutzeri and destroyed its structure. • OBS was toxic to the aerobic denitrification process of P. stutzeri. • OBS induced the production of ROS in P. stutzeri. • OBS affected the expression of key genes involved in denitrification and SOD. The toxicities of sodium perfluorononyloxy-benzenesulfonate (OBS) to animals and plants are similar to those of perfluorooctane sulfonate. However, the mechanism of its toxicity to aerobic denitrifying bacteria is still unclear. In the present study, the ecotoxicity of OBS on an aerobic denitrifying strain, Pseudomonas stutzeri, was evaluated. The results showed that a dosage of OBS clearly affected the growth and aerobic denitrification of P. stutzeri. When compared with an unamended control, the degradation efficiency of the total nitrogen decreased by 30.13% during exposure to 200 mg/L of OBS, and the complete degradation time of nitrate-nitrogen was delayed by 4 h. The lactate dehydrogenase and superoxide dismutase produced by the bacteria increased with the concentration of OBS, and reactive oxygen species were also detected by confocal laser scanning microscope imaging. Transmission electron microscope imaging revealed chromosome deformation of the cells and damage to cell content; moreover, outer membrane vesicles were secreted from the bacteria, which was the important detoxification mechanism of P. stutzeri to OBS. Expression of the genes involved in aerobic nitrification and oxidative stress were also changed under OBS stress, which further confirmed the toxicity of OBS to P. stutzeri. This study reveals the environmental exposure risk of OBS from the perspective of microorganisms.  相似文献

Characterization of redox conditions in groundwater contaminant plumes   总被引:2,自引:0,他引:2  
Evaluation of redox conditions in groundwater pollution plumes is often a prerequisite for understanding the behaviour of the pollutants in the plume and for selecting remediation approaches. Measuring of redox conditions in pollution plumes is, however, a fairly recent issue and yet relative few cases have been reported. No standardised or generally accepted approach exists. Slow electrode kinetics and the common lack of internal equilibrium of redox processes in pollution plumes make, with a few exceptions, direct electrochemical measurement and rigorous interpretation of redox potentials dubious, if not erroneous. Several other approaches have been used in addressing redox conditions in pollution plumes: redox-sensitive compounds in groundwater samples, hydrogen concentrations in groundwater, concentrations of volatile fatty acids in groundwater, sediment characteristics and microbial tools, such as MPN counts, PLFA biomarkers and redox bioassays. This paper reviews the principles behind the different approaches, summarizes methods used and evaluates the approaches based on the experience from the reported applications.  相似文献

Polyphenols in plants can protect proteins from degradation and improve the efficiency of conversion of plant protein to animal protein (meat and milk), but can this be achieved in a way that is environmentally sustainable, profitable to the livestock farmer and is consistent with aspirations for improved livestock nutrition, health, welfare and product quality? Given that grazed and conserved forage is the bedrock of sustainable ruminant production, this paper attempts to consider what challenges are ahead in terms of enhancing productive efficiency, reducing the polluting footprint of livestock agriculture and alleviating the endemic parasitism that occurs with grazing animals.  相似文献

Greywater from baths, showers and washbasins was collected separately from all other domestic wastewater at a university block of flats with a dual reticulation system and analysed for a range of contaminants including indicator organisms and pathogens. Greywater flow and temperature were also monitored and a diurnal variation was observed. Physical and chemical water quality parameters were similar to previously published data, although measured COD and BOD levels appeared to be lower, possibly due to settlement or biodegradation in the storage tanks. Plate counts and indicator organism concentrations were consistently high suggesting a high level of human bacterial contamination necessitating biological treatment and disinfection if the water is to be used for recycling. However, these high levels of indicator organisms did not correlate to pathogen presence and should not be used as pathogen indicators in greywater. One positive count of Salmonella veltereden was observed as well as low levels of Giardia. Cryptosporidium, Escherichia coli O157:H7, enteroviruses and Legionella were not identified in any of the samples. The research also highlighted a number of problems with the complexity of this type of sampling programme, such as identifying the most likely time to isolate pathogens and analysing an ‘unusual’ water source.  相似文献

硝化处理效率低是硝化处理工艺的主要问题。硝化过程主要由硝化细菌完成,因此硝化作用的效果基本取决于硝化菌体的生长状况。文章通过硝化细菌生长方式进行探讨,提出硝化菌体在载体表面选择附着生长模式,对于理解硝化作用与硝化菌体生长的关系,硝化菌全生物膜的生物特征,从而相应地改善载体的优化设计,取得较好的硝化处理效果。  相似文献

In this study, a denitrification (DN)–partial nitritation (PN)–anaerobic ammonia oxidation (Anammox) system for the efficient nitrogen removal of mature landfill leachate was built with a zone-partitioning self-reflux biological reactor as the core device, and the effects of changes in seasonal temperature on the nitrogen removal in non-temperature-control environment were explored. The results showed that as the seasonal temperature decreased from 34°C to 11.3°C, the total nitrogen removal rate of the DN-PN-Anammox system gradually decreased from the peak value of 1.42 kg/(m3?day) to 0.49 kg/(m3?day). At low temperatures (<20°C), when the nitrogen load (NLR) of the system is not appropriate, the fluctuation of high NH4+-N concentration in the landfill leachate greatly influenced the stability of the nitrogen removal. At temperatures of 11°C–15°C, the NLR of the system is controlled below 0.5 kg/(m3?day), which can achieve stable nitrogen removal and the nitrogen removal efficiency can reach above 96%. The abundance of Candidatus Brocadia gradually increased with the decrease of temperature. Nitrosomonas, Candidatus Brocadia and Candidatus Kuenenia as the main functional microorganisms in the low temperature.  相似文献
