The toxic equivalency factor (TEF) method has been developed to evaluate structurally related compounds, sharing a common mechanism of action. Because certain polycyclic aromatic hydrocarbons (PAHs) appear to fit this requirement and are ubiquitous environmental contaminants, these compounds are TEF candidates. Toxicity is often expressed relative to benzo[a]pyrene (BaP), the reference standard (e.g., TEF = EC50BaP/EC50PAH). BaP‐like toxicity or toxic equivalents (TEQs) can then be calculated: TEQ = S(PAH i × TEF i ), where PAH i and TEF i are the concentration and TEF, respectively, for individual PAH congeners. Representative TEFs for PAHs were compiled from studies measuring carcinogenic potency and surrogate biomarkers. This review examines the application of TEFs to PAHs with established criteria (i.e., demonstrated need, defined chemical grouping, broad toxicological database, endpoint consistency, additivity, common mechanism of action, consensus). Although all criteria are not satisfied (e.g., endpoint inconsistency, nonadditivity) and more rigorous validation studies are needed, the TEF method is useful when limitations are recognized. Refinements of the method might include incorporation of pharmacokinetic factors and segregation of TEFs derived from mammalian vs. nonmammalian systems to increase endopoint consistency. Probabilistic analysis may also prove useful to assess the range of TEF values reported both within and between human health and ecological risk assessments.  相似文献

Until several years ago dioxins were considered as just an unwanted by product of anthropogenic activities and stigmatized as the symbol of man-made environmental pollution. Natural processes, such as forest fires, can emit dioxins, but compared to industrial processes, usually very low quantities are emitted. However after a case of food contamination occurred in the United States of America in 1996 caused by kaolinitic clay a discussion on the provenience started. Besides the relatively high concentration also an unusual PCDD/F distribution pattern was found in these ball clay samples. This specific pattern related to none of the known anthropogenic sources for these contaminants and, in relation to a supposed natural formation, later it was named “natural formation pattern”. Hydrothermal carbonization (HTC) can transform biomass within hours into a brown coal-like product which resembles naturally occurring coal formation. HTC can also transform an already present PCDD/F contamination in a way to obtain a “natural formation pattern” characterized by an unusual high ratio between 1,2,3,7,8,9-HxCDD and 1,2,3,6,7,8-HxCDD and the absence of almost all chlorinated dibenzofurans. By varying the experimental conditions of the HTC process applied to sewage sludge samples contaminated with PCDD/Fs from anthropogenic sources, beside the “natural formation pattern” at a temperatures of 255 °C, a remarkable increase of the toxicity based on WHO–TEQ was observed.  相似文献

Biomonitoring of industrial chemicals in human tissues and fluids has shown that all people carry a “body burden” of synthetic chemicals. Although measurement of an environmental chemical in a person's tissues/fluids is an indication of exposure, it does not necessarily mean the exposure concentration is sufficient to cause an adverse effect. Since humans are exposed to multiple chemicals, there may be a combination effect (e.g., additive, synergistic) associated with low-level exposures to multiple classes of contaminants, which may impact a variety of organ systems. The objective of this research is to link measures of body burden of environmental chemicals and a “holistic” measure of wellness. The approach is demonstrated using biomonitoring data from the National Health and Nutrition Examination Surveys (NHANES). Forty-two chemicals were selected for analysis based on their detection levels. Six biological pathway-specific indices were evaluated using groups of chemicals associated with each pathway. Five of the six pathways were negatively associated with wellness. Three non-zero interaction terms were detected which may provide empirical evidence of crosstalk across pathways. The approach identified five of the 42 chemicals from a variety of classes (metals, pesticides, furans, polycyclic aromatic hydrocarbons) as accounting for 71% of the weight linking body burden to wellness. Significant interactions were detected indicating the effect of smoking is exacerbated by body burden of environmental chemicals. Use of a holistic index on both sides of the exposure-health equation is a novel and promising empirical “systems biology” approach to risk evaluation of complex environmental exposures.  相似文献
