Abstract:Di-n-butyl phthalate(DnBP) and diisobutyl phthalate(DiBP) are phthalate plasticizers being widely used in plastics, resins and rubber products, which can be frequently detected in various environmental matrices. Existing studies have shown that DnBP and DiBP have significant endocrine disrupting effects and adverse effects on humans and other animals, such as reproductive and developmental toxicity. Therefore, their use is restricted in many countries and regions. As a result, some substitutive plasticizers have emerged on the market. Based on domestic and international studies on DnBP, DiBP and their substitutes, this article reviews the endocrine disrupting effects of DnBP and DiBP at individual, cellular and molecular levels, and summarizes their mechanisms of endocrine disrupting effects and their substitutive plasticizers on the market. Four typical substitutes(diisononyl phthalate, diisononyl cyclohexane-1,2-dicarboxylate, dipropylene glycol dibenzoate and glyceryl triacetate) are selected for comparative analysis of their endocrine disrupting effects. Research results show that DnBP and DiBP can affect the expression of key genes in the steroid hormone synthesis pathway at low environmental concentrations, thereby changing the level of steroid hormones in the organism. They will cause damage to the reproductive organs and ultimately reduce reproductive capacity at high concentrations, and the same effect on offspring have been reported due to the exposure during pregnancy. The four alternatives of DnBP and DiBP have reduced the acute toxicity, persistence and bioaccumulation, and endocrine disrupting effects of them have also been significantly reduced. However, there is a lack of systematic studies to fully evaluate their substitutability. It is recommended that future safety assessment of alternatives should include more toxicity endpoints and in-depth study of mechanisms of related toxic effects to ensure the safety of substitutes.
