Abstract:A loop-mediated isothermal amplification(LAMP) technology was developed for rapidly detecting human astrovirus.The data showed that the optimal amplification condition was at 65℃ for 90 min with 4 mmol·L-1 magnesium ions and 1 mol·L-1 betaine.The specificity of RT-LAMP assay was further validated by restriction analgsis of the amplified products and we got two bands(84bp and 135bp) as expected.There is also no cross-reaction with different virus(human rotavirus and norovirus) and restriction analysis of the amplified products.The sensitivity of the LAMP and PCR were 5.04 copies·μL-1 and 5.04×10 copies·μL-1,respectively.Improved RT-LAMP and RT-PCR were both used for detection of human astrovirus in 12 reclaimed water samples.The detection rates of human astrovirus were 41.7%(5/12) and 33.3%(4/12),respectively.All these results suggested that LAMP was a simple,high sensitivity and specificity method which was potential for rapid detection of human astrovirus.
