摘    要:为了考察原核显微注射法制备转基因小鼠过程中M16和M2鼠卵营养液的PH的影响因素,测定了不同存放条件下的M2和M16营养液的PH,结果发现,M2 完全开放的空气环境中放置7h,PH升高2.6%(P〈0.001),在半封闭的实验环境中放置4d,PH升高3.7%(P〈0.001);M16在相对密闭的条件下放置7d和15d,PH分别升高4.5%(P〈0.001)和6.4%(P〈0.001),在半封闭的实

Abstract:s:In order to examine the interfering factors in the course of generation of transgenic mice by pronuclear microrinjection,the pH of M2 and M16 were measured under different conditions.pH of M2 increased by 2 6%(p<0 001)when it was placed in the open air for 7 hours,3 7%(p<0 001)in the semi closed condition for 4 days.In the case of M16,pH rise of 4 5%(p<0 001)and 6 4%(p<0 001)were found in the relatively closed condition,17 0%(p<0 001)and 18 6%(p<0 001)when placed in the semi closed condition for 7 days and 15 days,respectively,pH increased by 20 7%(p<0 001)in the open air only for 7 hours;while pH of M16 kept relatively constant in the CO 2 incubator of 5 0% CO 2 and 95 0% of air for consecutive days.It was found that the pH change of M16 and M2 medium was caused by alkaline gas in the ambient air.
