摘    要:自制了毒死蜱多克隆抗体,考察了离子强度和甲醇含量对抗原抗体竞争反应的影响,对反应体系进行了优化,最终建立了水样中毒死蜱的残留检测ELISA方法。结果表明,毒死蜱对抗体竞争性结合的I50为76.8μg/L,方法检出限为5.2μg/L。河水样品经简单前处理后分别采用ELISA和GC测定,用ELISA方法测得水样中毒死蜱的添加回收率为93.40%~102.1%,C.V为4.04%~5.18%,在水样中的最低检出浓度为0.08μg/L,检测灵敏度远高于GC,符合农药残留检测的要求。

Abstract:Using self-made polyclonal antibody the influence of ion-strength and the concentration of methanol on the competitive reaction were investigated,and the reaction system optimized.This paper developed an indirect competitive enzyme-linked immunosorbent assay(ELISA)for determining chloropyrifos residue in water.Surface water samples were detected with ELISA and GC after simple pre-processing with the detection limit of 0.08μg/L,result of ELISA being verified by GC method.
