富勒烯对PCR反应的抑制作用

摘    要:以110bp单链DNA为模板,研究富勒烯(C60)对聚合酶链式反应(polymerase chain reaction,PCR)的影响.实验结果表明,随着C60浓度的增加,PCR反应被显著抑制;将Taq DNA聚合酶、单链DNA模板与C60孵育后,其PCR扩增产物均显著减少;增加PCR反应体系中的Taq DNA聚合酶量,可消除C60的抑制作用,但增加起始单链DNA模板的数量,效应不明显,上述研究结果说明,C60不仅可抑制Taq DNA聚合酶活性,同时对DNA模板也具有一定的损伤作用.

Abstract:Based on a designed 110 bp single strand DNA template, a polymerase chain reaction (PCR) method was established to evaluate the effect of C60 on DNA in vitro replication. The results indicated that C60 could dose-dependently inhibit the generation of PCR products. After co-incubation with C60, the activity of treated Taq DNA polymerase was significantly inhibited, and the amplification product of the treated DNA template was also decreased. The inhibition effect induced by C60 could be eliminated by increasing the amount of Taq DNA polymerase in the PCR reaction solution, but could not be eliminated by increasing the quantity of DNA template. The results revealed that C60 could inhibit the activity of Taq DNA polymerase, and may have potential adverse effects on the DNA template.
