摘    要:为研究明日叶查尔酮(AC)对小鼠H22+肝癌细胞增殖的抑制作用,采用如下方法进行实验:1)将50只荷H22肝癌小鼠随机分为5组,每组10只.低、中、高剂量组每日分别灌胃给予5、20和40 mg·kg-1明日叶查尔酮,肿瘤对照组给予生理盐水,环磷酰胺(CTX)组隔天腹腔注射20 mg· kg-1的CTX.饲养10 d后处...

Abstract:To investigate the inhibitive effect of chalcone of Angelica keiskei(AC) on the proliferation of mice hepatocarcinoma cells,the following methods were used:1) Fifty mice inoculated hepatocarcinoma 22 cells were divided into five groups which was 10 mice in per group.5,20 and 40 mg·kg-1 AC were given daily to low,medium and high dose groups by mouth.The tumor control group was given saline.20 mg·kg-1 cytoxan(CTX) was given to the CTX group by injection every other day.After 10 days,all mice were sacrificed.Tumor weight,body weight and rate of tumor inhibition were determined.The expression levels of PCNA and Caspase-3 were measured with immunohistochemistry method.2) Mice hepatocarcinoma 22 cells were cultured with different concentrations of AC.MTT assy was used to determine the proliferative activity of hepatocarcinoma cells.The results were as follows:1) The tumor weight,exprssion level of PCNA and Caspase-3 of the tumor control group were(0.55±0.23) g,72.77% and 5%,respectively.Those of high-dose AC group were(0.31±0.05) g,28.33% and 38.52%,respectively.2)The cell proliferation activity of tumor control group and high-dose group were 1.135±0.032 and 0.711±0.028,respectively.All the differences between two groups were significant(p<0.05).The conclusion of this study is that AC can inhibit the proliferative activity of mice hepatocarcinoma 22 cells.
