鸡粪除臭菌的筛选、培养条件优化及其应用

摘    要:为了筛选出具有高效除臭能力的菌株,采用驯化富集、平板划线的方法从鸡粪中筛选除臭微生物,采用初筛、复筛相结合的方法检测所筛选微生物的除臭效果。初筛采用嗅阈值法,复筛以氨气和硫化氢的去除率为检测指标,最终获得除臭效果最好的菌株CCJZ022。经过形态学观察、生理生化鉴定和16S rRNA基因序列分析,确定菌株CCJZ022为红平红球菌(Rhodococcus erythropolis)。对其初始pH值、培养温度及接种量进行优化,发现该菌株在初始pH值为7.0、培养温度为30℃、接种量为12%时除臭效果最好,对氨气和硫化氢的去除率分别可达66.73%和54.51%。利用该菌株进行鸡粪除臭的小试也取得了很好的效果,表明该菌株可以应用于鸡粪除臭。

Abstract:The given paper is oriented to make a report of its study for a microorganism with high deodorant efficiency for the chicken manure.The said chicken manure is recommended to be supplied with certain concentrated ammonia and hydrogen sulfide to enrich the deodorant bacteria strains.According to the experimental results,the bacteria strains tend to be isolated and purified by using a dilution plate and streak plate,which consists entirely of 39 microorganisms isolated from the chicken manure,named CCJZ001-CCJZ039.After being screened,the seven bacterial strains with potential high deodorant efficiency can be isolated with the olfactory threshold method,and then,rescreened by comparing the removal rate of the ammonia and that of hydrogen sulfide,so as to get a strain entitled as CCJZ022,which is expected to be endowed with the highest removal rate of the ammonia and hydrogen sulfide,that is,the removal rate of the ammonia and hydrogen sulfide of 62.56% and 52.63%,respectively.Technically speaking,the strain CCJZ022 has been identified by the colony and cell morphology through a series of physiological and biochemical tests,as well as the gene analysis of 16S rRNA.In biology,the strain we have detected can be classified as Rhodococcus erythropolis.Furthermore,we have also investigated the optimal culturing conditions (temperature,inoculation and pH) of the strain CCJZ022 via a single factor experiment with its removal rate of the ammonia and hydrogen sulfide as its indexes.The results indicate that the change of the culture condition could change the removal rate of ammonia and hydrogen sulfide of the strain.The optimal culture conditions of the strain can be stated as follows:temperature 30 ℃,pH 7.0,and the total inoculation size 12% (v/v).Under the optimized conditions the removal rates of ammonia and hydrogen sulfide of the strain are expected to reach 66.73% and 54.51%,respectively.In order to verify the strain deodorizing effect in their actual application,we have laid out a small pilot test to check it,whose results tend to show that after being treated with the strain of chicken manure for 12 days,the strain can be practically applied to the chicken manure almost with no ammonia and hydrogen sulfide released whereas in contrast the control group can still smell the stench for the duration of time.
