血红蛋白-环氧苯乙烯加合物的分析

摘    要:采用环氧苯乙烯和血红蛋白中羧酸基和巯基位的加合物同时测定的方法,对大鼠血红蛋白SO加的进行了体外实 腹腔注射环氧苯乙烯和苯乙烯的体内实验。结果表明在血红蛋白中所分析的3种加合物(SG,1-PE,2-PE)均随着SO剂量而增加。

Abstract:An assay is described for measures of adducts of styrene oxide(SO)with hemoglobin(Hb)wich have been modified at residues containing eit her carboxylic acid or sulfhydryl groups.In the first, SO carboxylic acid adducts are hydrolyzed in basic medium to liberate st yrene glycol(SG).Th en, the remain ing residues are reacted wit h a reductive catalyst(Raney nickel)to liberate the two positional isomers of SO cysteine adducts, i.e., phenylet hanol(2-PE)and ph enylethanol(1-PE).T he products are derivat ized wit h pentafluorobenzoyl chloride and measured by GC NICI M S.The assay was evaluated w ith SO adducts of Hb which had been produced in the blood of S D rats follow ing(a)in vitro modification or(b)i.p.administration of eit her SO or st yrene.Levels of the three analytes(SG, 1-PE and 2-PE)increased w ith dose in hemoglob in.Binding of SO to cyst eine residues of blood prot eins is greatly preferred at the position rather t han the position both in vit ro and in vivo. The lowest detectable adduct concent rat ions are 8 pmol/g protein for SG, 5 pmol/g protein for 1-PE, and 0.6 pmol/g protein for 2-PE.
