Abstract:Zooplankton is an important part of aquatic ecosystem,which is sensitive to environmental factors and indicates the health status of aquatic ecology.Traditional morphological zooplankton survey can not identify copepod nauplius,and it is difficult to accurately understand zooplankton community composition.DNA macrobarcoding provides a new method for the accurate analysis of zooplankton community composition based on DNA sequence differences.In this study,the effects of different sampling methods(water filtration method and net enrichment method)and sampling volume on zooplankton biodiversity monitoring were compared.The results showed that different sizes of zooplankton should adopt different sampling methods,among which direct filtration of 1 L water sample is the best for rotifer diversity,and enrichment of 5~10 L water sample with zooplankton net is more suitable for monitoring cladocerans and copepods.This study initially achieved the standardization of zooplankton metabarcoding monitoring sampling,and also provided technical support for the future aquatic biodiversity monitoring by metabarcoding technology.
