摘    要:为了描述剩余污泥稳定化芦苇床系统中有机质矿化、甲烷排放及相关微生物多样性之间的内在关系,采用PCR-DGGE技术分析了不同芦苇床、不同季节的稳定化污泥中古细菌群落结构及其多样性。序列分析发现,与DGGE条带序列亲缘关系最近的种属为未培养古细菌和未培养产甲烷古细菌。古细菌群落结构呈季节性变化,且同一季节不同芦苇床的群落结构存在一定差异。具有通气结构芦苇床的剩余污泥中产甲烷古细菌的丰度较低,同时甲烷排放量也较低,因此设置通气结构的芦苇床设计更合理。

Abstract:The paper has made an investigation and exploration of the diversity of archaea community coming up from different seasons in the sludge treatment reed bed (STRB) systems so as to treat the surplus activated sludge since the year of 2012 by means of a denatured gradient gel electrophoresis (DGGE).For the research purpose,we have explored the microbial function and the biodegrading mechanism of the contaminants in STRB.In our experiment and investigation practice,it can be said that STRB system should be taken as a kind of eco-environment-friendly and alternative technology for the research purpose.The clustering analysis results from the fingerprints of DGGE prove that the communities of archaea are quite different in the samples we have taken from different reed beds in the same period,signifying that aeration has had clear influence on the archaea community structure.Besides,the communities of archaea can also be regarded as being different in the same reed bed for different periods,implying that the seasons and conditions of the reed tend to contribute to the archaeal community structure.What is more,the diversity indexes of the sludge samples from the aeriferous bed turn out to be lower than those from the non-aeriferous reed one,symbolizing that it is probable for the aeration to inhibit the activities of archaea in the reed bed.Furthermore,in the aeriferous reed bed,the aechaea diversity has been found increasing from the spring to autumn and began to decrease in the winter.But,in contrast,the aechaea diversity of the non-aeriferous reed bed tends to increase gradually with the extension of the stabilizing period.Of all the 12 bands which have been successfully sequenced for archaea,the obtained sequences should be submitted to the GenBank database for the alignment application via a basic local alignment search tool (BLASTN) algorithm of nucleotide in the 16S rRNA sequences of archaea database to determine their phylogenetic affiliations.The aforementioned sequences should be affiliated to the uncultured archaeon and uncultured methanogenic archaea,which should belong to the kingdom of Euryarchaeota,the dominant archaea in STRB.Moreover,it is necessary to point it out that archaea has been found less abundant in the aeriferous reed bed,which should have been contributable to the reduction of the methane production.
