烟草悬浮细胞抗氧化系统对微囊藻毒素-RR的响应

摘    要:采用0.1mg/L的微囊藻毒素-RR(MC-RR)处理烟草BY-2悬浮细胞,测定了细胞活力、细胞内活性氧(ROS)及抗氧化系统的变化.结果表明,处理144h后,BY-2的细胞活力与对照相比无显著差异,但是处理细胞的ROS含量随着MC-RR处理时间的延长而迅速上升,96h后,细胞的ROS含量与对照差异显著;毒素处理细胞的过氧化物酶(POD)和谷胱甘肽过氧化物酶(GPX)活性明显升高,分别在处理48h和72h后与对照差异显著;细胞中还原型谷胱甘肽(GSH)含量有一个先下降后升高的过程,96h后,MC-RR处理的细胞中GSH含量显著高于对照;然而,0.1mg/L的MC-RR处理144h后,过氧化氢酶(CAT)和超氧化物歧化酶(SOD)活性与对照相比没有显著差异.恢复处理168h后,抗氧化系统各个指标均恢复到对照水平.

Abstract:Adopting microcystin-RR(MC-RR) of 0.1mg/L to treat tobacco BY-2 suspension cell; the changes of cells reactivity, reactive oxygen species (ROS), in the cells and the antioxidant systems were determined. The cell activity of BY-2 after 144h treating was not different markedly against the contrast; but the ROS content of the cell treating raised rapidly with prolonging of MC-RR treating time; after 96h, the ROS content of the cell was different markedly against the contract; the content of GSH in the cell of MC-RR treating was higher markedly than the contract; while after 144h treating the MC-RR of 0.1mg/L, the activities of CAT and SOD were not different markedly against the contract. After 168h of recovering treating, each index of antioxidant system all recovered to the control level.
