摘    要:从传统中药大玉竹中提取低聚糖Poos,用吡啶-氯磺酸法制备了它的硫酸酯衍生物S-Poos,产物经红外鉴定,证实Poos硫酸酯化后羟基连上硫酸基,硫含量为8.47%,取代度(Ds)为0.6体外实验研究了Poos、S-Poos对非洲绿猴肾细胞(Vero)的细胞毒怀和抗单纯疱疹病毒2型(HSV-2)活性。结果表明,Poos和S-Poos都无细胞毒性。Poos本身无抗病毒活性,对HSV-2引起的细胞病变和

Abstract:Poos, a kind of oligosaccharide extracted from Polygonatum od oratum (Mill.) Druce (a kind of Chinese herb), was sulfated to obtain S-Poos with chlorosulfur acid-pyridine. IR showed adsorption at 1 250, 810 cm-1 . The sulfur content of S-Poos was found to be 8.47%. The average degree of su b stitution was 0.6. The hydroxy group of S-Poos was esterified by chlorosulfu r acid. Cytotoxicity in Vero cell, and antivtus activity of Poos and S-Poos we re investigated in vitro. Poos and S-Poos both showed no cytotoxi city in Vero cell. Poos itself did not inhibit the cytopathic effect and plaque forming of H erpes simplex virus type 2(HSV-2) in Vero cell, but its sulfated de rivative ob tained antivirus activity, which could well inhibit the cytopathic effect of HS V-2 at concentrations of 125~2 000 μg/mL, and completely inhibit plaque forming of HSV-2 at concentrations as low as 200 μg/mL.
