铁皮石斛种质资源的低温离体保存

摘    要:研究了在低温条件下保存铁皮石斛种质资源的方法和影响因素。结果表明,铁皮石斛苗在4℃黑暗条件下可连续保存12个月,并能恢复下正常生长。1/2MS和1/4MS培养基上的存活率高于MS。蔗糖深度以20-40g/L为宜。添加低浓度活性炭可明显改善保存材料的状态。甘露醇对铁皮石斛试管苗的保存效果不佳。

Abstract:The method and influencing factors of in vitro conservation of Dendrobium officinale germplasm were studied. Seedlings could be conservedcondition in vitro for 12 months at 4in dark, without subculture. The survival rates on 1/2 MS and 1/4 MS media were higher than that on MS medium. The suitable concentrations of sucrose were 20 to 40 g/L. Supplement with activated charcoal could improve the growth conditions of stored plantlets. However, mannitol was not beneficial to the conservation of D.officinale. Plate 1, Fig 1, Tab 3, Ref 21
