| 摘 要： | 采用ＰＣＲ技术，扩增了平胸龟、黄缘盒龟的Ｓｏｘ基因，扩增产物与人ｐＹ５３．３ＳＲＹ基因探针进行Ｓｏｕｔｈｅｒｎ杂交，证实了扩增产物是人ＳＲＹ基因的同源片段；采用ＳＣＰ分析技术，显示龟类该基因片段的序列与人有较大差异，而龟类种间差异则较小，种内雌雄个体间则无差异．本文结果支持Ｓｏｘ基因在系统演化上十分保守的结论．

THE PCR AMPLIFICATION
AND SSCP ANALYSIS OF Sox GENE
IN TURTLES (PLATYSTERNON MEGACEPHALUM AND
CISTOCLEMMYS FLAVOMARGINATUS) |

| Abstract: | The Sox genes of Platysternon megacephalum and Cistoclemmys flavomarginatus were amplified using PCR, and the products were proved to be the homologue fragment of human SRY gene by Southern bloting hybridization with probe of pY53.3. The results of SSCP analysis indicated that there was a bigger difference in Sox gene sequence between turtle and human than that between the two turtle species, and there was no difference in the Sox gene sequence between the male and female in species. The result of this paper supports the conclusion that the Sox gene is conservative in phylogenetics. |
