摘    要:采用小鼠全胚胎培养模型,将8.5 d小鼠胚胎在含有千里光碱的即刻离心血清中培养48 h(千里光碱的终浓度分别为100、50、25和12.5 μg·mL-1),观察千里光碱对胚胎生长发育(卵黄囊直径、颅臀长、头长和体节数)和组织器官形态分化(卵黄囊循环、尿囊、翻转、心、脑、尾神经管、视听嗅系统、腮弓、颌突和肢芽)的影响....

Abstract:Mouse whole embryo culture(WEC)was applied to culture the post-implantation(8.5 d)mouse embryos which were isolated from their mothers,then,were put into the medium of immediately centrifuged serum(ICS).Embryos cultured for 48 h in different concentrations of senecionine including 100 μg·mL-1,50 μg·mL-1,25 μg·mL-1 and 12.5 μg·mL-1,development(yolk sac diameter,crown-rump length,head length,somite number)and organic morphodifferentiation(yolk sac circulation,allantois,embryonic flexion,heart,brain,optic-otic-olfactory organ,branchial arch,maxillary,mandible,bud)of embryos were observed.Result showed that with the increasing of the concentrations of senecionine,the effect of senecionine on the development and organic morphodifferentiation of embryos were more and more severe,especially on yolk sac circulation,heart,optic organ,olfactory organ,mandible and hindible bud.It can be concluded that senecionine had obvious fetotoxicity in vitro WEC culture.It indicates that exposure of pregnant mice to senecionine may have potential risk on fetals.
