The association of the direct-acting mutagenicity of soluble organic fraction of airborne particles toward Salmonella typhimurium YG1024 strain with the direct emission was investigated at a roadside and at a residential area in Osaka, Japan. The direct-acting mutagenicity was evaluated as mutagenic activity per unit volume of ambient air (rev m−3) and/or that per airborne particulate weight collected on a filter (rev mg−1). The annual or diurnal changes of the mutagenicity of airborne particles at the residential site showed similar patterns to those of some gaseous pollutants such as NO2 and SO2, which were emitted from combustion processes. This result indicates that the mutagenicity is mainly attributable to the primary emissions. From the analysis of the relationship between the wind sector and the mutagenic intensity, rev m−3 and rev mg−1 values were strongly affected by the emissions from the fixed sources and from the mobile sources, respectively. The rev m−3 value and concentration of 1-nitropyrene (1-NP) in unit per m3 at the roadside were a factor of 2.6 and 2.8 higher than those at the residential site, respectively, but the rev mg−1 value and concentration of 1-NP in unit per mg at the roadside were substantially comparable to those at the residential area. These observations suggest that the characteristics of the airborne particles can be attributed to the automotive emissions even at the suburban area.  相似文献

Background, aim, and scope  

We recently developed a new isolation method for diesel exhaust particles (DEP), involving successive extraction with H2O, sodium bicarbonate, and sodium hydroxide, in which the sodium hydroxide extract was found to consist of phenolic components. Analysis of the extract revealed that vasodilative-active nitrophenols are in DEP in significantly higher concentrations than those estimated by an earlier method involving a combination of solvent extraction and repeated chromatography. These findings indicated that our new procedure offers a simple, efficient, and reliable method for the isolation and identification of bioactive substances in DEP. This encouraged us to extend our work toward investigating new vasodilatory substances in the sodium bicarbonate extract.  相似文献

The impacts of ultrafine airborne particles generated by diesel or gasoline engines on human lung cells have been investigated using a new in vitro cellular exposure technique. This technique enables direct deposition of the gasoline engine exhaust particles (GEP) and diesel engine exhaust particles (DEP) on human lung cells located at the air-cell interface on a transwell membrane in an exposure apparatus. The cellular responses to particle exposure were measured by the levels of IL-8 chemokines produced as a function of exposure time. The findings suggest that GEP and high-sulfur DEP induced the production of similar levels of IL-8 by unprimed A549 cells. The level of IL-8 produced by unprimed A549 cells in response to low-sulfur DEP was found lower than that produced in response to high-sulfur DEP and GEP. When cells were primed, simulating predisposed conditions, significant levels of IL-8 were produced. GEP triggered a much higher level of IL-8 production than DEP did. Furthermore, the time profile of IL-8 production induced by GEP was markedly different from that induced by DEP. The findings indicate that GEP could induce the production of higher levels of chemokines (i.e., IL-8) than DEP did, implying that exposure to GEP could be a greater health risk than exposure to DEP.  相似文献

Phenanthrene is a 3-ring polycyclic aromatic hydrocarbon which exists mainly in the gas-phase in the atmosphere. Recent concern over the presence of 9,10-phenanthrenequinone in ambient particles led us to study the products of the gas-phase reactions of phenanthrene with hydroxyl radicals, nitrate radicals and ozone. The formation yields of 9,10-phenanthrenequinone were measured to be ∼3%, 33±9%, and ∼2% from the OH radical, NO3 radical and O3 reactions, respectively. Calculations suggest that daytime OH radical-initiated and nighttime NO3 radical-initiated reactions of gas-phase phenanthrene may be significant sources of 9,10-phenanthrenequinone in ambient atmospheres. In contrast, the ozone reaction with phenanthrene is unlikely to contribute significantly to ambient 9,10-phenanthrenequinone.  相似文献

In total, 24 polycyclic aromatic hydrocarbons (PAHs) in both gas and particle phases and 35 nitro-PAHs in particle phase were analyzed in the exhaust from heavy-duty diesel vehicles equipped with after-treatment for particulate matter (PM) and NOX control. The test vehicles were carried out using a chassis dynamometer under highway cruise, transient Urban Dynamometer Driving Schedule (UDDS), and idle operation. The after-treatment efficiently abated more than 90% of the total PAHs. Indeed, the particle-bound PAHs were reduced by >99%, and the gaseous PAHs were removed at various extents depending on the type of after-treatment and the test cycles. The PAHs in gas phase dominated the total PAH (gas + particle phases) emissions for all the test vehicles and for all cycles; that is, 99% of the two-ring and 98% of the three-ring and 97% of the four-ring and 95% of the carcinogenic PAHs were in the gas-phase after a diesel particle filter (DPF) and not bound to the very small amount of particulate matter left after a DPF. Consequently, an evaluation of the toxicity of DPF exhaust must include this volatile fraction and cannot be based on the particle fraction only. The selective catalytic reduction (SCR) did not appear to promote nitration of the PAHs in general, although there might be some selective nitration of phenanthrene. Importantly the after-treatmtent reduced the equivalent B[a]P (B[a]Peq) emissions by >95%, suggesting a substantial health benefit.

Implications: This study demonstrated that after-treatments, including diesel particulate filters (DPF), diesel oxidation catalysts (DOC), and selective catalytic reduction (SCR), significantly reduce the emissions of PAHs from heavy-duty diesel engines. The gas-phase PAHs dominate the total PAH (gas + particle phases) emissions from heavy-duty diesel vehicles retrofitted with various DPFs and not bound to the very small amount of particulate matter left after a DPF. Consequently, an evaluation of the toxicity of DPF exhaust must also include this volatile fraction and cannot be based on the particle fraction only.

To identify major PM2.5 (particulate matter ≤2.5 μm in aerodynamic diameter) sources with a particular emphasis on the ship engine emissions from a major port, integrated 24 h PM2.5 speciation data collected between 2000 and 2005 at five United State Environmental Protection Agency's Speciation Trends Network monitoring sites in Seattle, WA were analyzed. Seven to ten PM2.5 sources were identified through the application of positive matrix factorization (PMF). Secondary particles (12–26% for secondary nitrate; 17–20% for secondary sulfate) and gasoline vehicle emissions (13–31%) made the largest contributions to the PM2.5 mass concentrations at all of the monitoring sites except for the residential Lake Forest site, where wood smoke contributed the most PM2.5 mass (31%). Other identified sources include diesel vehicle emissions, airborne soil, residual oil combustion, sea salt, aged sea salt, metal processing, and cement kiln. Residual oil combustion sources identified at multiple monitoring sites point clearly to the Port of Seattle suggesting ship emissions as the source of oil combustion particles. In addition, the relationship between sulfate concentrations and the oil combustion emissions indicated contributions of ship emissions to the local sulfate concentrations. The analysis of spatial variability of PM2.5 sources shows that the spatial distributions of several PM2.5 sources were heterogeneous within a given air shed.  相似文献

The automobile exhausts are one of the major sources of particulate matter in urban areas and these particles are known to influence the atmospheric chemistry in a variety of ways. Because of this, the oxidation of dissolved sulfur dioxide by oxygen was studied in aqueous suspensions of particulates, obtained by scraping the particles deposited inside a diesel truck exhaust pipe (DEP). A variation in pH showed the rate to increase with increase in pH from 5.22 to about ~6.3 and to decrease thereafter becoming very slow at pH?=?8.2. In acetate-buffered medium, the reaction rate was higher than the rate in unbuffered medium at the same pH. Further, the rate was found to be higher in suspension than in the leachate under otherwise identical conditions. And, the reaction rate in the blank reaction was the slowest. This appears to be due to catalysis by leached metal ions in leachate and due to catalysis by leached metal ions and particulate surface both in suspensions. The kinetics of dissolved SO2 oxidation in acetate-buffered medium as well as in unbuffered medium at pH?=?5.22 were defined by rate law: k obs ?=?k 0?+?k cat [DEP], where k obs and k 0 are observed rate constants in the presence and the absence of DEP and k cat is the rate constant for DEP-catalyzed pathway. At pH?=?8.2, the reaction rate was strongly inhibited by DEP in buffered and unbuffered media. Results suggest that the DEP would have an inhibiting effect in those areas where rainwater pH is 7 or more. These results at high pH are of particular significance to the Indian subcontinent, because of high rainwater pH. Conversely, it indicates the DEP to retard the oxidation of dissolved SO2 and control rainwater acidification.

The relationship between immune defense mechanisms and environmental pollutants has been a focus of intensive research during the last decade. In animal experiments, diesel exhaust particles (DEP) have been shown to exert adjuvant effects on the IgE response against aeroantigens and to compromise broncho-pulmonary immune defense. Important target cells are monocytes/alveolar macrophages which display the important functions of phagocytosis, antigenpresentation and modulation of inflammatory processes. To further evaluate the influence of DEP upon these cells, we investigated whether exposure to suspended DEP would change the phagocytic capacity and the tumor necrosis factor alpha (TNF-α) release of human peripheral monocytes/alveolar macrophages. Our results demonstrated a dose-dependent reduction of phagocytosis (p < 0.001) and an additional increase of an ongoing TNF-α response (p < 0.005). These findings may help to link exposure to environmental pollutants to such biologic effects as increased susceptibility to broncho-pulmonary disease or facilitated sensitization against aeroallergens.  相似文献

Relatively little is known about exposures to traffic-related particulate matter at schools located in dense urban areas. The purpose of this study was to examine the influences of diesel traffic proximity and intensity on ambient concentrations of fine particulate matter (PM2.5) and black carbon (BC), an indicator of diesel exhaust particles, at New York City (NYC) high schools. Outdoor PM2.5 and BC were monitored continuously for 4–6 weeks at each of 3 NYC schools and 1 suburban school located 40 km upwind of the city. Traffic count data were obtained using an automated traffic counter or video camera. BC concentrations were 2–3 fold higher at urban schools compared with the suburban school, and among the 3 urban schools, BC concentrations were higher at schools located adjacent to highways. PM2.5 concentrations were significantly higher at urban schools than at the suburban school, but concentrations did not vary significantly among urban schools. Both hourly average counts of trucks and buses and meteorological factors such as wind direction, wind speed, and humidity were significantly associated with hourly average ambient BC and PM2.5 concentrations in multivariate regression models. An increase of 443 trucks/buses per hour was associated with a 0.62 μg/m3 increase in hourly average BC at an NYC school located adjacent to a major interstate highway. Car traffic counts were not associated with BC. The results suggest that local diesel vehicle traffic may be important sources of airborne fine particles in dense urban areas and consequently may contribute to local variations in PM2.5 concentrations. In urban areas with higher levels of diesel traffic, local, neighborhood-scale monitoring of pollutants such as BC, which compared to PM2.5, is a more specific indicator of diesel exhaust particles, may more accurately represent population exposures.  相似文献

Aerosol particles were collected in the situation of the widespread dust suspension on 21 February 1991 at Qira in the southern edge of the Taklamakan Desert, western China. The collected particles were examined by a transmission electron microscope equipped with an energy-dispersive X-ray (EDX) analyzer in order to obtain the size and elemental composition of individual mineral particles.On the basis of EDX analyses for 386 particles, mineral particles were present in high number fractions (>99%) of particles in the radius range of 0.1–4 μm. Particles mainly composed of silicates comprised 76% of mineral particles. “Ca-rich” particles were detected in 7% of all the particles. Ca in the particles would be present not only as CaCO3 but also as an internal mixture of CaCO3 and CaSO4. Particles containing halite (NaCl) were detected in number proportions of about 10% and were mainly present in the radius range of 0.5 μm. Some halite particles would be modified by chemical reactions with sulfuric acid.  相似文献

A nitronaphthalene kinetics mechanism has been implemented and added to the photochemical smog mechanism, Carbon Bond-4. This mechanism was used to simulate the formation, decay, and partitioning of 1- and 2-nitronaphthalene and compare it to outdoor smog chamber data. The results suggest that these types of mechanisms can be used to model nitronaphthalene formation and decay in regional airmasses. The partitioning experiments were conducted at night. The sampling system consisted of two Teflon impregnated glass fiber filters followed by a gas-phase denuder. Evidence is provided that partitioning equilibrium is maintained even when the gas-phase components are decaying rapidly under sunlight. The photolysis rate constants were determined relative to kNO2 to be 0.07×kNO2 and 0.005×kNO2 for 1- and 2-nitronaphthalene, respectively. Our results confirm that gas-phase photolysis is the major degradation pathway for 1-nitronaphthalene, whereas for 2-nitronaphthalene other pathways may also be important. The photochemical formation of nitronaphthalenes was studied using a mixture of naphthalene, propylene, NOx, and diesel particles. 2-nitronaphthalene was observed to build up to higher levels than 1-nitronaphthalene, as the photodegradation of the latter was faster. Additionally, as a part of this study 1-nitronaphthalene has been identified and quantified in diesel exhaust.  相似文献

Aerosol mass spectrometer (AMS) measurements are used to characterize the evolution of exhaust particulate matter (PM) properties near and downwind of vehicle sources. The AMS provides time-resolved chemically speciated mass loadings and mass-weighted size distributions of nonrefractory PM smaller than 1 microm (NRPM1). Source measurements of aircraft PM show that black carbon particles inhibit nucleation by serving as condensation sinks for the volatile and semi-volatile exhaust gases. Real-world source measurements of ground vehicle PM are obtained by deploying an AMS aboard a mobile laboratory. Characteristic features of the exhaust PM chemical composition and size distribution are discussed. PM mass and number concentrations are used with above-background gas-phase carbon dioxide (CO2) concentrations to calculate on-road emission factors for individual vehicles. Highly variable ratios between particle number and mass concentrations are observed for individual vehicles. NRPM1 mass emission factors measured for on-road diesel vehicles are approximately 50% lower than those from dynamometer studies. Factor analysis of AMS data (FA-AMS) is applied for the first time to map variations in exhaust PM mass downwind of a highway. In this study, above-background vehicle PM concentrations are highest close to the highway and decrease by a factor of 2 by 200 m away from the highway. Comparison with the gas-phase CO2 concentrations indicates that these vehicle PM mass gradients are largely driven by dilution. Secondary aerosol species do not show a similar gradient in absolute mass concentrations; thus, their relative contribution to total ambient PM mass concentrations increases as a function of distance from the highway. FA-AMS of single particle and ensemble data at an urban receptor site shows that condensation of these secondary aerosol species onto vehicle exhaust particles results in spatial and temporal evolution of the size and composition of vehicle exhaust PM on urban and regional scales.  相似文献

To better understand the atmospheric behaviour of pesticides, heterogeneous ozonolysis of three herbicides (alachlor, terbuthylazine and trifluralin) adsorbed on silica particles were performed in a flow reactor. The experimental setup used in this study and previously validated (Pflieger et al., 2009) has been specially developed to investigate extremely slow reactivity. The pesticides were adsorbed on particles using a gas/solid adsorption equilibrium, in order to simulate atmospheric conditions. After exposure to ozone concentrations ranging from 5 to 41 ppm during 90 min to 6 h, the kinetics were calculated by comparing the initial and the remaining amounts of pesticides adsorbed on silica particles. This work offers the first results of heterogeneous ozonolysis of alachlor and trifluralin adsorbed on mineral particles. Although alachlor and terbuthylazine were expected to react with ozone, no degradation was observed which leads to a lifetime higher than 8 months towards ozonolysis (for 40 ppb of O3). A significant degradation of trifluralin adsorbed on silica particles by heterogeneous ozonolysis was observed. The experimental data could be fit by both the Langmuir–Rideal and the Langmuir–Hinshelwood models resulting in atmospheric lifetimes (towards heterogeneous ozonolysis) of 40 and 32 days respectively (for 40 ppb of O3). These results are discussed and compared to other studies.  相似文献

The potential problem of 1-nitropyrene (1-NP) formation during filter sampling of diesel emissions from dilution tubes is assessed. Rates of formation are calculated using data generated from several independent filter exposure studies. A portion of the 1-NP (12% average) found in participates collected from light-duty (LDD) and heavy-duty diesels (HOD) was found to be due to formation on the filter under average sampling conditions of 10-15/1 dilution at 44°C with 3 ppm NO2 for 23 min. On the average, the concentration of 1-NP in participates emitted from HDD is 16 times less than that found in LDD run under similiar transient conditions. The average emission rate of 1-NP generated from the LDD and HDD was 4.7 μg/km and 1.5 μg/km, respectively for the vehicles operating under Federal Test Procedure (FTP)-transient conditions. Under operating conditions which increase exhaust temperature (increased speed and load), the concentration of nitro-PAH is significantly reduced with a concurrent increase in the concentration of partially oxidized nitro-PAH and dinitro-PAH. The concentration of 1-NP in the particulates measured from HDD in these dilution tube studies (0.33-0.95 ppm, depending upon engine load) is comparable to that measured in highway tunnel experiments (0.54 ppm average).  相似文献

Diesel exhaust related airborne Particulate Matter (PM) has been linked to a myriad of adverse health outcomes, ranging from cancer to cardiopulmonary disease. The underlying toxicological mechanisms are of great scientific interest. A hypothesis under investigation is that many of the adverse health effects may derive from oxidative stress, initiated by the formation of reactive oxygen species (ROS) within affected cells. In this study, the main objective was to determine whether aged-diesel exhaust PM has a higher oxidant generation and toxicity than fresh diesel exhaust PM. The diesel exhaust PM was generated from a 1980 Mercedes-Benz model 300SD, and a dual 270 m3 Teflon film chamber was utilized to generate two test atmospheres. One side of the chamber is used to produce ozone–diesel exhaust PM system, and another side of the chamber was used to produce diesel exhaust PM only system. A newly optimized dithiothreitol (DTT) method was used to assess their oxidant generation and toxicity. The results of this study showed: (1) both fresh and aged-diesel exhaust PM had high oxidant generation and toxicity; (2) ozone–diesel exhaust PM had a higher toxicity response than diesel exhaust PM only; (3) the diesel exhaust PM toxicity increased with time; (4) the optimized DTT method could be used as a good quantitative chemical assay for oxidant generation and toxicity measurement.  相似文献

Size, morphology, and composition of airborne particles strongly affect human health and visibility, precipitation, and the kinetic characteristics of particles. In this study, the morphology and chemical composition of particles emitted from conventional (diesel and gasoline) and alternative (CNG and methanol) fuel vehicles were characterized through scanning electron microscopy (SEM) and energy-dispersive X-ray (EDX). The SEM images revealed that the size of primary particles (without agglomeration) was approximately 10 nm in the exhaust from all the tested vehicles. The particles emitted from gasoline vehicle (GV), CNG vehicle (CNGV), and methanol vehicle (MV) had the same median diameter, 62 nm, which was smaller than those from heavy diesel vehicle (HDV) and light diesel vehicle (LDV). Soot was observed in the HDV, LDV, and GV samples but not in the CNGV and MV. The fractal dimension, which was used to quantify the degree of irregularity of soot, was 1.752 ± 0.014, 1.789 ± 0.076, and 1.769 ± 0.006 in the exhaust from HDV, LDV, and GV samples, respectively. The particles discharged by all tested vehicles contained the elements C, O, Fe, and Na. The main element in the samples of HDV, LDV, and GV was C, while O was the main element in the samples of alternative fuel vehicles. The profiles of minor elements were more complex in the emissions of alternative fuel vehicles than those in the emissions of conventional fuel vehicles. The results improved our understanding of the morphology and elemental composition of particles emitted from vehicles powered by diesel, gasoline, CNG, and methanol.

Free radical generation potential of automobile exhaust gas was examined by measuring hydroxyl (OH) radical photo-formation rates in exhaust gas-scrubbing water. Effects of automobile exhausts on physiological status of Japanese red pine trees (Pinus densiflora Sieb. et Zucc.) were also investigated to elucidate the mechanism how the free radicals derived from exhaust gas damage higher plants. Gasoline and diesel exhaust gases were scrubbed into pure water. Potential photo-formation rates of OH radical in aqueous phase (normalized to sun light intensity of clear sky midday on May 1 at 34°N) for gasoline and diesel cars were ave. 51 and 107 μM h−1 m−3 of exhaust gas, respectively. Nitrite was a dominant source (ca. 70-90%) of photochemical formation of OH radical in both gasoline and diesel car exhausts. The scrubbed solution of diesel car exhaust gas was sprayed for six times per week to needles of pine tree seedlings in open top chambers. Control, exhaust + mannitol (added as OH radical scavenger), and nitrite + nitrate standard solution (equivalent levels existed in the exhaust gas) were also sprayed. Two months sprays indicated that the sprayed solutions of diesel exhaust and nitrite + nitrate caused a decrease of maximum photosynthetic rate and stomata conductance in pine needles while the control and exhaust + mannitol solution showed no effects on photosynthetic activities of pine needles. These results indicated that OH radicals generated mainly from photolysis of nitrite occurring in the scrubbing solution of exhaust gas are responsible for the decrease of photosynthetic activities of pine needles.  相似文献

GOAL, SCOPE, AND BACKGROUND: Diesel exhaust is believed to consist of thousands of organic constituents and is a major cause of urban pollution. We recently reported that a systematic separation procedure involving successive solvent extractions, followed by repeated column chromatography, resulted in the isolation of vasodilatory active nitrophenols. These findings indicated that the estimation of the amount of nitrophenols in the environment is important to evaluate their effect on human health. The isolation procedure, however, involved successive solvent extractions followed by tedious, repeated chromatography, resulting in poor fractionation and in a significant loss of accuracy and reliability. Therefore, it was crucial to develop an alternative, efficient, and reliable analytical method. Here, we describe a facile and efficient acid-base extraction procedure for the analysis of nitrophenols. MATERIALS AND METHODS: Diesel exhaust particles (DEP) were collected from the exhaust of a 4JB1-type engine (ISUZU Automobile Co., Tokyo, Japan). Gas chromatography-mass spectrometry (GC-MS) analysis was performed with a GCMS-QP2010 instrument (Shimadzu, Kyoto, Japan). RESULTS: A solution of DEP in 1-butanol was extracted with aqueous NaOH to afford a nitrophenol-rich oily extract. The resulting oil was methylated with trimethylsilyldiazomethane and subsequently subjected to GC-MS analysis, revealing that 4-nitrophenol, 3-methyl-4-nitrophenol, 2-methyl-4-nitrophenol, and 4-nitro-3-phenylphenol were present in significantly higher concentrations than those reported previously. DISCUSSION: Simple acid-base extraction followed by the direct analysis of the resulting extract by GC-MS gave only broad peaks of nitrophenols with a poor detection limit, while the GC-MS analysis of the sample pretreated with (trimethylsilyl)diazomethane gave satisfactorily clear chromatograms with sharp peaks and with a significantly lowered detection limit (0.5 ng/ml, approximately 100 times). CONCLUSION: The present method involving an acid-base extraction, in situ derivatization, and GC-MS analysis has shown to be a simple, efficient, and reliable method for the isolation and identification of the chemical substances in DEP.  相似文献

Aluminium (Al) is one of the trace inorganic metals present in atmospheric particles. Al speciation study is essential to better evaluate the mobility, availability, and persistence of trace Al and Al species in the atmosphere. This paper reports Al distribution and speciation in atmospheric particles with aerodynamic diameters >10.0, 10.0–2.5 and <2.5 μm in the urban area of Nanjing, China. Urban particles were collected with a high-volume sampling system equipped with a cascade impactor, which effectively separates the particulate matter into three size ranges. Particulate Al was fractionated into five different forms (insoluble, oxide, organic, carbonate, and exchangeable species) by the modified five-step Tessier's sequential extraction procedure. The main points are as follows: (1) The average levels of Al in PM2.5, PM2.5–10 and PM>10 are 2.02±0.35, 3.04±0.43 and 6.32±0.76 μg m−3, respectively, with PM2.5, PM2.5–10 and PM>10 constituting respectively, 17.8±3.1%, 26.7±3.8% and 55.5±6.7% of suspended particulate matter (SPM) mass (11.38 μg m−3). (2) The vertical profile of airborne Al in the above three size fractions has been estimated. A significant increase in airborne Al concentrations was found for PM2.5, PM2.5–10 and PM>10 as the sampling height above the ground increased from 2.5 to 17.5 m; however, there was an obvious decrease in airborne Al concentrations between 17.5 and 40.0 m. The maximum mean of total Al in PM2.5, PM2.5–10 and PM>10 occurred between 12.5 and 20.0 m above the ground. (3) The distribution of Al speciation was studied. It was found that the size distribution of airborne Al species followed the order: insoluble species>oxide species>organic species>carbonate species>exchangeable species.  相似文献
