The histology and ultrastructure of the epithelial cells of the outer and middle mantle folds of the bivalves Mytilus edulis (L), Cardium edule (L), Macoma balthica (L) and Nucula sulcata Bronn are described. The cells lining the inner face of the outer fold exhibit a prominent granular endoplasmic reticulum, and Golgi complexes which are concerned with the elaboration of granules and vesicles eventually incorporated into the periostracum. A gradual reduction in the protein synthetic apparatus occurs towards the tip of the fold. Within the cells, it is proposed that the ovoid inclusion bodies are lysosomes and that they control the rate of secretion. The cells of the middle fold are cuboidal in appearance. Those of M. edulis and N. sulcata exhibit prominent granules, whereas those of C. edule and M. balthica possess vesicles. The cells of M. edulis differ from the others in possessing stout bundles of filaments, which occupy large areas of the cell and constitute a cell web. The cells of the epithelium in all cases do not appear to be implicated in periostracum formation.  相似文献

Genetic relations between populations of Cardium (Cerastoderma) edule (L.) and C. lamarcki (Reeve) from Danish waters and C. glaucum (Bruguiére) from the Mediterranean Sea were studied using DNA analysis. Repetitive sequences of nuclear DNA were compared by Southern blotting of enzyme digested DNA fragments, separated on agarose gel by electrophoresis. Fragments were visualized by autoradiography using 32P labelled probes. These were nick translated, randomly sheared fragments of the whole chromosomal genomes of two species of distant taxa — Mytilus edulis and Mya arenaria, respectively. The number of detected DNA bands was 18 for C. edule, 32 for C. lamarcki, and 24 for C. glaucum. Measurements of genetic distances, based on comparisons of these repetitive sequences using a simple dissimilarity coefficients, show the greatest distance, i.e., 0.84 between C. edule and C. lamarcki, 0.69 between C. edule and C. glaucum, and the smallest, 0.52 between C. lamarcki and C. glaucum. Comparisons of the three Cardium types, using gene spliced and cloned C. edule nuclear DNA (nDNA) fragments as probes, accorded with these findings. Also, analysis of mitochondrial DNA (mtDNA) from eggs and hepatopancreas, although not in itself conclusive, accorded with the results of the analyses of nDNA. MtDNA was measured by electron micrograph to 15 000 base pairs (bp), and sizes of restriction enzyme digested fragments were compared by gel electrophoresis. The results of the comparisons based on repetitive sequences supported by the two other studies suggest that C. edule resembles the ancestral form more than the two others, and that C. lamarcki is the most different and recently evolved form. This hypothesis is discussed in relation to the geographical distribution of the species complex.  相似文献

We used natural abundance measurements of stable carbon and nitrogen isotopes and a short-term 15N-enrichment experiment to explore the importance of river-derived and autochthonous sources to the diet of intertidal suspension-feeding bivalves in the Ría de Arosa, an oceanic-dominated system in northwest Spain. Highly seasonal and intense phytoplankton "bloom" events occur in this estuary. However, relatively low phytoplankton standing crop and productivity are present during much of the year, suggesting that food sources other than phytoplankton must support bivalves during these periods. Bivalves (Cerastoderma edule, Tapes decussatus, Mytilus galloprovincialis) were sampled quarterly at four intertidal stations along the length of the estuary with increasing distance from the Ulla River. Muscle tissue was consistently enriched in 13C and 15N relative to stomach contents. The stable C isotope values of bivalves indicated that river-derived inputs were, at most, a minor component of diet. These values suggested the potential importance of benthic microalgae to bivalve diet much of the year and the incorporation of 15N label applied to the sediment surface into stomach contents and muscle tissue in a field experiment supported this conclusion. Other labeled components of the microbenthos such as bacteria could also have been ingested. 13C-depleted values of bivalves in May coincided with elevated offshore chlorophyll a concentrations, suggesting the increased use of a phytoplankton source during the spring. We propose that intertidal suspension-feeding bivalves in this estuary use primarily resuspended microbenthos during periods of low phytoplankton concentration, but that phytoplankton increases in relative dietary importance during bloom events.Communicated by P.W. The recent separation of Cardium glaucum Bruguière from C. edule L. and the scarcity of reports of populations containing both species has necessitated further ecological study to demonstrate differences in habitat preferences. Some mixed and unispecific populations of the cockles Cardium edule and C. glaucum, situated in fjords around the Kattegat and Baltic coasts of Denmark were studied; it was demonstrated that some degree of tidal influece was essential for the survival of C. edule, and that C. glaucum was being restricted by the exposure of the site. A study of the cockle populations of some étangs on the Mediterranean coast of France demonstrated that, despite earlier reports of its presence, C. edule was not present in that area. A brief survey of the Bassin d'Arcachon (France) showed that it contained a mixed cockle population. It was concluded that C. edule was unable to occupy localities having a tidal amplitude of less than 0.2 m, and that high temperatures also restricted occupation of semitidal habitats; similarly, the exposure of the site restricted colonisation by C. glaucum.  相似文献

Habitat selection of two bivalve species, Cardium edule (L.) and C. glaucum (Bruguière) is described for sympatric and allopatric populations of both sibling species. It is demonstrated that C. edule grows faster than C. glaucum and is absent from localities with periodic low food-availability. The ability to burrow of C. glaucum is low compared with that of C. edule; the former species does not occur in very loose, well-sorted sand near the coast. Earlier studies which suggest that tidal range determine the species distribution could not be confirmed.  相似文献

Polyphenols were located in all the layers of the periostracum and in inclusion bodies in the inner face of the outer-fold cells of Mytilus edulis L. Results, after 4 days periostracum repair, show that the proto-ostracal material is deposited in distinct layers. Inclusion bodies in the cells of the inner face of the outer fold appear to give rise to the polyphenols in the proto-ostracal material.  相似文献

Allometric relations were determined in bivalves collected from approximate mid-tide levels in Biscay, Spain, during March 1987. Species compared included the epifaunal suspension-feederMytilus edulis L. (9 to 1 108 mg dry soft-tissue weight) from a rocky-shore population at Meinakotz Beach, and the infaunal deposit-feederCerastoderma edule (L.) (1 to 192 mg dry soft-tissue weight) from the mudflats in Mundaka Ría. Relative to M. edulis, and compared per unit dry tissue weight,C. edule had similar palp areas but smaller gill areas. In addition, to help maximize absorption from organically-poor deposits,C. edule ingested three to four times as much food per hour, but had gut contents that were five to six times greater, so that gut-passage times available for the extraction of nutrients were 2.5 times longer. Metabolic faecal losses, which were comprised of endogenous materials lost from the bivalve into the gut, were two to three times greater inC. edule, but similar to those ofM. edulis when expressed per unit mass ingested by each species. These losses were very substantial, being equivalent to as much as 15% of the ingested mass, and represent a significant indirect cost that is presumably incurred largely by the intracellular digestion characteristic of bivalves. Weight exponents indicated that such metabolic investment represented an unchanging proportion of the total costs of growth. They also showed that age-related constraints on total production did not stem from decreasing gut content. Rather, associated exponents identified limitations to the production in each species as being linked with marked reductions of gutpassage time and ingestion rate, and indicated that these limitations do not derive from corresponding decreases in gill or palp areas.  相似文献

The biological effects of 5 surface active agents (the anionic ABS, LAS and LES 3 EO and the nonionic NP 10 EO and TAE 10 EO) on marine fishes, crustaceans and bivalves have been tested in, continuous-flow systems. Concentrations from 100 to 0.5 ppm were normally used. Fishes were found to be more susceptible (96 hLC 50 range: 0.8 to 6.5 ppm) than bivalves (96 hLC50 range: 5 to >100 ppm) while crustaceans were considerably more resistant (96 h LC 50 range: 25 to > 100 ppm). Within each of these 3 systematic groups, more active species were found to be more sensitive than less active species. Developmental stages were also more sensitive than adults. The resistance of crustaceans to surfactants decreases immediately after moulting. The most toxic agent for fishes and decapods was the soft anionic LAS, and for bivalves and barnacles the hard nonionic NP 10 EO. Ability to recover normal behaviour after exposure decreases with increasing concentration and time, and ceases earlier in anionic than in nonionic surfactants. The first reaction to the surfactants is increased activity (avoidance reaction of mobile species), followed successively by inactivation, immobilization and death. Nonionic surfactants affect the equilibrium in fishes. Sublethal effects appear as impaired locomotory activity and breathing rate in fishes and crustaceans, impaired byssus activity and shell closure in Mytilus edulis, burrowing in Cardium edule, Astarte montagui and Astarte sulcata. Siphon retraction is affected in Mya arenaria and Cardium edule, as is also the response to food of the Leander spp.  相似文献

The ultrastructural details of changes that occur in the inner face of the outer fold of Mytilus edulis during periostracum repair are described. Initially, after the periostracum is slit, increased secretory activity occurs in the cells, apparently controlled by lysosomes. Proto-ostracal material deposited outside the cells seems to be due to Golgi secretory granules, which dissociate and then re-assemble outside the cells to form an organised layer. During the later stages of repair, autophagic vacuole formation increases in the cells, possibly because of increased demands for synthetic material. When substantial amounts of proto-ostracal material are secreted and the outer fold is sealed off from the external environment, the cells return to their normal condition. During the early stages of repair, amoebocytes appear to be implicated in the laying down of proto-ostracal material.  相似文献

A method for determining filtration rates in undisturbed suspension-feeding bivalves is described. Concentrations of particulate matter in the water collected in the inhalant (C i) and exhalant (C e) currents were estimated with an electronic particle counter. The clearance was calculated as , where Fl=flow rate through the tube collecting exhalant water. Only above critical levels of water flow (Fl) were clearances representative of filtration rates. At 10° to 13°C, the filtration rates (F, 1 h-1) within one or two orders of magnitude of dry weight (w, g), in Cardium echinatum L., C. edule L., Mytilus edulis L., Modiolus modiolus (L.) and Arctica islandica (L.) followed the allometric equations: 4.22w 0.62, 11.60w 0.70, 7.45w 0.66, 6.00w 0.75 and 5.55w 0.62, respectively. Five species of bivalves [Spisula subtruncata (da Costa), Hiatella striata (Fleuriau), Cultellus pellucidus (Pennant), Mya arenaria L. and Venerupis pullastra (Montagu)] filtered with the same rates as individuals of Cardium echinatum and A. islandica of equivalent soft weight. In Pecten furtivus and P. opercularis filtration rates were about twice the rates measured in individuals of Mytilus edulis of comparable body weight. The gill area in M. edulis increases with size at the same rate as the filtration rate.  相似文献

Introduced Pacific oysters (Crassostrea gigas) have shown rapid expansion in the Oosterschelde estuary, while stocks of native bivalves declined slightly or remained stable. This indicates that they might have an advantage over native bivalve filter feeders. Hence, at the scale of individual bivalves, we studied whether this advantage occurs in optimizing food intake over native bivalves. We investigated feeding current characteristics, in which potential differences may ultimately lead to a differential food intake. We compared feeding currents of the invasive epibenthic non-siphonate Pacific oyster to those of two native bivalve suspension feeders: the epibenthic siphonate blue mussel Mytilus edulis and the endobenthic siphonate common cockle Cerastoderma edule. Inhalant flow fields were studied empirically using digital particle image velocimetry and particle tracking velocimetry. Exhalant jet speeds were modelled for a range of exhalant-aperture cross-sectional areas as determined in the laboratory and a range of filtration rates derived from literature. Significant differences were found in inhalant and exhalant current velocities and properties of the inhalant flow field (acceleration and distance of influence). At comparable body weight, inhalant current velocities were lower in C. gigas than in the other species. Modelled exhalant jets were higher in C. gigas, but oriented horizontally instead of vertically as in the other species. Despite these significant differences and apparent morphological differences between the three species, absolute differences in feeding current characteristics were small and are not expected to lead to significant differences in feeding efficiency.  相似文献

Rabbit antisera were raised against Cardium edule (L.) from the Waddensea locality, Skallingen, against Cardium lamarcki (Reeve) from the Danish locality, Dybsø Fjord, and against Cardium glaucum (Bruguière) from the French Mediterranean coast at Ile des Embiez. Allopatric and sympatric populations of C. lamarcki and C. edule from Swedish and Danish localities, and allopatric populations of C. glaucum from the Mediterranean Sea, Red Sea and Black Sea were studied by means of the described method and by starch gel electrophoresis of malate dehydrogenase (MDH). By means of crossed immuno-electrophoresis, using intermediary gels containing antibodies against one of the Cardium types and receiver gels containing antibodies against one of the others, type-specific and type-non-specific reactions were recorded. The results are compared with a similar investigation of Mytilus edulis, M. galloprovincialis and M. californianus. Based on immunological properties, it is shown that C. edule is a valid species different from the C. lamarcki/C. glaucum complex. It is further suggested that C. lamarcki and C. glaucum should still be referred to by these two names as significant genetic differences between them is revealed by this immunological study.  相似文献

The upper temperature tolerances of three European species of Cardium [C. (Cerastoderma) glaucum Bruguière, C. tuberculatum L., C. (Cerastoderma) edule L.] are compared by means of median lethal temperature (LT50) and median burial temperature (BT50) determinations for periods of exposure up to 96 h. The influence of previous acclimation temperatures on these determinations is also considered. C. glaucum from shallow water (0.5 to 1 m), lagoonal, habitats in the Mediterranean Sea shows a greater thermal tolerance than C. tuberculatum from deeper water (4 to 5 m) in the Mediterranean. The thermal tolerance of C. edule from the intertidal zone, but at higher latitudes, is intermediate. Young C. glaucum show a greater resistance to short exposures to high temperatures than adults, but for longer exposures their tolerance is the same. All 3 species exhibit a significant effect of previous acclimation temperature on the burrowing response, and C. glaucum and C. tuberculatum a significant but lesser effect on LT50. LT50 determinations with C. edule showed secondary mortality effects which may have obscured any effect of acclimation temperature.  相似文献

In 1983 a sample of cockles [Cerastoderma edule (L.)] was removed from the mid-tide level at Booterstown Strand, County Dublin, Ireland at 2-h intervals, for a period of 24 h. The gills, mantle and visceral mass tissues contained significant levels of lysozyme, with the gills containing the highest, weight-specific levels. A tidal rhythm of lysozyme activity in the gills, mantle and visceral mass was inversely related to the tidal cycle. The data were best described by a tidal curve shifted-5 hours out of phase, with maximum and minimum levels of activity occurring one hour after low and high tides, respectively. In marine bivalves, lysozyme may be involved in the extracellular digestion and absorption of bacteria during low tide. Cardium edule L. and C. glaucum Bruguière are two sibling species of cockles commonly found in littoral systems along the Portuguese coast, where they sometimes occur as mixed populations. The problem of morphological separation of these two cockle species has not been completely solved. Indeed, the distinctive morphological criterion proposed by Brock after identifying the specimens by means of electrophoretic patterns of hepatopancreatic malate dehydrogenase was not effective when applied to Portuguese Cardium spp. The object of the present study was to establish morphological discrimination criteria for distinguishing Portuguese C. edule and C. glaucum on the basis of initial identification of individuals through the enzymatic criterion established by Brock. Cockles were collected from 1988 to 1991 at 16 estuarine and lagoonal systems along the Portuguese coast. Hepatopancreatic extracts were submitted to horizontal starch gel electrophoresis following Brock's procedure. The malate dehydrogenase patterns obtained were identical to those found by Brock. Clear interspecific differences in the leucine aminopeptidase and esterase isozyme systems were also detected. The morphological study of the shells involved 7 morphometric and 9 qualitative and semiquantitative characters. Canonical variate analysis of the morphometric characters showed that these could not be used for identification purposes. However, a clear separation between the two species was obtained by a principal-component analysis of qualitative and semiquantitative characters. The characters mainly useful for distinguishing between the two species were valve profile, shape of ventral valve junction, type of calcareous scales and shell colour.  相似文献

The effect of physical disturbance in the form of trampling on the benthic environment of an intertidal mudflat was investigated. Intense trampling was created as unintended side-effect by benthic ecologists during field experiments in spring and summer 2005, when a mid-shore area of 25 × 25 m was visited twice per month by on average five researchers for a period of 8 months. At the putatively-impacted location (I) (25 × 25 m) and two nearby control locations (Cs) (25 × 25 m each), three sites (4 × 4 m) were randomly selected and at each site, three plots (50 × 50 cm) were sampled after 18 and 40 days from the end of the disturbance. Multivariate and univariate asymmetrical analyses tested for changes in the macrofaunal assemblage, biomass of microphytobenthos and various sediment properties (grain-size, water content, NH4 and NO3 concentrations in the pore water) between the two control locations (Cs) and the putatively-impacted location (I). There were no detectable changes in the sediment properties and microphytobenthos biomass, but variability at small scale was observed. Microphytobenthos and NH4 were correlated at I to the number of footprints, as estimated by the percentage cover of physical depressions. This indicated that trampling could have an impact at small scales, but more investigation is needed. Trampling, instead, clearly modified the abundance and population dynamics of the clam Macoma balthica (L.) and the cockle Cerastoderma edule (L.). There was a negative impact on adults of both species, probably because footsteps directly killed or buried the animals, provoking asphyxia. Conversely, trampling indirectly enhanced recruitment rate of M. balthica, while small-sized C. edule did not react to the trampling. It was likely that small animals could recover more quickly because trampling occurred during the growing season and there was a continuous supply of larvae and juveniles. In addition, trampling might have weakened negative adult-juvenile interactions between adult cockles and juvenile M. balthica, thus facilitating the recruitment. Our findings indicated that human trampling is a relevant source of disturbance for the conservation and management of mudflats. During the growing season recovery can be fast, but in the long-term it might lead towards the dominance of M. balthica to the cost of C. edule, thereby affecting ecosystem functioning.  相似文献

Three marine diatoms, Skeletonema costatum, Chaetoceros debilis, and Thalassiosira gravida were grown under no limitation and ammonium or silicate limitation or starvation. Changes in cell morphology were documented with photomicrographs of ammonium and silicate-limited and non-limited cells, and correlated with observed changes in chemical composition. Cultures grown under silicate starvation or limitation showed an increase in particulate carbon, nitrogen and phosporus and chlorophyll a per unit cell volume compared to non-limited cells; particulate silica per cell volume decreased. Si-starved cells were different from Si-limited cells in that the former contained more particulate carbon and silica per cell volume. The most sensitive indicator of silicate limitation or starvation was the ratio C:Si, being 3 to 5 times higher than the values for non-limited cells. The ratios Si:chlorophyll a and S:P were lower and N:Si was higher than non-limited cells by a factor of 2 to 3. The other ratios, C:N, C:P, C:chlorophyll a, N:chlorophyll a, P:chlorophyll a and N:P were considered not to be sensitive indicators of silicate limitation or starvation. Chlorophyll a, and particulate nitrogen per unit cell volume decreased under ammonium limitation and starvation. NH4-starved cells contained more chlorophyll a, carbon, nitrogen, silica, and phosphorus per cell volume than NH4-limited cells. N:Si was the most sensitive ratio to ammonium limitation or starvation, being 2 to 3 times lower than non-limited cells. Si:chlorophyll a, P:chlorophyll a and N:P were less sensitive, while the ratios C:N, C:chlorophyll a, N:chlorophyll a, C:Si, C:P and Si:P were the least sensitive. Limited cells had less of the limiting nutrient per unit cell volume than starved cells and more of the non-limiting nutrients (i.e., silica and phosphorus for NH4-limited cells). Juvenile bivalves may be dispersed by entering a bysso-pelagic phase where they drift through the water mass aided by a long thread. The ability to resuspend and control the specific weight in two bivalve species, the cockle Cerastoderma edule (L.) and the Japanese clam Ruditapes philippinarum (Adams and Reeves), was documented with juveniles through flume and still-water experiments. Cockle juveniles initially placed on an unsuitable substratum were exposed to two shear velocities (u *). At the end of the experiment, 42 (±15)% (for u *=0.51 cm s−1) and 79 (±9)% (for u *=0.99 cm s−1) of individuals were retrieved from the sand area which represents only 8% of the total flume surface. Most juveniles (70.5%) with shell lengths <2.5 mm migrated from the unsuitable Plexiglas substratum to the sand array by resuspension in the water column. The percentage was lower (21.5%) for larger individuals. The same experimental design was applied to clams, which immediately adhered to the Plexiglas substratum and remained attach to it. Sinking rates of live and dead specimens of both species were measured in a 1 m long transparent PVC tube. Cockle fall velocities showed severe deceleration, probably due to byssus secretion (up to 15-fold slower than dead cockles), sometimes interrupted by brutal acceleration probably due to byssal rupture. Cockles were able to reduce their sinking rate for shell lengths up to 4.25 mm. By contrast, clam sinking rates were constant, and similar to dead clam sinking rates. Specific weights of all experimental juveniles were calculated in relation to their lengths, and their passive motion into the boundary layer was theoretically assessed with Shields curve. In short, C. edule and R. philippinarum can both exhibit dense populations in the field with a good capacity to colonize, although juveniles display different abilities to resuspend in the water column.
